Purpose: Based on a quasi-experimental design, this study sought to investigate the effects of two different cognitive styles, field independence and field dependence, on students' learning behaviors of online database search ***/methodology/approach: An experiment was carried out among senior students in a Chinese ***: The field independent(FI) subjects performed better in terms of their search strategy scores. When comparing how many people in each cognitive style group learned the targeted search strategies, more field dependent(FD) subjects were successors, whereas the FI subjects were more inclined to learn from their past experience. When analyzing the reasons for the subjects' selection of search strategies, we found that the FI subjects demonstrated more rational thinking behaviors than the FD *** limitations: Only 28 students participated in the study, which was a relatively small sample size. A larger sample will give more information and therefore more precise *** implications: This research can provide some suggestions to the information system designers on how the system interface can be better designed to suit the cognitive styles of different ***/value: So far, few studies have been published about the effects of users' cognitive styles on their learning behaviors of online database search strategies.

CRISPR/Cas9 is a versatile genome-editing tool which is widely used for modifying the genome of both prokaryotic and eukaryotic organisms for basic research and applications. An increasing number of reports have demonstrated that CRISPR/Cas9-mediated genome editing is a powerful technology for gene therapy. Here, we review the recent advances in CRISPR/Cas9-mediated gene therapy in animal models via different strategies and discuss the challenges as well as future prospects.

Genome editing through adeno-associated viral(AAV) vectors is a promising gene therapy strategy for various diseases,especially genetic disorders. However, homologous recombination(HR) efficiency is extremely low in adult animal models. We assumed that increasing AAV transduction efficiency could increase genome editing activity, especially HR efficiency, for in vivo gene therapy. Firstly, a mouse phenylketonuria(PKU) model carrying a pathogenic R408W mutation in phenylalanine hydroxylase(Pah) was generated. Through co-delivery of the general AAV receptor(AAVR), we found that AAVR could dramatically increase AAV transduction efficiency in vitro and in vivo. Furthermore, co-delivery of SaCas9/sgRNA/donor templates with AAVR via AAV8 vectors increased indel rate over 2-fold and HR rate over 15-fold for the correction of the single mutation in Pah;mice. Moreover, AAVR co-injection successfully increased the site-specific insertion rate of a 1.4 kb Pah cDNA by 11-fold, bringing the HR rate up to 7.3% without detectable global off-target effects. Insertion of Pah cDNA significantly decreased the Phe level and ameliorated PKU symptoms. This study demonstrates a novel strategy to dramatically increase AAV transduction which substantially enhanced in vivo genome editing efficiency in adult animal models, showing clinical potential for both conventional and genome editing-based gene therapy.

The steady progress in genome editing, especially genome editing based on the use of clustered regularly interspaced short palindromic repeats(CRISPR) and programmable nucleases to make precise modifications to genetic material, has provided enormous opportunities to advance biomedical research and promote human health. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges. We also summarize the tools of genome editing and the insights derived from their applications, hoping to accelerate new discoveries and therapies in biomedicine.

This study aims at proposing a reasonable roughness parameter that can reflect the peak shear strength(PSS)of rock ***,the contribution of the asperities with different apparent dip angles to shear strength is *** the shear strength of the entire joint asperities is *** results showed that the PSS of the entire joint asperities is proportional to a key parameter hs,which is related to the geometric character of the joint surface and the joint material *** parameter hsis taken as the new roughness parameter,and it is reasonable to associate the PSS with the geometric characteristics of the joint *** on the new roughness parameter and shear test results of 20 sets of joint specimens,a new PSS model for rock joints is *** new model is validated with the artificial joints in this paper and real rock joints in published *** showed that it is suitable for different types of rock joints except for gneiss *** new model has the form of the Mohr-Coulomb model,which can directly reflect the relationship between the 3 D roughness parameters and the peak dilation angle.

At present,the development trend of dressing materials is being multifunctional for convenient and long-term nursing care process of some complicated ***,basing on the theory of wound moist healing,an injectable and self-healing hydrogel comprising of collagen(COL),chitosan(CS)and oxidation modified Konjac glucomannan(OKGM),which acts as a macromolecular cross-linker to construct dynamic Schiff-base bonds was smartly *** strategy of introducing the silver nanoparticles(Ag NPs)into the COL-CS-OKGM hydrogel matrix achieved a markedly enhanced antibacterial activity derived from the synergistical effect between the Ag^(+)and the mild photothermal efficacy of Ag NPs,which also improved the local capillary blood circulation of the wound area to further facilitate wound healing *** excellent syringeability and self-healing behaviors endowed the COL-CS-OKGM-Ag hydrogel with self-adapting ability for the wounds with irregular and large area needing frequent applying and changing without secondary *** vitro and in vivo evaluations verified that so-designed COL-CS-OKGM-Ag hydrogel also with hemostatic performance is a promising multifunctional dressing for the treatments of infected wound with not only good biocompatibility and convenient use,but also with desired regenerative healing prognoses benefited from hydrogel moist environment and physiotherapy.
