Melittin induces human gastric cancer cell apoptosis via activation of mitochondrial pathway

AIM: To investigate the apoptotic effects of melittin on SGC-7901 cells via activation of the mitochondrial signaling pathway in ***: SGC-7901 cells were stimulated by melittin, and its effect on proliferation and apoptosis of was investigated by methyl thiazolyl tetrazolium assay, morphologic structure with transmission electron microscopy, annexin-V/propidium iodide double-staining assay, measuring mitochondrial membrane potential(MMP) levels, and analyzing reactive oxygen species(ROS) concentrations were analyzed by flow cytometry. Cytochrome C(Cyt C), apoptosis-inducing factor(AIF), endonuclease G(Endo G), second mitochondria-derived activator of caspases(Smac)/direct IAP binding protein with low isoelectric point(Diablo), and FAS were analyzed by western blot. The expression of caspase-3 and caspase-8 was measured using activity assay ***: Melittin was incubated at 1.0, 2.0, 4.0, or 6.0 μg/m L for 1, 2, 4, 6, or 8 h and showed a timeand concentration-dependent inhibition of SGC-7901 cell growth. Melittin induced SGC-7901 cell apoptosis, which was confirmed by typical morphological changes. Treatment with 4 μg/m L melittin induced early apoptosis of SGC-7901 cells, and the early apoptosis rates were 39.97% ± 3.19%, 59.27% ± 3.94%, and 71.50% ± 2.87% vs 32.63% ± 2.75% for 1, 2, and 4 h vs 0 h(n = 3, P < 0.05); the ROS levels were 616.53% ± 79.78%, 974.81% ± 102.40%, and 1330.94% ± 93.09% vs 603.74% ± 71.99%(n = 3, P < 0.05); the MMP values were 2.07 ± 0.05, 1.78 ± 0.29, and 1.16 ± 0.25 vs 2.55 ± 0.42(n = 3, P < 0.05); caspase-3 activity was significantly higher compared to the control(5492.3 ± 321.1, 6562.0 ± 381.3, and 8695.7 ± 449.1 vs 2330.0 ± 121.9), but the caspase activity of the non-tumor cell line L-O2 was not different from that of the control. With the addition of the caspase-3 inhibitor(Ac-DEVD-CHO), caspase-3 activity was significantly decreased compared to the control group(1067.0 ± 132.5 U/g vs 8695.7 ± 449.1 U/g). The expression

Flavonoids with cytotoxicities from the seeds of Pongamia pinnata(L.)Pierre

Twenty-eight compounds,including flavones(2,4-5,7-11),flavonols(1,3,6,21,22),flavanones(12-18),isoflavones(19,20),chalcones(23,24),phenylpropanoids(25,26),and others(27,28)were isolated from the ethanol extract of the seeds of Pongamia pinnata(L.)Pierre and identified on the basis of physic-chemical constants and spectral analysis(NMR,ECD,[α]^20 D).Among them,compounds 16,18,21,23,25 and 26 were obtained from the genus for the first *** cytotoxicities of the purified flavonoids against H292 cells were evaluated using MTT *** a result,compounds 14 and 15 displayed moderate cytotoxicities.

＜正＞The development of kernel methods,in particular support vector machines,has been further extended into a new class of methods,which could effectively solve the nonlinear problems in chemistry by using simple linear transformation manner+.In fact,the kernel function used in kernel methods might be regarded as a general protocol to deal with nonlinear data in *** this paper,the basic idea and modularity of kernel methods,together with some simple examples,are discussed in detail to give an in-depth understanding for kernel *** key ingredients of kernel methods,namely dual form,nonlinear mapping and kernel function,construct a consistent framework of the kernel-based algorithms（See Figure 1）.The modularity of kernel methods allows linear algorithms to combine with any kernel ***,some commonly used chemometrics algorithms are easily extended into their kernel versions.

Monostatic and Bistatic Low Frequency Ultrawideband SAR Imaging Experiment

Low frequency ultrawideband (LF UWB) synthetic aperture radar (SAR) has lately become of a particular interest to SAR community. Monostatic and bistatic LF UWB SAR system has the well foliage penetrating capability, high-resolution imaging and providing the increased information. In 2015, a monostatic and bistatic LF UWB SAR imaging experiment was conducted. In this experiment, the monostatic and bistatic data were collected simultaneously by operating a moving vehicle-based radar in the SAR mode, in conjunction with a stationary ground-based receiver. The aim was to investigate the imaging property of the bistatic LF UWB SAR system. The one pulse per second (1 PPS) signal in combination with the global position system (GPS) disciplined 100 MHz oscillator from the GPS receivers was used to implement the time and frequency synchronization in this SAR system. The bistatic SAR image was obtained by the subaperture spectrum-equilibrium method integrated with the fast factorized back projection (FFBP) algorithm. Bistatic experiment results are show to prove the validity of the bistatic LF UWB SAR imaging experiment.

Chan-su(toad venom) has been used as an analgesic agent in China from ancient to modern ***,a non-peptide toxin extracted from toad venom,is considered as one of the analgesic *** molecular mechanism underlying the anti-nociceptive effects of bufalin remains unclear so *** the present study,we investigated the pharmacological effects of bufalin on painrelated ion channels as well as animal models through patch clamp,calcium imaging and animal behavior *** the whole-cell recording,bufalin caused remarkable suppressive effect on the peak currents of voltage-gated sodium channels(VGSCs) of dorsal root ganglion neuroblastomas(ND7-23 cell) in a dose-dependent *** prompted the voltage-dependent activation and induced a negative shift on the fast inactivation of *** recovery kinetics of VGSCs were significantly slowed and recovery proportion were reduced after administering *** bufalin produced insignificant effect not only on voltage-dependent Kv4.2,Kv4.3 and BK channels heterologously expressed in HEK293 T cells,but also on the capsaicin and allyl isothiocyanate(AITC)induced Ca2+***’s more,bufalin could observably relieve formalin-induced spontaneous flinching and licking response as well as carrageenan-induced thermal and mechanical hyperalgesia in dose-dependent manner,which was in agreement with the results of in vitro *** present results suggest that the remarkable anti-nociceptive effects produced by bufalin are probably ascribed to its specific regulation on voltage-gated sodium *** inhibited the VGSCs in a dose-dependent manner,which provide references for the optimal dose selection of analgesia drugs.

An Improved CFD-synthesis Method for Predicting Inlet Dynamic Distortion

This article presents an improved computational fluid dynamics （CFD）-synthesis method to predict dynamic distortion. A steady-state flow field is derived from a CFD solution, through which are acquired total pressure, density, turbulence kinetic energy and others in steady-state at an aerodynamic interface plane （AIP）. Back-propagation artificial neural network （BP ANN） is used to find out the relationship between the measured flight turbulence and the CFD-computed turbulence parameters. The dynamic pressure is obtained by incorporating CFD-found total steady-state pressure with fluctuating pressure. Finally, the dynamic distortion is predicted by means of the synthesized dynamic pressure. The fairly good agreement between the computed inlet surface pressure and the flight test data bears out the reliability of CFD solution used in this article. To validate the proposed method, six sets of flight test data are used and the results show that the predicted dynamic distortion is well in line with the distortion displayed in flight tests. An examination of the traditional method is also accomplished and the comparison also shows that the proposed method is superior to the traditional one in higher consistency with flight test data.
