An improved association-mining research for exploring chinese herbal property theory: based on data of the Shennong's Classic of materia medica

Knowledge Discovery in Databases is gaining attention and raising new hopes for traditional chinese medicine （TCM） researchers. It is a useful tool in understanding and deciphering TCM theories. Aiming for a better understanding of chinese herbal property theory （CHPT）, this paper performed an improved association rule learning to analyze semistructured text in the book entitled Shennong＇s Classic of materia medica. The text was firstly annotated and transformed to well-structured multidimensional data. Subsequently, an Apriori algorithm was employed for producing association rules after the sensitivity analysis of parameters. From the confirmed 120 resulting rules that described the intrinsic relationships between herbal property （qi, flavor and their combinations） and herbal efficacy, two novel fundamental principles underlying CHPT were acquired and further elucidated：（1） the many-to-one mapping of herbal efficacy to herbal property; （2） the nonrandom overlap between the related efficacy of qi and flavor. This work provided an innovative knowledge about CHPT, which would be helpful for its modern research.

The current quality control（QC） pattern for chinese materia medica（CMM） lacks suitable methods and indicators to evaluate their safety and efficacy effectively, which impedes the smooth development of CMM. In this review, main problems of the current QC pattern for CMM, principally focused on the content determination of constituents,were summarized and the inspiration from the QC of biological products was introduced. With the aim at introducing a suitable tool to the QC of CMM, biopotency assay and its feasibility in the QC pattern for CMM were analyzed and confirmed by relevant researches with years of practice. From the applications of biopotency assays in the QC of CMM in the last 10 years, we propose that biopotency assays should be an integral part of the QC pattern for CMM, for these assays can make the QC indicators related to the clinical safety and efficacy, supplementing the existed QC system of CMM.

It is one of the key points for modernization and internationalization of traditional chinese medicines to construct the Good Agricultural Practice (GAP) base of chinese materia medica (CMM). GAP helps to minimize contamination and improve the quality of CMM during the plantation and the production of chinese crude drugs. In this article, the status and development of CMM production bases of GAP in Guangdong Province, China, are presented. The suggestions upon the problems during the development of GAP for chinese crude drugs are also provided.

The property theory of chinese materia medica is one of the foundations of traditional chinese medicine.The property of chinese materia medica(PCMM)is a multi-dimensional expression of the effect of chinese materia medica(CMM),and it is related to the clinical prescription that fully reflects the clinical effect evaluation of CMM in a holistic,systematic,and scientific way.This paper discusses the source,development,and application of the PCMM by considering not only the five dimensions that constitute the PCMM but also the recognition of the human body and disease as given in traditional chinese medicine.This paper aims to provide theoretical guidance for the rational use and development of CMM.

Jiawei Xiaoyao San(JWXYS)has shown excellent clinical efficacy in anxiety disorder,but has not yet attracted widespread attention.The animal experiments,clinical trials and mechanism studies of JWXYS were reviewed in this article,which may provide a reference for developing new anxiolytic drugs based on this prescription.The literature was searched in PubMed and CNKI and the documents written in English or with English abstracts were selected.JWXYS could reduce the anxiety symptoms of patients alone and reduce the adverse reactions when it is used in combination with other drugs in the clinic.In preclinical studies,JWXYS also showed therapeutic effects in reducing anxiety-like behavior.The mechanisms may include improving the hypothalamic–pituitaryadrenal(HPA)axis and hormone disorders,increasing neurotransmitter content,neurogenesis,and regulating the synthesis of related enzymes.This article shows that JWXYS could effectively treat anxiety disorders by regulating the central nervous system.In the future,with the participation of more researchers,it is expected to develop innovative drugs for the treatment of anxiety disorders based on JWXYS.

Quantitative determination of multi-class bioactive constituents for quality control of Yiqi Jiangzhi Granules

Objective:To establish a reliable and sensitive method for evaluating quality of Yiqi Jiangzhi Granules(YQJZG).Methods:Ultra performance liquid chromatography electrospray ionization tandem mass spectrometry(UPLC-ESI-MS/MS)was employed for simultaneous determination of eight marker components.Separation was performed on an AQUITY UPLC;HSS T3 column,the mobile phase consisted of acetonitrile as the organic phase and 0.1%(volume percentage)formic acid as the aqueous.Eight marker components,ginsenoside Rg1(GRg1),ginsenoside Re(GRe),ginsenoside Rb1(Gb1),typhaneoside(TEO),isorhamnetin-3-O-neohespeidoside(IN),hesperidin(HPD),aurantio-obtusin-6-O-β-D-glucoside(AG)and curcumin(CCM),were detected by multiple reaction monitoring(MRM)mode.The chinese Pharmacopoeia(2020 edition)was regarded as the guidance document for this method validation.Results:The method showed good linearity(R^(2)≥0.9990).The relative standard deviation(RSD)values for the instrument precision,intermediate precision and repeatability were less than 2.91%,2.88%,and 3.54%,respectively.The average recovery varied from 91.08%to 103.89%,with RSD below 3.81%.Sample solutions were found to be stable within 24 h at 4℃(RSDchinese medicine analogous formulae.

Phenylalanine ammonia-lyase(PAL),which catalyzes the conversion from L-phenylalanine to trans-cinnamic acid,is a well-known key enzyme and a connecting step between primary and secondary metabolisms in the phenylpropanoid biosynthetic pathway of plants and microbes.Schisandra chinensis,a woody vine plant belonging to the family of Magnoliaceae,is a rich source of dibenzocyclooctadiene lignans exhibiting potent activity.However,the functional role of PAL in the biosynthesis of lignan is relatively limited,compared with those in lignin and flavonoids biosynthesis.Therefore,it is essential to clone and characterize the PAL genes from this valuable medicinal plant.In this study,molecular cloning and characterization of three PAL genes(ScPAL1−3)from S.chinensis was carried out.ScPALs were cloned using RACE PCR.The sequence analysis of the three ScPALs was carried out to give basic characteristics followed by docking analysis.In order to determine their catalytic activity,recombinant protein was obtained by heterologous expression in pCold-TF vector in Escherichia coli(BL21-DE3),followed by Ni-affinity purification.The catalytic product of the purified recombinant proteins was verified using RP-HPLC through comparing with standard compounds.The optimal temperature,pH value and effects of different metal ions were determined.Vmax,Kcat and Km values were determined under the optimal conditions.The expression of three ScPALs in different tissues was also determined.Our work provided essential information for the function of ScPALs.

目的:对丹参50%乙醇提取物的体内外物质基础进行系统全面的分析。创新点:首次采用高效液相色谱-质谱联用(HPLC-LTQOrbitrap)方法对丹参50%乙醇提取物的体内外化学和代谢成分进行全面分析。方法:建立HPLC-LTQ-Orbitrap方法,对丹参50%乙醇提取物的化学成分以及给药后大鼠血浆、尿液、粪便和胆汁生物样品中的化学和代谢成分进行分析。结论:在正负离子模式下,在丹参提取物中鉴定出共69个化合物,包括丹酚酸类化合物23个,丹参酮类化合物33个,以及未知化合物13个;在大鼠灌胃给予丹参提取物后的生物样本中鉴定出共118个化合物,包括血浆中35个,尿液中63个,粪便中62个,以及胆汁中18个。

Medicinal plants are renowned for their abundant production of secondary metabolites,which exhibit notable pharmacological activities and great potential for drug development.The biosynthesis of secondary metabolites is highly intricate and influenced by various intrinsic and extrinsic factors,resulting in substantial species diversity and content variation.Consequently,precise regulation of secondary metabolite synthesis is of utmost importance.In recent years,genome sequencing has emerged as a valuable tool for investigating the synthesis and regulation of secondary metabolites in medicinal plants,facilitated by the widespread use of high-throughput sequencing technologies.This review highlights the latest advancements in genome sequencing within this field and presents several strategies for studying secondary metabolites.Specifically,the article elucidates how genome sequencing can unravel the pathways for secondary metabolite synthesis in medicinal plants,offering insights into the functions and regulatory mechanisms of participating enzymes.Comparative analyses of plant genomes allow identification of shared pathways of metabolite synthesis among species,thereby providing novel avenues for obtaining cost-effective biosynthetic intermediates.By examining individual genomic variations,genes or gene clusters associated with the synthesis of specific compounds can be discovered,indicating potential targets and directions for drug development and the exploration of alternative compound sources.Moreover,the advent of gene-editing technology has enabled the precise modifications of medicinal plant genomes.Optimization of specific secondary metabolite synthesis pathways becomes thus feasible,enabling the precise editing of target genes to regulate secondary metabolite production within cells.These findings serve as valuable references and lessons for future drug development endeavors,conservation of rare resources,and the exploration of new resources.
