STING agonist-conjugated metal-organic framework induces artificial leukocytoid structures and immune hotspots for systemic antitumor responses

Radiotherapy is widely used for cancer treatment, but its clinical utility is limited by radioresistance and its inability to target metastases. Nanoscale metal-organic frameworks(MOFs) have shown promise as high-Z nanoradiosensitizers to enhance radiotherapy and induce immunostimulatory regulation of the tumor microenvironment. We hypothesized that MOFs could deliver small-molecule therapeutics to synergize with radiotherapy for enhanced antitumor efficacy. Herein, we develop a robust nanoradiosensitizer,GA-MOF, by conjugating a STING agonist, 2,3-cyclic guanosine monophosphate–adenosine monophosphate(GA), on MOFs for synergistic radiosensitization and STING activation. GA-MOF demonstrated strong anticancer efficacy by forming immune-cell-rich nodules(artificial leukocytoid structures) and transforming them into immunostimulatory hotspots with radiotherapy. Further combination with an immune checkpoint blockade suppressed distant tumors through systemic immune activation. Our work not only demonstrates the potent radiosensitization of GA-MOF, but also provides detailed mechanisms regarding MOF distribution, immune regulatory pathways and long-term immune effects.

The tension and compression of face-centered-cubic high-entropy alloy(HEA) nanowires are significantly asymmetric, but the tension–compression asymmetry in nanoscale body-centered-cubic(BCC) HEAs is still unclear. In this study,the tension–compression asymmetry of the BCC Al Cr Fe Co Ni HEA nanowire is investigated using molecular dynamics simulations. The results show a significant asymmetry in both the yield and flow stresses, with BCC HEA nanowire stronger under compression than under tension. The strength asymmetry originates from the completely different deformation mechanisms in tension and compression. In compression, atomic amorphization dominates plastic deformation and contributes to the strengthening, while in tension, deformation twinning prevails and weakens the HEA *** tension–compression asymmetry exhibits a clear trend of increasing with the increasing nanowire cross-sectional edge length and decreasing temperature. In particular, the compressive strengths along the [001] and [111] crystallographic orientations are stronger than the tensile counterparts, while the [110] crystallographic orientation shows the exactly opposite trend. The dependences of tension–compression asymmetry on the cross-sectional edge length, crystallographic orientation,and temperature are explained in terms of the deformation behavior of HEA nanowire as well as its variations caused by the change in these influential factors. These findings may deepen our understanding of the tension–compression asymmetry of the BCC HEA nanowires.

The majority of the projectiles used in the hypersonic penetration study are solid flat-nosed cylindrical projectiles with a diameter of less than 20 *** study aims to fill the gap in the experimental and analytical study of the evolution of the nose shape of larger hollow projectiles under hypersonic *** the hypersonic penetration test,eight ogive-nose AerMet100 steel projectiles with a diameter of 40 mm were launched to hit concrete targets with impact velocities that ranged from 1351 to 1877 m/*** erosion of the projectiles was observed during high-speed penetration of heterogeneous targets,and apparent localized mushrooming occurred in the front nose of recovered *** examining the damage to projectiles,a linear relationship was found between the relative length reduction rate and the initial kinetic energy of projectiles in different penetration ***,microscopic analysis revealed the forming mechanism of the localized mushrooming phenomenon for eroding penetration,i.e.,material spall erosion abrasion mechanism,material flow and redistribution abrasion mechanism and localized radial upsetting deformation ***,a model of highspeed penetration that included erosion was established on the basis of a model of the evolution of the projectile nose that considers radial upsetting;the model was validated by test data from the literature and the present *** upon the impact velocity,v0,the projectile nose may behave as undistorted,radially distorted or *** to the effects of abrasion of the projectile and enhancement of radial upsetting on the duration and amplitude of the secondary rising segment in the pulse shape of projectile deceleration,the predicted DOP had an upper limit.

The dissolution and diffusion of CO_(2)in oil and water and its displacement mechanism were investigated by laboratory experiment and numerical simulation for Block 9 in the Tahe oilfield,a sandstone oil reservoir with strong bottom-water drive in Tarim Basin,Northwest *** parameters were analyzed as solubility ratio of CO_(2)in oil,gas and water,interfacial tension,in-situ oil viscosity distribution,remaining oil saturation distribution,and oil *** results show that CO_(2)flooding could control water coning and increase oil *** the early stage of the injection process,CO_(2)expanded vertically due to gravity differentiation,and extended laterally under the action of strong bottom water in the intermediate and late *** CO_(2)got enriched and extended at the oil-water interface,forming a high interfacial tension zone,which inhibited the coning of bottom water to some extent.A miscible region with low interfacial tension formed at the gas injection front,which reduced the in-situ oil viscosity by about 50%.The numerical simulation results show that enhanced oil recovery(EOR)is estimated at 5.72%and the oil exchange ratio of CO_(2)is 0.17 t/t.

Dear Editor,Three dimensional(3D)bioprinted extracellular matrix(ECM)can be used to provide both biochemical and biophysical cues to direct mesenchymal stem cells(MSCs)differentiation,and then differentiated cells were isolated for implantation in vivo using surgical ***,the reduced cell activity after cell isolation from 3D constructs and low cell retention in injured sites limit its application[1].Methacrylated gelatin(GelMA)hydrogel has the advantage of fast crosslinking,which could resemble complex architectures of tissue construct in vivo[2].Here,we adopted a noninvasive bioprinting procedure to imitate the regenerative microenvironment that could simultaneously direct the sweat gland(SG)and vascular differentiation from MSCs and ultimately promote the replacement of glandular tissue in situ(Fig.1a).

A New Curve Fitting Method for forming Limit Experimental Data

The forming limit curve （FLC） can be obtained by means of curve fitting the limit strain points of different strain paths. The theory of percent regression analysis is applied to the curve fitting of forming limit experimental *** intervals of FLC percentiles can be calculated. Thus reliability and confidence level can be considered. The theoretical method to get the limits of limit strain points distributing region is presented, and the FLC position can be adjusted according to practical requirement. Method for establishing FLC with high reliability using small samples is presented at the same time. This method can make full use of the current experimental data and the previous *** with the traditional method that can only use current experimental data, fewer specimens are required in the present method to obtain the same precision and the result is more accurate with the same number of specimens.

本文以SiO_(2)为载体和以CMC和SDBS及两者复配修饰得到硫化亚铁浆剂,发现CMC/SDBS 1:3质量比修饰SiO_(2)/FeS(表活剂和FeS质量比为1:1)的分散性和抗氧化性的效果可双向提高。在研究材料中,CMC/SDBS 1:3复配修饰SiO_(2)/FeS浆剂的粒径最小(D_(50)=0.66μm)、Zeta电位绝对值最大(值达到30 mV以上)。SiO_(2)/FeS-CMC/SDBS 1:3对镉污染溶液修复效果好,且Cd(Ⅱ)主要通过离子交换和化学沉淀作用形成CdS和CdSO_(4)产物(产物形态稳定)。在空气环境和氮气环境中,SiO_(2)/FeS-CMC/SDBS 1:3对镉的吸附量差距不大,30 d达到158 mg/g左右。特别地,在修复低浓度镉污染时,SiO_(2)/FeS-CMC/SDBS 1:3的吸附量比SiO_(2)/FeS-CMC和SiO_(2)/FeS-SDBS的吸附量大,且没有酸化风险。本研究揭示了CMC和SDBS复配修饰SiO_(2)/FeS分散稳定性的提高及其在镉污染地下水修复中的应用,为Cd污染地下水原位修复提供科学依据。
