Bioinformatics and in vitro study reveal the roles of microRNA-346 in high glucose-induced human retinal pigment epithelial cell damage

AIM:To study microRNAs(miRNAs)and their potential effects in high glucose-induced human retinal pigment epithelial cell ***:We screened the GSE52233 miRNA expression dataset for differentially expressed miRNAs(DEMs).The target genes of the top 10 DEMs were predicted using miRWalk 2.0 database,followed by function enrichment and protein-protein interaction *** expression was determined in the human retinal pigment epithelial cell line ARPE-19 treated with high glucose(HG)by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Cell proliferation was determined using cell counting kit(CCK)-8 *** cycle,apoptosis,and reactive oxygen species(ROS)levels were determined by flow *** direct interaction between miRNA and targets was validated using dual-luciferase reporter ***:Thirty-nine DEMs were screened,and we predicted 125 miRNA-mRNA pairs for the top 10 DEMs,including 119 target genes of seven DEMs such as miR-346,which was upregulated in diabetic retinopathy(DR).miR-346 target genes were substantially enriched in the regulation of intracellular transport and retinoic acidinducible gene I(RIG-I)-like receptor signaling *** of three upregulated and downregulated miRNAs were verified by qRT-PCR in HG-treated ARPE-19 *** of miR-346 was elevated in HG treated ARPE-19 cells in a dose-dependent *** inhibited cell proliferation and induced apoptosis,which were partly reversed by transfecting an miR-346 inhibitor,which even decreased the ROS levels elevated due to *** 2(AGO2)was a target of ***:miR-346 is a key miRNA and plays an important role in HG-induced damage in human retinal pigment epithelial cells.

Clinical research of EX-PRESS drainage device and modified trabeculectomy combined with intravitreal conbercept treatment for neovascular glaucoma

AIM:To evaluate the efficacy and safety of modified trabeculectomy(experimental group) and implantation of EX-PRESS drainage device(control group),combined with intravitreal conbercept injection for neovascular glaucoma(NVG).METHODS:Totally 30 patients with NVG were selected from June 2014 to June 2017,and randomly divided into experimental group and control *** patients were underwent intravitreal conbercept(0.5 mg/0.05 mL) treatment before *** trabeculectomy was performed in MT group,while EX-PRESS drainage device implantation was performed in EX *** success rates,best corrected visual acuity(BCVA),intraocular pressure(IOP),filtering bleb and complications were observed and ***:The differences of success rate,BCVA and filtering bleb were not statistically significant 12mo after the surgery(P>0.05),however,the difference of IOP at 1d,1wk,1,3,and 6mo after surgery was statistically significant(F_(time)=390.64,P_(time)0.05),while the results at other time point indicate statistical differences(P<0.05).CONCLUSION:The modified trabeculectomy and the implantation of EX-PRESS drainage device have clinical application value in reducing IOP and postoperative complications of refractory NVG.

BACKGROUND Lipemia retinalis(LR)is a rare disease related to ***,the symptoms of hypertriglyceridemia are insidious and difficult to detect without blood *** fundus is the only site where blood vessels can be observed *** the specific performance of LR in multimodal imaging fundus examinations can help diagnose more patients with abnormal *** SUMMARY A 29-year-old woman with type 2 diabetes presented to our clinic complaining of a six-day loss of visual acuity in the left *** fundus color images showed typical LR:Arteries and veins were the same pink-white *** images showed hyperinfrared reflections of the arteries and *** coherence tomography(OCT)showed numerous high point-like reflections in the retinal section,corresponding to different calibers of blood vessel *** reflections were seen in the big vessels of the *** fluorescein angiography(FFA)and optical coherence tomography angiography(OCTA)showed no significant *** examination found a total cholesterol level of 13.98 mmol/L,triglyceride 20.55 mmol/L,which confirmed the diagnosis of *** treatment to lower blood lipids and control blood glucose,the fundus imaging showed that the blood lipids in the patient had returned to *** LR shows specific changes in fundus color photography,infrared photography,and *** and OCTA were not sensitive to LR changes.

Loss of Anterior Chamber-Associated Immune Deviation during Normal Pregnancy in Primates

Purpose: We recently found that loss of anterior chamber-associated immune deviation was associated with normal pregnancy in rabbits. The purpose of this study is to further investigate whether the same events occurred in nonhuman primates. Methods: Mid-pregnant cynomolgus monkeys were randomly selected. Bovine serum albumin (BSA) was inoculated in anterior chamber of eyes of nonpregnant and mid-pregnant monkeys that were subsequently immunized with BSA in adjuvant. Delayed-type hypersensitivity (DTH) was assessed by skin ***: Non-pregnant monkeys of intracameral BSA proved able to acquire antigen-specific suppression of delayed-type hypersensitivity. By contrast, inoculation of BSA to anterior chamber of pregnant monkeys abolished the DTH-suppression effect. Conclusions: This is the first demonstration in primates that loss of anterior chamber-associated immune deviation occurred during normal pregnancy. Bimanual microincision cataract surgery with implantation of the new Incise~? MJ14 intraocular lens through a 1.4 mm incision

AIM：To analyze the visual outcomes and the posterior capsule opacification（PCO）with the new Incise？MJ14intraocular lens（IOL）implanted through a 1.4 mm clear corneal incision（CCI）in patients who underwent bimanual microincision cataract surgery（B-MICS）.METHODS：Eighty eyes which underwent cataract surgery using B-MICS technique performed by the same experienced surgeon were included in the study：40 eyes were implanted with an Incise？MJ14 IOL through a 1.4 mm CCI（group A）without enlargement of the main CCI,while 40 eyes were implanted with an Akreos？MI60 IOL with enlargement of the main CCI to 1.8 mm（group B）.Best corrected visual acuity（BCVA）,astigmatism and endothelial cell loss were evaluated before and after surgery at 7,30d and *** segment-optical coherence tomography（AS-OCT）of CCI was performed at 1,3,7,30d,6 and *** incidence was evaluated at 18mo using EPCO 2000 ***：Mean BCVA improvement and endothelial cell loss were statistically significant at 18mo in both groups with no difference between the two groups;no statistically significant difference in surgically induced astigmatism（SIA）was noticed in the two *** AS-OCT the only significant alterations in the CCI were endothelial gaping and local detachment of Descemet’s membrane at 1 and 7d after surgery;no statistically significant alterations were found at 1,6 and *** score at 18mo was 0.03±0.07for group A and 0.08±0.18 for group B（P=0.11）with no sign of central optic plate invasion in both ***：The implant of the new Incise？MJ14 IOL through a 1.4 mm CCI and B-MICS technique appeared to be a safe and effective procedure with rapid visual *** rate resulted very low and the CCI presented few morphological alterations which were only detectable in the first days postoperatively and achieved fast corneal healing during the long-term follow-up.

Pan-retinal ganglion cell markers in mice, rats, and rhesus macaques

Univocal identification of retinal ganglion cells(RGCs) is an essential prerequisite for studying their degeneration and neuroprotection. Before the advent of phenotypic markers, RGCs were normally identified using retrograde tracing of retinorecipient areas. This is an invasive technique, and its use is precluded in higher mammals such as monkeys. In the past decade, several RGC markers have been described. Here, we reviewed and analyzed the specificity of nine markers used to identify all or most RGCs, i.e., pan-RGC markers, in rats, mice, and macaques. The best markers in the three species in terms of specificity, proportion of RGCs labeled, and indicators of viability were BRN3A, expressed by vision-forming RGCs, and RBPMS, expressed by vision-and non-vision-forming RGCs. NEUN, often used to identify RGCs, was expressed by non-RGCs in the ganglion cell layer, and therefore was not RGC-specific. γ-SYN, TUJ1, and NF-L labeled the RGC axons, which impaired the detection of their somas in the central retina but would be good for studying RGC morphology. In rats, TUJ1 and NF-L were also expressed by non-RGCs. BM88, ERRβ,and PGP9.5 are rarely used as markers, but they identified most RGCs in the rats and macaques and ERRβ in mice. However, PGP9.5 was also expressed by non-RGCs in rats and macaques and BM88 and ERRβ were not suitable markers of viability.

Accuracy of biometric formulae for intraocular lens power calculation in a teaching hospital

AIM: To evaluate the accuracy of three commonly used biometric formulae across different axial lengths(ALs) at one United States Veterans Affairs teaching ***: A retrospective chart review was conducted from November 2013 to May 2018. One eye of each patient who underwent cataract surgery with a monofocal intraocular lens(IOL) was included. The range of postoperative follow-up period was from 3 wk to 4 mo. The Holladay 2, Barrett Universal II, and Hill-Radial Basis Function(Hill-RBF) formulae were used to predict the postoperative refraction for all cataract surgeries. For each formula, we calculated the prediction errors [including mean absolute prediction error(MAE)] and the percentage of eyes within ±0.25 diopter(D) and ±0.5 D of predicted refraction. We performed subgroup analyses for short(AL25.0 mm).RESULTS: A total of 1131 patients were screened, and 909 met the inclusion criteria. Resident ophthalmologists were the primary surgeons in 710(78.1%) cases. We found no statistically significant difference in predictive accuracy among the three formulae over the entire AL range or in the short, medium, and long eye subgroups. Across the entire AL range, the Hill-RBF formula resulted in the lowest MAE(0.384 D) and the highest percentage of eyes with postoperative refraction within ±0.25 D(42.7%) and ±0.5 D(75.5%) of predicted. All three formulae had the highest MAEs(>0.5 D) and lowest percentage within ±0.5 D of predicted refraction(<55%) in short ***: In cataract surgery patients at our teaching hospital, three commonly used biometric formulae demonstrate similar refractive accuracy across all ALs. Short eyes pose the greatest challenge to predicting postoperative refractive error.

Corneal collagen cross‑linking epithelium‑on ***‑off:a systematic review and meta‑analysis

Background:The purpose of the study was to determine the advantages and disadvantages of epi-on corneal crosslinking(CXL)techniques compared with standard epi-off ***:We searched MEDLINE and EMBASE for randomized controlled trials(RCTs)and non-randomized studies of interventions(NRSIs)and we evaluated the selected papers according to the Cochrane risk of bias *** considered,as primary outcomes,average Kmax flattening,changes in uncorrected and corrected distance visual acuity(UDVA and CDVA);as secondary outcomes,we considered changes in pachymetry values and endothelial cell density(ECD).We also investigated adverse events related to the treatments and treatment ***-analysis was conducted with a fixed or random-effects model using weighted mean difference(MD)with 95%confidence interval(CI)as the effect ***:A total of 15 studies were included and among these 15 trials,9 were RCTs and 6 were NRSIs,but only 4 studies showed no high risk of bias and were included in this *** analysis revealed significant postoperative differences in CDVA(MD=0.07;95%CI 0.04 to 0.10;P0.05).Epi-on CXL protocol was found to be significantly less prompt to have risks of delay in epithelial healing(P=0.035)and persistent stromal haze(P=0.026).Conclusion:Epi-on CXL is as effective as epi-off *** for a higher significant improvement in CDVA with current epi-on protocols,our meta-analysis demonstrates that epi-on and epi-off CXL have comparable effects on visual,topographic,pachymetric,and endothelial ***-on CXL has clinical advantages in terms of comfort and avoidance of complications as it reduces the risk of developing delay in epithelial healing and persistent stromal haze.

Induction of oxidative stress in human aqueous and vitreous humors by Nd:YAG laser posterior capsulotomy

AIM： To evaluate whether the Q-switched Nd：YAG laser treatment applied in routine capsulotomy elicits oxidative stress in aqueous and vitreous humors. METHODS： Thirty-six patients who had to undergo a 25 gauge pars plana vitrectomy due to vitreoretinal disorders were enrolled, 15 of them underwent a Q-switched Nd：YAG laser capsulotomy 7 d before vitrectomy due to posterior capsule opacification（PCO）（Nd：YAG laser group） while the remaining 21 patients were not laser treated before vitrectomy（no Nd：YAG laser group）. Samples of the aqueous and vitreous humors were collected during vitrectomy from all patients for the assessment of oxidative parameters which were compared between the Nd：YAG laser group and no Nd：YAG laser group. Thiobarbituric acid reactive substances（TBARS）, a product of membrane lipid peroxidation, nitrite levels, the antioxidative activities of SOD and catalase, the 4-HNE-protein conjugate formation, indicating structural modifications in proteins due to lipoperoxidation, were assessed in aqueous and vitreous samples. RESULTS： In the human vitreous humor TBARS levels are significantly higher in the Nd：YAG laser group compared to the no Nd：YAG laser group and importantly, there is a significant correlation between the TBARS levels and the total energy of Nd：YAG laser used during *** the anti-oxidative activities of SOD and catalase were significantly decreased by Nd：YAG laser treatment, both in aqueous and vitreous humors. In accordance with the TBARS data and anti-oxidative enzyme activities, significantly higher levels of proteins were conjugated with the lipoperoxidation product 4-HNE in the aqueous and vitreous humors in the Nd：YAG laser-treated group in comparison to no Nd：YAG laser group. CONCLUSION： These data, clearly suggest that any change that Q-switched Nd：YAG photo disruption may cause in the aqueous and vitreous compartments, resulting in a higher level of oxidative damage might be of considerable clinical significance
