The optimum concentration of Na_2 WO_4 was explored in relation to the cell density and astaxanthin content in Haematococcus pluvialis. Then, the cellular morphology, nitrate reductase(NR) activity, soluble sugar and protein contents, and chlorophyll ?uorescence were measured, and the transcriptional expression of carotenogenic genes was determined by quantitative real-time PCR. The results showed that 3.0 mmol/L of Na_2 WO_4 was the optimum concentration to induce astaxanthin accumulation, with a maximum content of 49.41±0.13 pg/cell reached on the tenth day. The NR activity decreased signi?cantly and continually after Na_2 WO_4 treatment. The soluble sugar content increased gradually during the experimental period and was eventually signi?cantly higher than that in the control. The soluble protein content increased rapidly,reached a maximum in day 0.5 and day 1 and then decreased. The ef fective photochemical effciency of PSII( F v'/F m') and light saturation( E k) ?rst decreased and then tended to stabilize, and NADP +-glyceraldehyde-3-phosphate dehydrogenase(GAPDH) gene expression was correlated with photosynthesis. The transcriptional expression of ipi, psy and bkt was signi?cantly increased compared with that in the control after application of Na_2 WO_4, and the relative expression of ipi reached the highest level on the ?fth day, with a 98.03±1.92-fold increase. Our results describe a new approach to promote the ef fective accumulation of astaxanthin in H. pluvialis by NR inhibitor Na_2 WO_4.

Copepod community structure during upwelling and non-upwelling seasons in coastal waters off Cochin, southwest coast of India

Vypeen, an island of Cochin estuarine system, acts as the spawning site of several marine and estuarine fishes. We assumed that, physical process(upwelling) make changes in hydrography and the production of chlorophyll a in coastal waters off Vypeen. These alterations can influence the zooplankton abundance and copepod community structure in that area. For justifying this hypothesis, samples were collected from Vypeen at 10 m and 30 m locations during January(pre-southwest monsoon), August(late-southwest monsoon) and November(postsouthwest monsoon) 2014. During August, subsurface water column was cool, nutrient rich and less oxygenated(signatures of upwelling) than in November and January. Maximum concentrations of nitrate and chlorophyll a were recorded during August. In the present study, 15 zooplankton groups were recorded;of which copepods were the most predominant group(73%–90%). Copepod density in the present study ranged between 527.2 ind./m^3 and 5 139.2 ind./m^3. Totally, 37 copepods species were reported in present study and copepod species richness was higher during January. The abundance of zooplankton and copepods was high in August during late-southwest monsoon. These variations were closely associated with the coastal upwelling in August and weakening of moderate upwelling in November. In SIMPER analysis, it was found that the copepods species distribution was similar within season and dissimilar between the three seasons. Moreover, the higher abundance of upwelling indicator species Temora turbinata was recorded in August, which confirms the signs of seasonal upwelling in Vypeen. The present study emphasized on the influence of hydrographical parameters associated with physical process, in governing the copepod community organization of the Vypeen Island.

Twenty species of seaweed were collected from the coast of Zhejiang, China, extracted with ethanol, and screened for algicidal activity against red tide microalgae H eterosigma akashiwo and Prorocentrum micans. Inhibitory eff ects of fresh and dried tissues of green alga U lva intestinalis were assessed and the main algicidal compounds were isolated, purifi ed, and identifi ed. Five seaweed species, U. intestinalis, U. fasciata, Grateloupia romosissima, Chondria crassicaulis, and Gracilariopsis lemaneiformis, were investigated for their algicidal activities. Fresh tissues of 8.0 and 16.0 mg/m L of U. intestinalis dissolved in media signifi cantly inhibited growth of H. akashiwo and P. micans, respectively. Dried tissue and ethyl acetate(Et OAc) extracts of U. intestinalis at greater than 1.2 and 0.04 mg/m L, respectively, were fatal to H. akashiwo, while its water and Et OAc extracts in excess of 0.96 and 0.32 mg/m L, respectively, were lethal to P. micans. Three algicidal compounds in the Et OAc extracts were identifi ed as 15-ethoxy-(6z,9z,12z)-hexadecatrienoic acid(I),(6E,9E,12E)-(2-acetoxy- β- D-glucose)-octadecatrienoic acid ester(II) and hexadecanoic acid(III). Of these, compound II displayed the most potent algicidal activity with IC_(50) values of 4.9 and 14.1 μg/m L for H. akashiwo and P. micans, respectively. Compound I showed moderate algicidal activity with IC_(50) values of 13.4 and 24.7 μg/m L for H. akashiwo and P. micans, respectively. These fi ndings suggested that certain macroalgae or products therefrom could be used as ef fective biological control agents against red tide algae.

Taxonomic dependency and spatial heterogeneity in assembly mechanisms of bacteria across complex coastal waters

Background Understanding community assembly mechanisms across taxa and space is fundamental for microbial ecology. However, the variability and determinants of assembly processes over taxa and space remain unclear. Here, we investigated taxonomic dependency and spatial heterogeneity in bacterial assembly mechanisms across coastal waters in the East China Sea using neutral and null models with customized visualization *** Overall, bacterial assembly mechanisms varied across broad taxonomic groups(phyla and proteobacterial classes) and space at the regional scale. A determinism–stochasticity balanced mechanism governed total bacterial assembly, while taxonomic dependency existed in assembly mechanisms and ecological processes. Among community ecological features, niche breadth and negative-to-positive cohesion ratio were strongly associated with the determinism-to-stochasticity ratio of bacterial groups. Bacterial assembly mechanisms commonly exhibited spatial heterogeneity, the extent and determinants of which varied across taxonomic groups. Spatial assembly of total bacteria was directly driven by many environmental factors and potential interactions between taxa, but not directly by geographic factors. Overall, the bacterial groups with higher spatial heterogeneity in assembly mechanisms were more related to environmental and/or geographic factors(except Bacteroidetes), while those with lower heterogeneity were more related to ecological *** Our results confirm the pervasiveness of taxonomic dependency and spatial heterogeneity in bacterial assembly, providing a finer understanding about regulation across complex coastal waters.

Using a new functional analysis tool, multiscale window transform(MWT), and the MWT-based localized multiscale energetics analysis and canonical transfer theory, this study reconstructs the Kuroshio system on three scale windows, namely,the mean flow window, the interannual-scale(low-frequency) window, and the transient eddy window, and investigates the climatological characteristics of the intricate nonlinear interactions among these windows. Significant upscale energy transfer is observed east of Taiwan, where the mean Kuroshio current extracts kinetic energy from both the interannual and eddy *** is found that the canonical transfer from the interannual variability is an intrinsic source that drives the eddy activities in this region. The multiscale variabilities of the Kuroshio in the East China Sea(ECS) are mainly controlled by the interaction between the mean flow and the *** mean flow undergoes mixed instabilities(i.e., both barotropic and baroclinic instabilities) in the southern ECS, while it is barotropically stable but baroclinically unstable to the north. The multiscale interactions are found to be most intense south of Japan, where strong mixed instabilities occur; both the canonical transfers from the mean flow and the interannual scale are important mechanisms to fuel the eddies. It is also found that the interannual-scale energy mainly comes from the barotropically unstable jet, rather than the upscale energy transfer from the high frequency eddies.

Baroclinic wave activity in the North Pacific exhibit peaks in late fall and early spring, and a local minimum in midwinter, when by linear baroclinic instability theory it should attain its maximum. This counterintuitive phenomenon, or"midwinter suppression"(MWM) as called, is investigated with a functional analysis apparatus, multiscale window transform(MWT), and the MWT-based theory of canonical transfer and localized multi-scale energetics analysis, together with a feature tracking technique, using the data from the European Centre for Medium-Range Weather Forecasts ReAnalysis(ERA-40). It is found that the MWM results from a variety of different physical processes, including baroclinic canonical transfer, diabatic effect, energy flux divergence, and frictional dissipation. On one hand, baroclinic canonical transfer and diabatic effect achieve their respective maxima in late fall. More transient available potential energy is produced and then converted to transient kinetic energy, resulting in a stronger storm track in late fall than in midwinter. On the other hand, in early spring, although baroclinic instability and buoyancy conversion are weak, energy flux convergences are substantially strengthened, leading to a net energy inflow into the storm track. Meanwhile, frictional dissipation is greatly reduced in spring; as a result, less transient energy is dissipated in early spring than in midwinter. It is further found that the weakening of baroclinic canonical transfer in midwinter(compared to late fall) is due to the far distance between the storm and the jet stream(located at its southernmost point), which suppresses the interaction between them. Regarding the increase in energy flux convergence in early spring, it appears to originate from the increase(enhancement) in the number(strength) of storms from the upstream into the Pacific.

Effects of different dietary lipid sources on growth performance and tissue fatty acid composition of juvenile swimming crab Portunus trituberculatus

This study was conducted to evaluate the effects of dietary lipid sources on the growth performance and fatty acid composition of the swimming crab, P ortunus trituberculatus. Four isonitrogenous and isoenergetic experimental diets were formulated to contain four separate lipid sources, including fish, soybean, rapeseed, and linseed oils(FO, SO, RO, and LO, respectively). With three replicates of 18 crabs each for each diet, crabs(initial body weight, 17.00 ±0.09 g) were fed twice daily for 8 weeks. There were no significant differences among these groups in terms of weight gain, specific growth rate, and hepatosomatic index. However, the RO groups' survival rate was significantly lower than FO groups. The feed conversion and protein efficiency ratios of RO groups were poorer than other groups. The proximate compositions of whole body and hepatopancreas were significantly affected by these dietary treatments. Tissue fatty acid composition mainly reflected dietary fatty acid compositions. Crabs fed FO diets exhibited significantly higher arachidonic, eicosapentaenoic, and docosahexaenoic acid contents in muscle and hepatopancreas compared with VO crabs. Linoleic, oleic, and linolenic acids in muscle and hepatopancreas were the highest in the SO, RO, and LO groups, respectively. The present study suggested that SO and LO could substitute for FO in fishmeal-based diets for swimming crabs, without affecting growth performance and survival.

KIF3A and KIF3B are two N-terminal motor proteins belonging to the kinesin-II superfamily that play essential roles in *** understand the roles played by KIF3 A/3B during spermatogenesis of large yellow croaker Larimichthys crocea,we studied the testis characteristics at different developmental stages of ***,and determined the spatiotemporal expression patterns of kif3a and kif3b during *** real-time PCR(qR T-PCR)showed that the overall trends of kif3 a/3 b m RNA abundance during testis development are *** stage Ⅱ to stage V,kif3a/3b m RNA abundances first increased and then fell after reaching a peak at stage ***,the m RNA abundances of both genes at stage V were higher than those at stages Ⅱ and Ⅲ.In addition,it is worth of noting that kif3 b m RNA abundance was higher than that of kif3a at all *** in situ hybridization results revealed that kif3a/3b m RNA abundance dynamics were consistent with the migration of mitochondria,the deformation of nucleus,and the formation of *** m RNA hybridization signals of both genes first appeared either around the nuclear periphery or on the side of the nuclei,then appeared at one side of nuclei,and finally were mainly on the tail during *** findings contributed to better understanding the molecular mechanisms of spermiogenesis in fish;and suggested that KIF3A and KIF3B may participate in the intracellular transport of mitochondria,nuclear deformation,and the formation of tail during the spermiogenesis in ***.
