To the Editor:With an aging global population,the incidences of community-acquired pneumonia(CAP)and chronic obstructive pulmonary disease(COPD)have signicantly increased.[1]Previous studies have conrmed that COPD and asthma are independently associated with the prevalence of *** use of inhaled corticosteroid(ICs),the cornerstone of treatment for asthma,COPD with frequent acute exacerbations,and asthma-COPD overlap(ACO)may induce changes in the local lung microbiome and abnormal lung immunity,ultimately,causing a signicantly increased risk of ***,in cases of pneumonia,the effect of the use of ICs on CAP mortality remains *** data from one study favored the prior use of ICs,which was associated with a signicantly lower short-term mortality rate,[2]other studies have identied no impact on *** date,data on the impact of the use of ICs on mortality,prehospitalization or during hospitalization,are scarce,particularly in the older ***,this multicenter,retrospective study explored the association between the use of ICs during hospitalization and short-term mortality in older patients with CAP and those with chronic pulmonary disease(CPD).

Trisomy 12 in a Case of Multiple Cutaneous squamous Cell Carcinoma in Association with Chronic Lymphocytic Leukemia

Chronic lymphocytic leukaemia （CLL）, which shares clinical and morphological overlap with small lymphocytic lyjmphoma （sLL）, is a low-grade clonal B-cell lymphoproliferative disorder that accounts for 25% of all cases of leukaemia in Western countries, while it is considered rare in Oriental patients and is thought to constitute only 2% of all leukemias in these patients. CLL is associated with an increased incidence of secondary malignant neoplasms, such as brain tumors, melanomas, and gastrointestinal-tract carcinomas. However, the simulataneous occurrence of CLL and cutaneous squamous cell carcinoma （sCC） is rarely reported. We present here a case of CLL with multiple sCC on the face. subsequent studies demonstrated the patient to have a trisomy 12 identified in bone marrow specimen.

sequential expression of cyclooxygenase-2, glutamate receptor-2, and platelet activating factor receptor in rat hippocampal neurons after fluid percussion injury

Traumatic brain injury causes gene expression changes in different brain regions. Occurrence and development of traumatic brain injury are closely related, involving expression of three factors, namely cyclooxygenase-2, glutamate receptor-2, and platelet activating factor receptor. However, little is known about the correlation of these three factors and brain neuronal injury. In this study, primary cultured rat hippocampal neurons were subjected to fluid percussion injury according to scott’s method, with some modifications. RT-PCR and semi-quantitative immunocytochemical staining was used to measure the expression levels of cyclooxygenase-2, glutamate receptor-2, and platelet activating factor receptor. Our results found that cyclooxygenase-2 expression were firstly increased post-injury, and then decreased. Both mRNA and protein expression levels reached peaks at 8 and 12 hours post-injury, respectively. similar sequential changes in glutamate receptor 2 were observed, with highest levels mRNA and protein expression at 8 and 12 hours post-injury respectively. On the contrary, the expressions of platelet activating factor receptor were firstly decreased post-injury, and then increased. Both mRNA and protein expression levels reached the lowest levels at 8 and 12 hours post-injury, respectively. Totally, our findings suggest that these three factors are involved in occurrence and development of hippocampal neuronal injury.

Human amniotic epithelial cell transplantation for the repair of injured brachial plexus nerve: evaluation of nerve viscoelastic properties

The transplantation of embryonic stem cells can effectively improve the creeping strength of nerves near an injury site in animals. Amniotic epithelial cells have similar biological properties as em-bryonic stem cells; therefore, we hypothesized that transplantation of amniotic epithelial cells can repair peripheral nerve injury and recover the creeping strength of the brachial plexus nerve. In the present study, a brachial plexus injury model was established in rabbits using the C6root avulsion method. A suspension of human amniotic epithelial cells was repeatedly injected over an area 4.0 mm lateral to the cephal and caudal ends of the C6 brachial plexus injury site （1 × 106 cells/mL, 3μL/injection, 25 injections） immediately after the injury. The results showed that the decrease in stress and increase in strain at 7,200 seconds in the injured rabbit C6 brachial plexus nerve were mitigated by the cell transplantation, restoring the viscoelastic stress relaxation and creep properties of the brachial plexus nerve. The forepaw functions were also signiifcantly improved at 26 weeks after injury. These data indicate that transplantation of human amniotic epithelial cells can effec-tively restore the mechanical properties of the brachial plexus nerve after injury in rabbits and that viscoelasticity may be an important index for the evaluation of brachial plexus injury in animals.

High-resolution sonographic Measurements of Lower Extremity Bursae in chinese Healthy Young Men

Background： Lower extremity bursae are very vulnerable to injury during strenuous physical *** the imaging characteristics of normal bursae is essential for early diagnosis of morphological ***, we evaluated the normal range of lower extremity bursae in healthy young men using high-resolution ultrasound （HR-Us） ***： Bursae in the lower extremities were examined by HR-Us in 290 chinese healthy young men with a median age of 18 years （range, 18-23 years）.The bilateral suprapatellar bursa （sPB）, deep infrapatellar bursa （DIPB）, popliteal bursa （PB）, and retrocalcaneal bursa （RCB） were imaged and measured for ***： The HR-Us identification rates of the sPB, DIPB, PB, and RCB were 89.0% （517/580）, 55.0% （319/580）, 29.4% （171/580）, and 49.5% （287/580）, *** the assumption that the bursae were normal in 95% of the study participants, the length and width values at the maximal cross-section of the sPB, DIPB, PB, and RCB were ≤18.00 and 6.09 mm, 8.10 and 2.1 1 mm, 7.67 and 3.93 mm, and 7.82 and 2.04 mm, ***： Using HR-Us imaging, we were able to analyze lower extremity bursae with high detection rates in healthy young *** normal ranges of lower extremity bursa dimensions in healthy young men measured by HR-Us in this study could be used as reference values for evaluation of bursa abnormalities in the lower extremity.

TRICHOsTATIN A INHIBITs PROLIFERATION,INDUCEs APOPTOsIs AND CELL CYCLE ARREsT IN HELA CELLs

Objective： The histone deacetylase inhibitors （HDACIs） have been shown to inhibit cancer cell proliferation, stimulate apoptosis, an induce cell cycle arrest. Our purpose was to investigate the antiproliferative effects of a HDACI, trichostatin A （TsA）, against human cervical cancer cells （HeLa）. Methods： HeLa cells were treated in vitro with various concentrations of TsA. The inhibitory effect of TsA on the growth of HeLa cells was measured by MTT assay. To detect the characteristic of apoptosis chromatin condensation, HeLa cells were stained with Hoechst 33258 in the presence of TsA. Induction of cell cycle arrest was studied by flow cytometry. Changes in gene expression of p53, p21wafl and p27Kipl were studied by semiquantitative RT-PCR. Results： TsA inhibited cell growth in a time- and dose-dependent manner. Hoechst 33258 staining assay showed that TsA induced apoptosis. Cell cycle analysis indicated that treatment with TsA decreased the proportion of cells in s phase and increased the proportion of cells in G0/G1 and/or G2/M phases of the cell cycle. This was concomitant with overexpression of genes related to malignant phenotype, including an increase in p53, p21wall and p27Kipl. Conclusion： These results suggest that TsA is effective in inhibiting growth of HeLa cells in vitro. The findings raise the possibility that TsA may prove particularly effective in treatment of cervical cancers.

Gene expression profile changes in brain regions following traumatic brain injury at the gene level cannot sufficiently elucidate gene expression time, expression amount, protein post-translational processing or modification. Therefore, it is necessary to quantitatively analyze the gene expression profile using proteomic techniques. In the present study, we established a rat model of closed brain injury using Marmarou＇s weight-drop device, and investigated hippocampal differential protein expression using two-dimensional gel electrophoresis and surface-enhanced laser desorption ionization-time of flight-mass spectrometry. A total of 364 protein peaks were detected on weak cation exchange-2 protein chips, including 37 differential protein peaks. 345 protein peaks were detected on immobilized metal affinity capture arrays-Cu, including 12 differential protein peaks Further examination of these differential proteins revealed that glucose-regulated protein and proteasome subunit alpha type 3 expression were significantly upregulated post-injury. These results indicate that brain injury can alter protein expression in the hippocampus, and that glucose-regulated protein and proteasome subunit alpha type 3 are closely associated with the occurrence and development of traumatic brain injury.

studies on Human Fetal Lens Crystallins Under Oxidative stress and Protective Effects of Tea Polyphenols

Purpose: To investigate the oxidative modification of water-soluble crystallins of human fetal lens with H2O2 and fourteen metal ions with or without EDTA. Tea-polyphenols (TP) was added to above solutions in order to testing their antioxidative abilities. Methods: The experiments were performed at 37℃ with final concentration of 2.5mg/ml protein, 0.1 mM metal ions, 0.3 mM EDTA and 1.0 mM H2O2. Then the TP was added to the solution with CusO4 and H2O2, after 5 or 24 hours, the crystallins were analysed with sDs-PAGE and *** : There were marked oxidative modifications of lens protein in H2O2 and copper without EDTA. In sDs-PAGE patterns, we found an increase in those species above of bands higher than 30 kD and some diffuse bands from 30 to 17 kD after 5 hours. In IEF patterns, there were a general increase on acidity with loss of the more basic species. Pedicale screw system plus ACPC perfusion to treat fractures of thoracolumbar vertebrae

In recent years the pedicle screw system has been widely used in treating thoracolumbar vertebral fractures. The effect to recover the injured vertebrae height, the spinal physiological curve and to decompress vertebral canal has been confirmed. But the problems of internal fixation bending, loosening and breaking, which result in the loss of the vertebral height and spinal angulation deformity, are quite common and cause a lot of difficulties for surgeons. To solve these problems and decrease the sequelae from treating thoracolumbar vertebrae fractures with vertebral pedicle screw system, we have tried using vertebral pedicle screw system plus ACPC perfusion to treat 18patients with thoracolumbar vertebral *** results have been obtained.

Building the Pharmacophore Model of HIV-1 Integrase strand Transfer Inhibitors and studying Their Inhibition Mechanism

The replication of HIV-1 requires the integration of its cyclic DNA into host DNA by HIV-1 integrase （IN）, which includes two important reactions, 3＇-processing and strand transfer, both catalyzed by HIV-1 IN. Disrupting either of the reactions will fulfill the purpose of inhibiting the replication of HIV-1. In this paper, pharmacophore modeling and molecular docking are employed to investigate the inhibition mechanism of the HIV-1 IN strand transfer inhibitors （INsTIs）. Based on the results, we suggest that the inhibition mechanism of INsTIs involves the inhibitor chelating the cofactors Mg2＋ and its forming hydrogen bonds with some crucial residues adjacent to the DDE active center.
