Tumor inflammation-specific self-amplifying fluorescent molecular probes induce an anti-tumor immune response

Enhancing the concentration of exogenous molecular drugs within the tumor microenvironment through enzyme-catalyzed polymerization presents a novel strategy for cancer therapy. Nonetheless, the optimization of the catalytic efficiency is often impeded by the inefficient expression of enzymes. Herein, we reported a self-amplifying fluorescent molecular probe, Bis-HTPICG, for photodynamic therapy(PDT) and subsequent PDT-induced immunoreaction. The Bis-HTP-ICG probe possesses a noticeable enzyme-catalyzed polymerization facilitated by myeloperoxidase(MPO), a crucial enzyme secreted by neutrophils at inflammation sites. Upon exposure to laser irradiation, Bis-HTP-ICG showed a high PDT efficacy, inducing an acute inflammatory response that stimulates further recruitment of neutrophils and then elevated MPO secretion. The heightened level of MPO enhances the accumulation of the Bis-HTP-ICG via self-polymerization or binding with intratumoral proteins following MPO enzyme catalysis, instigating a self-amplifying chain reaction cycle involving Bis-HTP-ICG, neutrophils and ***, PDT efficiently incites immunogenic cell death(ICD) in tumor cells, initiating an anti-tumor immune response including dendritic cells(DCs) maturation, T cell proliferation and reprogramming of tumor-associated neutrophils(TANs). This work portrays a promising strategy for self-amplification of fluorescent molecular probes through adjustable enzyme levels,potentially offering a unique avenue to enhance the tumor accumulation of molecular drugs for improved tumor therapy.

Local-contrastive-learning machine with both generalization and adversarial robustness: A statistical physics analysis

Distinct from human cognitive processing, deep neural networks trained by backpropagation can be easily fooled by adversarial examples. To design a semantically meaningful representation learning, we discard backpropagation, and instead, propose a local contrastive learning, where the representations for the inputs bearing the same label shrink(akin to boson) in hidden layers,while those of different labels repel(akin to fermion). This layer-wise learning is local in nature, being biologically plausible.A statistical mechanics analysis shows that the target fermion-pair-distance is a key parameter. Moreover, the application of this local contrastive learning to MNIST benchmark dataset demonstrates that the adversarial vulnerability of standard perceptron can be greatly mitigated by tuning the target distance, i.e., controlling the geometric separation of prototype manifolds.

Establishing a new testing protocol of the electrochemical stability window for electric double-layer capacitors

The electrochemical stability window(ESW) is crucial for determining the energy density of electric double-layer capacitors(EDLCs). However, current ESW testing protocols of EDLCs are strongly affected by artificial factors, thereby leading to unreliable result. To solve this problem, we first identify the key shortcomings of the traditional protocols by analyzing their test principles. Subsequently, taking an aqueous carbon-based EDLC as an example, we design a side reaction descriptor to represent the response difference in the charging capacitance to side reactions between the cyclic voltammetry and galvanostatic chargedischarge methods. With the aid of this descriptor, we propose a new ESW testing protocol, which is not only able to completely avoid the adverse influence of subjective factors but also fully consider the operating condition limitations. The protocol can be extended from the single electrode to the full device. This work proposes a more rigorous definition of ESW and provides a reliable protocol for evaluating the ESW of EDLCs.

Statistical analysis on the validity of the cold plasma approximation for chorus waves based on Van Allen Probe observations and their effects on radiation belt electrons

Theoretical analysis has demonstrated that the dispersion relation of chorus waves plays an essential role in the resonant interaction and energy transformation between the waves and magnetospheric *** quantitative analyses often simplified the chorus dispersion relation by using the cold plasma ***,the applicability of the cold plasma assumption is doubtful,especially during geomagnetic *** here present a systematic statistical analysis on the validity of the cold plasma dispersion relation of chorus waves based on observations from the Van Allen Probes over the period from 2012 to *** statistical results show that the observed magnetic field intensities deviate substantially from those calculated from the cold plasma dispersion relation and that they become more pronounced with an increase in geomagnetic activity or a decrease in background plasma *** region with large deviations is mainly concentrated in the nightside and expands in both the radial and azimuthal directions as the geomagnetic activity increases or the background plasma density *** addition,the bounce-averaged electron scattering rates are computed by using the observed and cold plasma dispersion relation of chorus *** with usage of the cold plasma dispersion relation,usage of the observed dispersion relation considerably lowers the minimum resonant energy of electrons and lowers the scattering rates of electrons above tens of kiloelectronvolts but enhances those ***,these differences are more pronounced with the enhancement of geomagnetic activity or the decrease in background plasma density.

Researchers have recently achieved significant advances in deep learning techniques, which in turn has substantially advanced other research disciplines, such as natural language processing, image processing, speech recognition, and software engineering. Various deep learning techniques have been successfully employed to facilitate software engineering tasks, including code generation, software refactoring, and fault localization. Many studies have also been presented in top conferences and journals, demonstrating the applications of deep learning techniques in resolving various software engineering tasks. However,although several surveys have provided overall pictures of the application of deep learning techniques in software engineering,they focus more on learning techniques, that is, what kind of deep learning techniques are employed and how deep models are trained or fine-tuned for software engineering tasks. We still lack surveys explaining the advances of subareas in software engineering driven by deep learning techniques, as well as challenges and opportunities in each subarea. To this end, in this study, we present the first task-oriented survey on deep learning-based software engineering. It covers twelve major software engineering subareas significantly impacted by deep learning techniques. Such subareas spread out through the whole lifecycle of software development and maintenance, including requirements engineering, software development, testing, maintenance, and developer collaboration. As we believe that deep learning may provide an opportunity to revolutionize the whole discipline of software engineering, providing one survey covering as many subareas as possible in software engineering can help future research push forward the frontier of deep learning-based software engineering more systematically. Early identification and treatment of stroke can greatly improve patient outcomes and quality of *** clinical tests such as the Cincinnati Pre-hospital Stroke Scale(CPSS)and the Face Arm Speech Test(FAST)are commonly used for stroke screening,accurate administration is dependent on specialized *** this study,we proposed a novel multimodal deep learning approach,based on the FAST,for assessing suspected stroke patients exhibiting symptoms such as limb weakness,facial paresis,and speech disorders in acute *** collected a dataset comprising videos and audio recordings of emergency room patients performing designated limb movements,facial expressions,and speech tests based on the *** compared the constructed deep learning model,which was designed to process multi-modal datasets,with six prior models that achieved good action classification performance,including the I3D,SlowFast,X3D,TPN,TimeSformer,and *** found that the findings of our deep learning model had a higher clinical value compared with the other ***,the multi-modal model outperformed its single-module variants,highlighting the benefit of utilizing multiple types of patient data,such as action videos and speech *** results indicate that a multi-modal deep learning model combined with the FAST could greatly improve the accuracy and sensitivity of early stroke identification of stroke,thus providing a practical and powerful tool for assessing stroke patients in an emergency clinical setting.

Homologous CoS2 interface with directed electron transfer and strong chloridion repulsion for efficient seawater oxidation

Seawater electrolysis is a highly promising method to produce high energy density hydrogen,especially in dry areas that have scarce freshwater ***,application of seawater electrolysis is confronted by two major problems associated with catalysts employed for the anodic oxygen evolution reaction （OER） including the competitive hypochlorite evolution reaction（HCER） and strong chloridion （Cl–） triggered *** studies aimed at overcoming these problems,a facile method has been developed to synthesize a homologous CoS2heterojunction as an OER *** to a difference in work functions at the interface and increased electron density,this material has directed electron transfer and strong chloridion repulsion properties,*** have shown that CoS2achieves low overpotentials of 289 and 322 mV at 10 mA cm-2current density in respective alkaline （1 M KOH） and alkaline simulated seawater ***,the outstanding stability of CoS2is reflected in a negligible decline in activity during promotion of the OER for 100 *** of density functional theory calculations reveal that the homologous CoS2heterojunction promotes directed electron transport for optimizing adsorption/desorption of reactive species and heightens the Cl-adsorption energy for strongly repulsing Cl-,phenomena that enhance OER performance in alkaline simulated seawater.

We study the characteristics of temperature fluctuation in two-dimensional turbulent Rayleigh–Benard convection in´a square cavity by direct numerical *** Rayleigh number range is 1×10^(8)≤Ra≤1×10^(13),and the Prandtl number is selected as Pr=0.7 and Pr=*** is found that the temperature fluctuation profiles with respect to Ra exhibit two different distribution *** the thermal boundary layer,the normalized fluctuationθrms/θrms,max is independent of Ra and a power law relation is identified,i.e.,θrms/θrms,max∼(z/δ)0.99±0.01,where z/δis a dimensionless distance to the boundary(δis the thickness of thermal boundary layer).Out of the boundary layer,when Ra≤5×10^(9),the profiles ofθrms/θrms,max descend,then ascend,and finally drop dramatically as z/δ*** for Ra≥1×10^(10),the profiles continuously decrease and finally overlap with each *** two different characteristics of temperature fluctuations are closely related to the formation of stable large-scale circulations and corner ***,there is a critical value of Ra indicating the transition,beyond which the fluctuation hθrmsiV has a power law dependence on Ra,given by hθrmsiV∼Ra−0.14±0.01.

A Mathematical Modeling Method Elucidating the Integrated Gripping Performance of Ant Mandibles and Bio-inspired Grippers

The ability to grip unhatched eggs is a skill exploited by the ants Harpegnathos venator,as they care their brood in tunneled nests,which is of extreme difficulty to keep the eggs intact while *** this paper we propose a mathematical modeling method to elucidate the mechanism of such a gripping behavior in the ant *** new method can be subdivided into following *** a preliminary,the concavity geometry and mandible kinematics are examined ***,coordinate transformation is used to predict the real-time spatial topology of the ***,we come up with a new method to quantify the workspace required to grip and the contact area between the concavity and ant *** model indicates that the biaxial rotation fashion with specialized concavities can reduce workspace by 40%and increase contact area by 53%on average compared with the uniaxial rotation pattern,which augments success rate of gentle *** methodology may have applications in evaluating mechanical performance in both natural and artificial grippers.
