Background: Huaier granule is an important medicinal fungus extract widely used in cancer treatment. Previous retrospective studies have reported its effectiveness in breast cancer patients, but the imbalanced baseline characteristics of participants could have biased the results. Therefore, this retrospective study aimed to examine the efficacy of Huaier granule on the prognosis of breast cancer ***: In this single-center cohort study, breast cancer patients diagnosed and treated at the Guangdong Provincial hospital of chinese medicine between 2009 and 2017 were selected. The data were retrospectively analyzed and divided into two groups according to whether the patients received Huaier granules. The propensity score matching (PSM) method was used to eliminate selection bias. The disease-free survival (DFS) and overall survival (OS) for these groups were compared using the Kaplan–Meier method and the Cox ***: This study included 214 early invasive breast cancer patients, 107 in the Huaier group and 107 in the control group. In the Kaplan–Meier analysis, the 2-year and 5-year DFS rates were significantly different in the Huaier group and control group (hazard ratio [HR], 0.495; 95% confidence interval [CI], 0.257–0.953;P = 0.023). The 2-year and 5-year OS rates were also significantly different (HR, 0.308; 95% CI, 0.148–0.644;P = 0.001). On multivariable Cox regression, Huaier granule was associated with improved DFS (HR, 0.440; 95% CI, 0.223–0.868;P = 0.018) and OS (HR, 0.236; 95% CI, 0.103–0.540;P = 0.001).Conclusion: In this retrospective study, Huaier granules improved the DFS and OS of early invasive breast cancer patients, providing real-world evidence for further prospective studies on treating breast cancer with Huaier granules.

FSD1 inhibits glioblastoma diffuse infiltration through restriction of HDAC6-mediated microtubule deacetylation

The infiltration of glioblastoma multiforme(GBM) is predominantly characterized by diffuse spread, contributing significantly to therapy resistance and recurrence of GBM. In this study, we reveal that microtubule deacetylation, mediated through the downregulation of fibronectin type III and SPRY domain-containing 1(FSD1), plays a pivotal role in promoting GBM diffuse infiltration. FSD1 directly interacts with histone deacetylase 6(HDAC6) at its second catalytic domain, thereby impeding its deacetylase activity on α-tubulin and preventing microtubule deacetylation and depolymerization. This inhibitory interaction is disrupted upon phosphorylation of FSD1 at its Ser317 and Ser324 residues by activated CDK5, leading to FSD1 dissociation from microtubules and facilitating HDAC6-mediated α-tubulin deacetylation. Furthermore, increased expression of FSD1 or interference with FSD1 phosphorylation reduces microtubule deacetylation, suppresses invasion of GBM stem cells, and ultimately mitigates tumor infiltration in orthotopic GBM xenografts. Importantly, GBM tissues exhibit diminished levels of FSD1 expression, correlating with microtubule deacetylation and unfavorable clinical outcomes in GBM patients. These findings elucidate the mechanistic involvement of microtubule deacetylation in driving GBM cell invasion and offer potential avenues for managing GBM infiltration.

Objective: To evaluate the clinical efficacy and safety of Yangxue Qingnao Pills(YXQNP) combined with amlodipine in treating patients with grade 1 hypertension. Methods: This is a multicenter, randomized, double-blind, and placebo-controlled study. Adult patients with grade 1 hypertension of blood deficiency and Gan(Liver)-yang hyperactivity syndrome were randomly divided into the treatment or the control groups at a 1:1 ratio. The treatment group received YXQNP and amlodipine besylate, while the control group received YXQNP's placebo and amlodipine besylate. The treatment duration lasted for 180 days. Outcomes assessed included changes in blood pressure, chinese medicine(CM) syndrome scores, symptoms and target organ functions before and after treatment in both groups. Additionally, adverse events, such as nausea, vomiting, rash, itching, and diarrhea, were recorded in both groups. Results: A total of 662 subjects were enrolled, of whom 608(91.8%) completed the trial(306 in the treatment and 302 in the control groups). After 180 days of treatment, the standard deviations and coefficients of variation of systolic and diastolic blood pressure levels were lower in the treatment group compared with the control group. The improvement rates of dizziness, headache, insomnia, and waist soreness were significantly higher in the treatment group compared with the control group(P0.05). No serious treatment-related adverse events occurred during the study period. Effect of herb-partitioned moxibustion for primary dysmenorrhea:a randomized clinical trial

OBJECTIVE: To observe the effect of herb-partitioned moxibustion(HPM) for primary ***: Six hundred and forty patients were randomized assigned(1∶1) to HPM group and control group. Duration of treatment was 3 months with 3 month follow-up. The primary outcome was pain relief measured by visual analogue scale(VAS). The second outcomes were Cox Menstrual Symptom Scale(CMSS), menstrual pain duration and frequency of analgesics usage. The exploratory outcome included quality of life,RESULTS: After the 3-month treatment and follow-ups, the pain intensity measured by VAS was significantly reduced in both groups compared with baseline(P < 0.05), and it was significantly decreased in HPM group than that of control group(P < 0.001). The higher proportion of participants in the HPM group had a decrease of at least 50% in VAS at the end of treatment, as compared with the control group(P < 0.001). At the 3 rd and 6 th month, the menstrual pain duration, CMSS score and frequency of analgesics usage in HPM group were significantly lower than those of control group(P < 0.05). After 3 month treatment and follow-ups,the scores of physical, psychological, social and environmental domains were significantly increased than baseline in both groups(P < 0.05),and the sores of physical, psychological and environmental domains were significantly higher in HPM group than those of control group(P < 0.05).CONCLUSION: Herb-partitioned moxibustion reduced menstrual pain and improved quality of life,these were sustained for up to 3 months after treatment. Further research is needed to understand long term effect and the mechanism of the intervention.

Rehmannia has been shown to be clinically effective in treating neurodegenerative diseases; however, the neuroprotective mechanisms remain unclear. In this study, we established a model of neurodegenerative disease using PC12 cytotoxic injury induced by glutamate. The cells were treated with 20 mM glutamate in the absence or presence of water extracts of dried Rehmannia root of varying concentrations（70%, 50% and 30%）. The different concentrations of Rehmannia water extract significantly increased the activity of glutamate-injured cells, reduced the release of lactate dehydrogenase, inhibited apoptosis, increased the concentrations of NADH, NAD and ATP in cells, ameliorated mitochondrial membrane potential, and reduced the levels of light chain 3. Taken together, our findings demonstrate that Rehmannia water extracts exert a cytoprotective effect against glutamate-induced PC12 cell injury via energy metabolism-related pathways.
