Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Studying molecular recognition in aqueous environments is crucial for understanding biological processes. Herein, we present a series of N-doped endo-functionalized anthracene-based cavities by introducing inward-directed pyridine units. The subtle modifications can significantly affect the binding properties of these receptors, which is similar to the mutations in biological systems. Combining theoretical calculations and molecular recognition experiments demonstrated that these “mutations” have an impact on the hydrophobicity and charge distribution of the hosts, consequently significantly influencing molecular *** is crucial for understanding the intricate mechanisms of molecular-level interactions in biological receptors.

Evaluation of thermal sprayed coating using ultrasonic inspection by means of bottom echo back reflection

Thermal spraying technique is widely used in various mechanical parts as a surface reforming ***,as demand to maintain superior mechanical performance in harsh operating environment increases,the need for non-destructive evaluation method for thermal spray coating becomes more *** this purpose,we thinned the thickness of the thermal sprayed coating by abrasion with blasting and used ultrasonic inspection by means of bottom echo reflection for effective measurement of abrasion quantity in thermal sprayed *** results obtained are summarized as *** the thickness of thermal sprayed coating becomes thin,the echo height *** is because thermal sprayed coatings absorb ultrasonic *** energy absorbed by Al2O3 is smaller compared with Fe-13Cr *** sprayed coatings submerged in water have a lower echo height compared with *** mentioned above,the thermal sprayed coating thickness can be estimated using ultrasonic inspection by means of bottom echo back reflection.

Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SESN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SESN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SESN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SESN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

Effect of reagent vibrational excitation and isotope substitution on the stereo-dynamics of the Ba+HF→BaF+H reaction

Based on an extended London-Eyring-Polanyi-Sato （LEPS） potential energy surface （PES）, the Ba q-HF reaction has been studied by the quasi-classical trajectory （QCT） method. The reaction integral cross section as a function of collision energy for the Ba q- HF --＊ BaF q- H reaction is presented and the influence of isotope substitution on the differential cross sections （DCSs） and alignments of the product＇s rotational angular momentum have also been studied. The results suggest that the integral cross sections increase with increasing collision energy, and the vibrational excitation of the reagent has great influence on the DCS. In addition, the product＇s rotational polarization is very strong as a result of heavy-heavy-light （HHL） mass combination, and the distinct effect of isotope substitution on the stereodynamics is also revealed.

Two new cytotoxic sesquiterpenoids from Solanum lyratum

Acute central nervous system injuries,including ischemic stro ke,intracerebral hemorrhage,subarachnoid hemorrhage,traumatic brain injury,and spinal co rd injury,are a major global health *** optimal therapies and improving the long-term neurological functions of patients with acute central nervous system injuries are urgent *** are susceptible to damage after acute central nervous system injury,and this leads to the release of toxic levels of reactive oxygen species,which induce cell ***,a selective form of autophagy,is crucial in eliminating redundant or damaged mitochondria during these *** evidence has highlighted the significant role of mitophagy in acute central nervous system *** this review,we provide a comprehensive overview of the process,classification,and related mechanisms of *** also highlight the recent developments in research into the role of mitophagy in various acute central nervous system injuries and drug therapies that regulate *** the final section of this review,we emphasize the potential for treating these disorders by focusing on mitophagy and suggest future research paths in this area.

Preparation and Evaluation in vitro of Salicylic Acid-pachyman Nanoparticles

Pachyman based nanoparticles loading salicylic acid as model drug （SA-PNPs） were prepared by an inverse microemulsion crosslinking approach using epichlorohydrin （ECH） as crosslinker. The effects of crosslinking reaction time, initial volume ratio of oil to aqueous phase and dosage of crosslinker on the particle size of SA-PNPs were optimized by orthogonal experimental design. SA-PNPs prepared under the optimal conditions had the average size of 230 nm and high encapsulation efficiency of 90%. The in vitro drug release was also investigated and the release data were analyzed using zero order, first order and Higuchi＇s kinetics model. According to the determined coefficients, release data fitted to Higuchi＇s model, which suggested that the release of SA from SA-PNPs in phosphate buffer （pH 7.4） was diffusion controlled release. The experimental results indicated that pachyman possesses a promising potential to be applied as nanocarriers for controlled drug release.

Effects of antibiotic antitumor drugs on nucleotide levels in cultured tumor cells:an exploratory method to distinguish the mechanisms of antitumor drug action based on targeted metabolomics

Nucleotide pools in mammalian cells change due to the influence of antitumor drugs,which may help in evaluating the drug effect and understanding the mechanism of drug *** this study,an ion-pair RP-HPLC method was used for a simple,sensitive and simultaneous determination of the levels of 12 nucleotides in mammalian cells treated with antibiotic antitumor drugs(daunorubicin,epirubicin and dactinomycin D).Through the use of this targeted metabolomics approach to find potential biomarkers,UTP and ATP were verified to be the most appropriate ***,a holistic statistical approach was put forward to develop a model which could distinguish 4 categories of drugs with different mechanisms of *** model can be further validated by evaluating drugs with different mechanismsof *** targeted metabolomics study may provide a novel approach to predict the mechanism of action of antitumor drugs.
