Periodontitis is closely related to many systemic diseases linked by different periodontal pathogens.To unravel the relationship between periodontitis and systemic diseases,it is very important to correctly discriminate major periodontal pathogens.To realize convenient,effcient,and high-accuracy bacterial species classification,the authors use Raman spectroscopy combined with machine learning algorithms to distinguish three major periodontal pathogens Porphyromonas gingivalis(Pg),Fusobacterium nucleatum(Fn),and Aggregatibacter actinomycetemcomitans(Aa).The result shows that this novel method can successfully discriminate the three abovementioned periodontal pathogens.Moreover,the classification accuracies for the three categories of the original data were 94.7%at the sample level and 93.9%at the spectrum level by the machine learning algorithm extra trees.This study provides a fast,simple,and accurate method which is very beneficial to differentiate periodontal pathogens.

Development and Application of Artificial Intelligence Recognition Systems in Rectal Cancer Staging Whether for surgical treatment or for neoadjuvant chemoradiotherapy,imaging evaluation has become an important basis to perform the treatment plans.[1]The reading of imaging results requires a large number of experienced radiologists to complete,but shortages and uneven distributions of personnel cause delays and biases in imaging results.Therefore,independent research and development of automatic recognition systems of rectal cancer staging based on artificial intelligence(AI)platforms aim to partially replace practitioners’work and achieve rapid and accurate identification of rectal cancer staging.

Ethnopharmacology relevance:Panax ginseng C.A.Mayer.is a traditional chinese herbal medicine and has beneficial effects for those with physical and mental exhaustion,including fatigue and anxiety.Ginsenosides are recognized as its major active ingredient which reportedly play an important role in the anxiolytic effects of ginseng.Dammarane sapogenins(DS),the deglycosylated product of ginsenosides,possesses high pharmacological activity and has higher bioavailability than ginsenosides.However,the anxiolytic-like effects of DS in mice subjected to chronic social defeat stress(CSDS)and its underlying mechanisms are unknown.Aim of the study:This study aimed to investigate the anxiolytic-like effects of DS in CSDS model mice and potential mechanisms related to corticosterone(CORT),monoamine neurotransmitters and brain-derived neurotrophic factor(BDNF) signalling regulation.Materials and methods:Fifty C57 BL/6 mice were divided into a non-CSDS control group(con),a chronic social defeat model group(CSDS),CSDS+DS groups(40 or 80 mg/kg) and a CSDS+ fluoxetine group(10 mg/kg).Except for the control group,the groups were exposed to CSDS for 21 days.The anxiolytic effect of DS was evaluated by the social interaction test(SIT),open field test(OFT),elevated plus maze(EPM) and novelty-suppressed feeding(NSF) test.Furthermore,serum CORT levels and levels of monoamine neurotransmitters in the hippocampus and frontal cortex were tested using an enzyme-linked immunosorbent assay(ELISA) kit.The protein expression levels of brain-derived neurotrophic factor(BDNF),phosphorylated CREB and ERK1/2 in the hippocampus and frontal cortex were detected by western blot.Results:Oral administration of DS improved anxiolytic-like effects in mice,as indicated by increased time spent in the centre of the field in the OFT,latency to eat in the NSF test and the percentage of open arm entries and time spent in the open arms in the EPM.DS treatment significantly reduced serum CORT levels and increased the neurotransmitter concentration(5-HT and NE) in

It is known that social stress could alter oxytocin(OT)and arginine-vasopressin(AVP)expression in specific regions of brains which regulate the aggressive behavior of small rodents,but the effects of density-induced social stress are still unknown.Brandt’s voles(Lasiopodomys brandtii)are small herbivores in the grassland of China,but the underlying neurological mechanism of population regulation is still unknown.We tested the effects of housing density of Brandt’s voles on OT/AVP system with physical contact(allowing aggression)and without physical contact(not allowing aggression)under laboratory conditions.Then,we tested the effects of paired-aggression(no density effect)of Brandt’s voles on OT/AVP system under laboratory conditions.We hypothesized that high density would increase aggression among animals which would then increase AVP but reduce OT in brains of animals.Our results showed that high housing density induced more aggressive behavior.We found high-densityinduced social stress(with or without physical contact)and direct aggression significantly increased expression of mRNA and protein of AVP and its receptor,but decreased expression of mRNA and protein of OT and its receptor in specific brain regions of voles.The results suggest that density-dependent change of OT/AVP systems may play a significant role in the population regulation of small rodents by altering density-dependent aggressive behavior.

The evolutionarily conserved Wnt signaling pathway plays a central role in develop-ment and adult tissue homeostasis across species.Wnt proteins are secreted,lipid-modified signaling molecules that activate the canonical(β-catenin dependent)and non-canonical(β-catenin independent)Wnt signaling pathways.Cellular behaviors such as proliferation,differ-entiation,maturation,and proper body-axis specification are carried out by the canonical pathway,which is the best characterized of the known Wnt signaling paths.Wnt signaling has emerged as an important factor in stem cell biology and is known to affect the self-renewal of stem cells in various tissues.This includes but is not limited to embryonic,hematopoietic,mesenchymal,gut,neural,and epidermal stem cells.Wnt signaling has also been implicated in tumor cells that exhibit stem cell-like properties.Wnt signaling is crucial for bone formation and presents a potential target for the development of therapeutics for bone disorders.Not surprisingly,aberrant Wnt signaling is also associated with a wide variety of diseases,including cancer.Mutations of Wnt pathway members in cancer can lead to unchecked cell proliferation,epithelial-mesenchymal transition,and metastasis.Altogether,advances in the understand-ing of dysregulated Wnt signaling in disease have paved the way for the development of novel therapeutics that target components of the Wnt pathway.Beginning with a brief overview of the mechanisms of canonical and non-canonical Wnt,this review aims to summarize the cur-rent knowledge of Wnt signaling in stem cells,aberrations to the Wnt pathway associated with diseases,and novel therapeutics targeting the Wnt pathway in preclinical and clinical studies.

The study of tumor nanovaccines(NVs)has gained interest because they specifically recognize and eliminate tumor cells.However,the poor recognition and internalization by dendritic cells(DCs)and insufficient immunogenicity restricted the vaccine efficacy.Herein,we extracted two molecular-weight Astragalus polysaccharides(APS,12.19 k D;APSHMw,135.67 k D)from Radix Astragali and made them self-assemble with OVA257–264directly forming OVA/APS integrated nanocomplexes through the microfluidic method.The nanocomplexes were wrapped with a sheddable calcium phosphate layer to improve stability.APS in the formed nanocomplexes served as drug carriers and immune adjuvants for potent tumor immunotherapy.The optimal APS-NVs were approximately 160 nm with uniform size distribution and could remain stable in physiological saline solution.The FITC-OVA in APS-NVs could be effectively taken up by DCs,and APS-NVs could stimulate the maturation of DCs,improving the antigen cross-presentation efficiency in vitro.The possible mechanism was that APS can induce DC activation via multiple receptors such as dectin-1 and Toll-like receptors 2 and 4.Enhanced accumulation of APS-NVs both in draining and distal lymph nodes were observed following s.***.injection.Smaller APS-NVs could easily access the lymph nodes.Furthermore,APS-NVs could markedly promote antigen delivery efficiency to DCs and activate cytotoxic T cells.In addition,APS-NVs achieve a better antitumor effect in established B16-OVA melanoma tumors compared with the OVA+Alum treatment group.The antitumor mechanism correlated with the increase in cytotoxic T cells in the tumor region.Subsequently,the poor tumor inhibitory effect of APS-NVs on the nude mouse model of melanoma also confirmed the participation of antitumor adaptive immune response induced by NVs.Therefore,this study developed a promising APS-based tumor NV that is an efficient tumor immunotherapy without systemic side effects.

To the Editor:As the novel coronavirus disease 2019(COVID-19)pandemic rapidly spreads around the world,it has resulted in unprecedented economic loss,social turmoil,and detrimental effects on public mental health.In particular,the implementation of disease control measures such as nationwide lockdowns,social distancing,school closures,and home quarantine has given rise to a wide range of social anxiety(SA)around the world,from nonclinical manifestations to clinical social anxiety disorder(SAD).[1]Given the potentially severe impairments from SA and the limited mental health care resources,it is vital to identify whether there are vulnerability/risk neuropsychological factors contributing to the progression of SA during the pandemic.

OBJECTIVE Ginsenoside Rb1(Rb1),an important bioactive ingredient of Panax ginseng,has potent neuroprotective effects.The objective of the study is to elucidate the impact of Rb1 treatment on chronic social defeat stress(CSDS)-induced depressive-like behaviors and its related mechanism.METHODS AND RE⁃SULTS The daily oral administration of Rb1(35 and 70 mg·kg-1)and imipramine(15 mg·kg-1)for 28 d significantly reversed the social avoidance behavior,anhedonia,and behavioral despair via CSDS exposure,as demonstrated by the consid⁃erable elevation in the time in the zone in social interaction test and consumption of sucrose solu⁃tion in sucrose preference test and decrease of immobility time in forced swim test.Moreover,Rb1 obviously restored the CSDS-induced decrease of BDNF-signaling pathway and hippo⁃campal neurogenesis.Rb1 significantly increased the hippocampal levels of ERK,AKT,and CREB phosphorylation and increased the number of DCX+cells in DG.Importantly,the antidepres⁃sant effects of Rb1 were completely blocked in mice by using K252a(the nonselective tyrosine kinase B inhibitor).CONCLUSION Rb1 exerts promising antidepressant-like effects in mice with CSDS-induced depression,and its effects was facilitated by enhancing the BDNF signaling cas⁃cade and up-regulation of hippocampal neuro⁃genesis.
