A novel chitosan nanocapsule for enhanced skin penetration of cyclosporin A and effective hair growth in vivo

Hair loss due to medical conditions,such as alopecia,male pattern baldness,and cancer chemotherapy treatment,has been a comm onproblem for many in *** n A(CsA),a fungal metabolite,has been reported to be a hair growth modulatory agent and is apotential drug for hair ***,the effect of topical application of CsA is limited by its poor water *** deliverysystems developed to enhance its solubility still showed poor skin *** overcome these limitations,in this study,we have developed a novel chitosan nanocapsule platform using Pluro nic F127 and chitosan without any chemical crosslinking or complicated preparati on steps for the enhanced water solubility and high tran sdermal penetration of *** chitosan nanocapsules(ChiNCs)optimized in terms of structural stability by using chitosan with various molecular weights ranging from 3 to 100 kDa enhanced the skin permeation of CsA through humancadaver skin in *** administration of the CsA loaded ChiNCs in creased the hair follicles by c.a.7 times higher than that of thecontrol group,and effectively induced hair growth in C57BL/6 mice in *** results suggest that ChiNCs could be used as a platform foreffective transdermal delivery of various hydrophobic drugs.

Objective:This study aimed to investigate the effects of TH/CTS/β-CD microspheres by oral administration on ovalbumin allergic asthma of a rat ***:Spray drying method was applied for preparing the TH/CTS/β-CD microspheres,which were used in treatment of asthmatic *** a predetermined time,the levels of eosinophil(Eos),protein,lactate dehydrogenase(LDH),aspartate transaminase(AST),glutamic pyruvic transaminase(GPT),and creatinine(Cr)were ascertained by automatic hematology and biochemical *** tissue histology was performed by hematoxylin-eosin ***:No significant differences were found about the contents of Eos in the blood,and the contents of Eos,protein and LDH in the bronchoalveolar lavage fluid between the microsphere-treated and dexamethasone-treated groups,but the levels were lower in both treated groups than in the model *** the microsphere-treated group,the levels of Cr,AST,and GPT in the serum of rats showed no significant differences compared with the normal *** on lung histopathological findings,the microspheres attenuated inflammatory cell infiltration and mucus hypersecretion compared with the model ***:This study demonstrated that TH/CTS/β-CD microspheres exerted an anti-inflammatory effect and could serve as a novel promising drug delivery system for asthma treatment.

Bacterial infections are grave threats to human health,particularly those caused by the most common Grampositive *** massive administration of broad-spectrum antibiotics to treat various bacterial infections has led to the evolution and spread of drug *** a universal antimicrobial technique unapt to induce drug resistance,photothermal therapy(PTT)is attracting extensive attention in recent ***,its unspecific killing capability and side effects towards adjacent mammalian cells severely impede the practical ***,we proposed a metabolic engineering strategy to selectively inactivate Gram-positive bacteria by PTT.A bioorthogonal photothermal agent was prepared by the conjugation of IR-780 iodide and dibenzocyclooctyne(IR780-DBCO).Upon pre-metabolizing with 3-azido-D-alanine,Gram-positive bacteria rather than Gramnegative ones,such as Staphylococcus aureus and vancomycinresistant Enterococcus faecalis(VRE),could be specifically tied up by the explosive IR780-DBCO via copper-free click ***,they spontaneously detonated under 15 min near-infrared light irradiation and inactivated nearly 100% Gram-positive bacteria in ***,superbug VRE-induced infection was significantly inhibited by this approach in a mouse skin wound *** metabolic labelling-based photothermal ablation strategy specific to Gram-positive microbes would stimulate the development of precise antibacterial candidates for preclinical applications.

OBJECTIVE:To investigate the safety profiles of Motherwort injection(MI).METHODS:A multi-center,prospective and drugderived hospital intensive monitoring method was conducted to assess the safety of MI in real world *** study was based on a very large population after the injection was approved and marketed in *** patients using the injection in participating hospitals were monitored to determine the incidence,pattern,severity and outcome of associated adverse ***:The post-marketing surveillance was performed in 10 094 female patients from April to December,*** incidence of adverse drug reactions(ADRs) was 0.79‰(8/10 094).Among the 8 patients,the reported adverse events mainly included systemic abnormalities,such as fever,chills and eyelid edema;skin and appendages disorders,such as pruritus and rash;gastrointestinal disorders,such as nausea,abdominal distension and pain;heart rate and rhythm disorders,such as palpitation and increased heart *** of these ADRs were mild in ***:In this study the ADRs incidence rate of MI is very low,which supports that it is generally safe for use in obstetric and gynecological ***,the total number of 8 ADRs recorded over a relatively short time span seems limited,and the low number of reports could not represent an absolute guarantee of safety.
