Objective Recent studies have suggested that the gut microbiota is altered in children with juvenile idiopathic arthritis(JIA). However, age, sex, body mass index(BMI) were not matched in the previous studies, and the results are inconsistent. We conducted an age-, sex-, and BMI-matched cross-sectional study to characterize the gut microbiota in children with JIA, and evaluate its potential in clinical *** A total of 40 patients with JIA and 42 healthy controls, ranging from 1 to 16 years, were enrolled in this study at two tertiary hospitals in Zhejiang province, China. Fecal samples were collected for 16 S rDNA sequencing. The data was analyzed using QIIME software and R packages. Specifically, the random forest, a commonly used machine learning method, was used to identify biomarkers, and the receiver operating characteristic curve and the decision curve analysis were used to evaluate model *** The Shannon-Wiener index in the JIA group was smaller than that in the control group(p = 0.031). The Bray-Curtis dissimilarity differed significantly between the two groups(p = 0.019, R2 = 0.021). The relative abundance of four genera, Anaerostipes, Dialister, Lachnospira, and Roseburia, decreased significantly in the JIA group compared with those in the control group(adjusted p = 0.031, 0.013, 0.041, and 0.011, respectively). The four genera were microbes that produce short-chain fatty acids(SCFAs) and were correlated with some rheumatic indices. Moreover, twelve genera were identified as potential biomarkers by using the nested cross-validation function of the random forest. A random forest model constructed using these biomarkers was able to differentiate the patients with JIA from the healthy controls, and the area under the receiver operating characteristic curve was 0.7975. OBJECTIVE:To evaluate the efficacy and safety of Zhumian Tang formula granules(助眠汤配方颗粒)combined with eszopiclone for treating poor sleep ***:This multi-center,dynamic blockrandomized,parallel-group superiority clinical trial included 130 *** combined treatment group received Zhumian Tang formula granules combined with eszopiclone treatment,and the control group received eszopiclone treatment *** group allocation ratio was 1∶*** duration of treatment was 2 *** were asked to complete questionnaires before treatment,after 1 week of the intervention,after 2 weeks of the intervention,and at the follow-up on week *** primary outcomes were the Pittsburgh Sleep Quality Index(PSQI)score and the total effective rate of *** secondary outcome was the rate of adverse ***:Compared with the eszopiclone treatment group,the PSQI score of the combined treatment group was significantly lower after 2 weeks of the intervention(6.98 vs 8.26,P<0.05).However,there was no significant difference in the mean PSQI score after 1 week of the intervention(9.89 vs 9.15,P=0.124).After the follow-up on week 3,the PSQI score of the combined treatment group remained significantly lower than that of the eszopiclone treatment group(6.12 vs 8.31,P<0.001).The total effective rates of treatment of the combined group and the eszopiclone group were 36.92%vs 35.38%(Z=0.033,P=0.855)after 1 week of the intervention,83.08%vs 58.46%(Z=9.519,P<0.05)after 2 weeks of the intervention,and 83.08%vs 61.54%(Z=7.530,P<0.05)and after the follow-up on week 3,*** was no significant difference in the overall rate of adverse reactions between the combined and eszopiclone treatment groups(21.53%vs 31.8%,P=0.318).CONCLUSION:The combination of Zhumian Tang formula granules with eszopiclone was found to be safe and more effective in improving sleep quality than eszopiclone *** Chinese medicine can enhance the effectiveness of Western m

Intestinal dysbiosis exacerbates the pathogenesis of psoriasis-like phenotype through changes in fatty acid metabolism

Qualitative and quantitative assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream by HPLC-TOF-MS and HPLC

Related substances in pharmaceutical formulations are associated with their safety, efficacy and stability. However, there is no overall study already published on the assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream. In this work, a reliable HPLC-TOF-MS qualitative method was developed for the analysis of related substances in this preparation with a quick and easy extraction procedure. Besides the active pharmaceutical ingredients, two compounds named ketoconazole impurity B′ optical isomer and ketoconazole impurity E were identified. Furthermore, a new HPLC method for qualitative and quantitative assessment on related substances and degradation products, which were found in the stability test, was established and validated. The single standard to determine multi-components method was applied in the quantitative analysis, which was an effective way for reducing cost and improving accuracy. This study can provide a creative idea for routine analysis of quality control of the Compound Ketoconazole and Clobetasol Propionate Cream.

Ionic liquids on uncharged and charged surfaces: In situ microstructures and nanofriction

In situ changes in the nanofriction and microstructures of ionic liquids(ILs)on uncharged and charged surfaces have been investigated using colloid probe atomic force microscopy(AFM)and molecular dynamic(MD)*** representative ILs,[BMIM][BF_(4)](BB)and[BMIM][PF_(6)](BP),containing a common cation,were selected for this *** torsional resonance frequency was captured simultaneously when the nanoscale friction force was measured at a specified normal load;and it was regarded as a measure of the contact stiffness,reflecting in situ changes in the IL microstructures.A higher nanoscale friction force was observed on uncharged mica and highly oriented pyrolytic graphite(HOPG)surfaces when the normal load increased;additionally,a higher torsional resonance frequency was detected,revealing a higher contact stiffness and a more ordered IL *** nanofriction of ILs increased at charged HOPG surfaces as the bias voltage varied from 0 to 8 V or from 0 to−8 *** simultaneously recorded torsional resonance frequency in the ILs increased with the positive or negative bias voltage,implying a stiffer IL layer and possibly more ordered ILs under these *** simulation reveals that the[BMIM]+imidazolium ring lies parallel to the uncharged surfaces preferentially,resulting in a compact and ordered IL *** parallel“sleeping”structure is more pronounced with the surface charging of either sign,indicating more ordered ILs,thereby substantiating the AFM-detected stiffer IL layering on the charged *** in situ observations of the changes in nanofriction and microstructures near the uncharged and charged surfaces may facilitate the development of IL-based applications,such as lubrication and electrochemical energy storage devices,including supercapacitors and batteries.

Water Cooperation Priorities in the Lancang-Mekong River Basin Based on Cooperative Events Since the Mekong River Commission Establishment

The Lancang-Mekong River has attracted much attention from researchers, but the cooperation on water issues in this river basin has been limited, even after the establishment of the Mekong River Commission(MRC). Cooperation on water resources has been determined as one of the key priority areas in the Lancang-Mekong Cooperation Mechanism, but there are no details of targets. In order to establish the priorities of water cooperation under the mechanism, we adopted nine categories to classify the objectives of 87 water cooperation events based on the ‘Lancang-Mekong Water Cooperative Events Database' from 1995 to 2015. Based on the occurrence of cooperative events, cooperative objectives, cooperative scales, and approaches to cooperation, we conducted statistical, correlation, and text analyses. Our analyses indicated the following results: under the impact of economic conditions inside and outside the river basin, full cooperation appeared more difficult than bilateral and multilateral cooperation. Each of the partners adopted different preferences for cooperation targets. Cooperation with more definite objectives was easier to establish than cooperation with broader and more complex objectives. The potential objectives for water cooperation were navigation, hydropower, joint management, data sharing, flood control and water use. Because hydropower development is controversial, and because water cooperation is avoided by most existing regional cooperation mechanisms due to its complexity, we suggest the following priority areas for water cooperation in the Lancang-Mekong River Basin. 1) Navigation and flood control/drought relief are attractive objectives for all the riparian countries across the whole watershed. 2) Data sharing should be a priority for cooperation in the watershed due to its laying the foundation for the equitable and reasonable utilization of transboundary waters. 3) Hydropower is an objective best implemented mainly through bilateral cooperation,

Hepatitis B immunoglobulin （HBIG） is important in the management of hepatitis B virus （HBV） infection. Aiming to develop recombinant monoclona# antibodies as an alternative to HBIG, we report the successful identification of HBV surface antigen （HBsAg）-specific antibodies from a full-length human antibody library displayed on mammalian cell surface. Using total RNA of peripheral blood mononuclear cells of a natively immunized donor as template, the antibody repertoire was amplified. Combining four-way ligation and the FIp recombinase-mediated integration （FIp-ln） system, we constructed a mammalian cell-based, fully human, full-length antibody display library in which each cell displayed only one kind of antibody molecule. By screening the cell library using fluorescence-activated cell sorting （FACS）, eight cell clones that displayed HBsAg-specific antibodies on cell surfaces were identified. DNA sequence analysis of the antibody genes revealed three unique antibodies. FACS data indicated that fluorescent strength of expression （FSE）, fluorescent strength of binding （FSB） and relative binding ability （RBA） were all different among them. These results demonstrated that by using our antibody mammalian display and screening platform, we can successfully identify antigen-specific antibodies from an immunized full-length antibody library. Therefore, this platform is very useful for the development of therapeutic antibodies.

Constructing parallel long-message signcryption scheme from trapdoor permutation

A highly practical parallel signcrypUon scheme named PLSC from trapdoor permutations （TDPs for short） was built to perform long messages directly. The new scheme follows the Idea ＂scramble all, and encrypt small＂, using some scrambling operation on message m along with the user＇s Identities, and then passing, In paraliel, small parts of the scrambling result through corresponding TOPs. This design enables the scheme to flexibly perform long messages of arbitrary length while avoid repeatedly invoking TDP operations such as the CBC mode, or verbosely black-box composing symmetric encryption and slgncryption, resulting in noticeable practical sevlngs in both message bandwidth and efficiency. Concretely, the signcryptlon scheme requires exactly one computation of the ＂receiver＇s TDP＂（for ＂encryptlon＂） and one Inverse computation of the ＂sender＇s TDP＂（for ＂authentication＂）, which Is of great practical significance in directly performing long messages, since the major bottleneck for many public encryptlon schemes is the excessive computational overhead of performing TDP operations. Cutting out the verbosely repeated padding, the newly proposed scheme Is more efficient than a black-box hybrid scheme. Most importantly, the proposed scheme has been proven to be tightly semanUcaiiy secure under adaptive chosen clphertext attacks （iND-CCA2） and to provide integrity of clphertext （INT-CTXT） as well as non-repudiation in the random oracle model. All of these security guarantees are provided in the full multi-user, insider-security setting. Moreover, though the scheme is designed to perform long messages, it may also be appropriate for settings where It is Impractical to perform large block of messages （i.e. extremely low memory environments such as smart cards）.
