Enhanced a novel β-agarase production in recombinant Escherichia coli BL21(DE3) through induction mode optimization and glycerol feeding strategy

Agarases are hydrolytic enzymes that act on the hydrolysis of agar and have a broad range of applications in food,cosmetics and pharmaceutical industries. In this study, a glycerol feeding strategy based on induction mode optimization for high cell density and β-agarase production was established, which could effectively control acetate yield. First, exponential feeding strategy of glycerol with different overall specific growth rates（μ） was applied in the pre-induction phase. The results showed that the low μ（μ=0.2） was suggested to be the optimal for cell growth and β-agarase production. Second, the effects of induction temperature and the inducer concentration on cell growth and β-agarase production were investigated in the post-induction phase. When induced by isopropyl-β-d-thiogalactoside（IPTG）, the strategy of 0.8 mmol/L IPTG induction at 20℃ was found to be optimal for β-agarase production. When cultivation was induced by continuous lactose feeding strategy of 1.0 g/（L·h）, the β-agarase activity reached 112.5 U/mL, which represented the highest β-agarase production to ***, the β-agarase was capable of degrading G. lemaneiformis powder directly to produce neoagarooligosaccharide, and the hydrolysates were neoagarotetraose（NA4） and neoagarohexaose（NA6）. The overall research may be useful for the industrial production and application of β-agarase.

Currently,the manual contact rail measurement that was basically adopted in China has low detection efficiency,poor accuracy and poor *** order to improve the function of the system,we propose a non-contact measurement method based on the flatness and verticality ruler *** flatness measurement model was built by employing the string measurement *** addition,the verticality measurement model was built by the dihedral method to measure the rail *** extracting curvature information of feature points,in this system,each laser sensor is used to collect rail profile curves.A large number of three-dimensional point clouds data are generated by the unit quaternion method of coordinate transformation,and the contour curves of the characteristic points of the four laser sensors are matched with the corresponding point sets one to one,and the rail contour splicing is finally *** experimental results show that this method has better measurement effect compared with the traditional manual measurement method.

Objective:To evaluate the existing randomized controlled trials(RCTs)for evidence of the efficacy and safety of head acupuncture(HA)plus Schuell’s language rehabilitation(SLR)in post-stroke ***:Seven databases including Embase,PubMed,Cochrane Library,Technology Periodical Database,the China National Knowledge Infrastructure,SinoMed and Wanfang Data Information Site were searched for RCTs published from database inception until November 14,*** that compared HA plus SLR with sham(or blank)control,acupuncture therapy alone,certain language rehabilitation therapy alone or other therapies for post-stroke aphasia were *** were extracted and assessed,and the quality of RCTs was ***-effects model was used,with meta-influence analysis,meta-regression,and regression-based sub-group analyses applied for exploration of *** bias was estimated by funnel plots and Egger’s ***:A total of 32 RCTs with1,968 patients were included and 51 comparisons were conducted classified as types of strokes and aphasia.(1)For patients with aphasia after ischemic stroke,HA plus PSA showed significantly higher accumulative markedly effective rate[relative risk(RR)=1.55,95% confidence interval(CI):1.19-2.02,I^(2)=0%]and accumulative effective rate(RR=1.22,95%CI:1.09-1.36,I^(2)=0%).(2)For patients with comprehensive types of stroke,HA plus PSA was more effective in increasing recovery rate(RR=1.89,95%CI:1.39-2.56,I^(2)=0%),accumulative markedly effective rate(RR=1.53,95%CI:1.36-1.72,I^(2)=9%)and accumulative effective rate(RR=1.14,95%CI:1.09-1.19,I^(2)=34%).(3)For patients with aphasia after stroke,HA plus PSA was superior to PSA alone with statistical significance in increasing recovery rate(RR=2.08,95%CI:1.24-3.46,I^(2)=0%),accumulative markedly effective rate(RR=1049,95%CI:1.24-1.78,I^(2)=0%)and accumulative effective rate(RR=1.15,95%CI:1.06-1.24,12=39%).(4)For patients with multiple types of aphasia,HA plus PSA also demonstrat

Highly sensitive ethanol sensors have been widely utilized in environmental protection,industrial monitoring,and drink-driving *** this work,a fully self-powered ethanol detector operating at room temperature has been developed based on a triboelectric nanogenerator(TENG).The gas-sensitive oxide semiconductor is selected as the sensory component for the ethanol detection,while the resistance change of the oxide semiconductor can well match the“linear”region of the load characteristic curve of ***,the output signal of TENG can directly reveal the concentration change of ethanol *** accelerator gearbox is applied to support the operation of the TENG,and the concentration change of ethanol gas can be visualized on the Liquid Crystal *** fully self-powered ethanol detector has excellent durability,low fabrication cost,and high selectivity of 5 ***,the ethanol detector based on TENG not only provides a different approach for the gas detection but also further demonstrates the application potential of TENG for various sensory devices.

Polygala tenuifolia is a perennial medicinal plant that has been widely used in traditional Chinese medicine for treating mental ***,the lack of genomic resources limits the insight into its evolutionary and biological *** the present work,we reported the *** genome,the first genome assembly of the Polygalaceae *** sequenced and assembled this genome by a combination of Illumnina,PacBio HiFi,and Hi-C *** assembly includes 19 pseudochromosomes covering∼92.68%of the assembled genome(∼769.62 Mb).There are 36463 protein-coding genes annotated in this *** comparative genome anal-ysis revealed that *** experienced two rounds of whole genome duplication that occurred∼39–44 and∼18–20 million years ago,***,we systematically reconstructed ancestral chromosomes of *** and inferred its chromosome evolution trajectories from the common ancestor of core eudicots to the present *** on the transcriptomics data,enzyme genes and transcription factors involved in the synthesis of triterpenoid saponin in *** were *** analysis demonstrated that whole-genome duplications and tandem duplications play critical roles in the expansion of P450 and UGT gene families,which contributed to the synthesis of triterpenoid *** genome and transcriptome data will not only provide valuable resources for comparative and functional genomic researches on Polygalaceae,but also shed light on the synthesis of triterpenoid saponin.
