Brillouin microscopy, which maps the elastic modulus from the frequency shift of scattered light, has evolved to a faster speed for the investigation of rapid biomechanical changes. Impulsive stimulated Brillouin scattering (ISBS)spectroscopy has the potential to speed up measurement through the resonant amplification interaction from pulsed excitation and time-domain continuous detection. However, significant progress has not been achieved due to the limitation in signal-to-noise ratio (SNR) and the corresponding need for excessive averaging to maintain high spectral precision. Moreover, the limited spatial resolution also hinders its application in mechanical imaging. Here, by scrutinizing the SNR model, we design a high-speed ISBS microscope through multi-parameter optimization including phase, reference power, and acquisition time. Leveraging this, with the further assistance of the Matrix Pencil method for data processing, three-dimensional mechanical images are mapped under multiple contrast mechanisms for a millimeter-scale polydimethylsiloxane pattern immersed in methanol, enabling the identification of these two transparent materials without any contact or labeling. Our experimental results demonstrate the capability to maintain high spectral precision and resolution at a sub-millisecond integration time for one pixel. With a two-order improvement in the speed and a tenfold improvement in the spatial resolution over the state-of-the-art systems, this method makes it possible for ISBS microscopes to sensitively investigate rapid mechanical changes in time and space.

Purpose:Observational studies have reported positive associations between glaucoma and stroke;however,controversial results ***,the nature of the relationship remains unknown since previous studies were not designed to test ***,we aimed to investigate the possible causal relationships between glaucoma and ***:Our two-sample Mendelian randomization(MR)encompassed multi-ethnic large-scale genome-wide association studies with more than 20000 cases and 260000 controls for glaucoma,and more than 80000 cases and 630000 controls for *** effect estimates for each SNP were combined using the inverse-variance weighted(IVW)*** avoid potential pleiotropic effects,we adjusted the main results by excluding genetic variants associated with metabolic *** weighted median and MR-Egger methods were also used for the sensitivity ***:Our MR analysis revealed that glaucoma and its subtypes,including primary open-angle glaucoma and primary angle-closure glaucoma,exhibited no causal role in relation to any stroke(AS),any ischemic stroke(AIS),large-artery atherosclerotic stroke(LAS),small-vessel stroke(SVS),or cardioembolic stroke(CES)across MR analyses(all P>0.05).The null associations remained robust even after adjusting for metabolic-related traits and were consistent in both the European and Asian ***,reverse MR analyses also did not indicate any significant causal effects of AS,AIS,LAS,or CES on glaucoma ***:Evidence from our series of causal inference approaches using large-scale population-based MR an-alyses did not support causal effects between glaucoma and *** findings suggest that the relationship of glaucoma management and stroke risk prevention should be carefully evaluated in future *** turn,stroke diagnosis should not be simply applied to glaucoma risk prediction.

This study aimed to develop a pan-genotypic and multifunctional small interfering RNA(siRNA)against hepatitis B virus(HBV)with an efficient delivery system for treating chronic hepatitis B(CHB),and explore combined RNA interference(RNAi)and immune modulatory modalities for better viral *** synthetic siRNAs targeting consensus motifs distributed across the whole HBV genome were designed and *** lipid nanoparticle(LNP)formulation was optimized by adopting HO-PEG_(2000)-DMG lipid and modifying the molar ratio of traditional polyethylene glycol(PEG)lipid in LNP *** efficacy and safety of this formulation in delivering siHBV(tLNP/siHBV)along with the mouse IL-2(mIL-2)mRNA(tLNP/siHBVIL2)were evaluated in the rAAVHBV1.3 mouse model.A siRNA combination(terms“siHBV”)with a genotypic coverage of 98.55%was selected,chemically modified,and encapsulated within an optimized LNP(tLNP)of high efficacy and security to fabricate a therapeutic formulation for *** results revealed that tLNP/siHBV significantly reduced the expression of viral antigens and DNA(up to 3log_(10)reduction;vs PBS)in dose-and time-dependent manners at single-dose or multi-dose frequencies,with satisfactory safety *** studies showed that tLNP/siHBVIL2 enables additive antigenic and immune control of the virus,via introducing potent HBsAg clearance through RNAi and triggering strong HBV-specific CD4^(+)and CD8^(+)T cell responses by expressed mIL-2 *** adopting tLNP as nucleic acid nanocarriers,the co-delivery of siHBV and mIL-2 mRNA enables synergistic antigenic and immune control of HBV,thus offering a promising translational therapeutic strategy for treating CHB.

Comparison of Ex‑PRESS implantation versus trabeculectomy combined with phacoemulsification in primary open‑angle glaucoma:a retrospective in vivo confocal microscopy study

Background:To compare the efficacy of Ex-PRESS implantation versus trabeculectomy combined with ***:A retrospective 12-month study on patients with coincident primary open-angle glaucoma(POAG)and *** patients underwent combined phacoemulsification and Ex-PRESS implant(Phaco-ExPRESS,n=35)or phacotrabeculectomy(Phaco-Trab,n=35).The morphological structures of the filtering bleb were examined by slitlamp,anterior segment optical coherence tomography(AS-OCT)and in vivo confocal microscopy(IVCM).Complete success was defined as postoperative intraocular pressure(IOP)<18 mmHg without the use of anti-glaucoma *** success was defined as postoperative IOP<18 mmHg with or without anti-glaucoma *** data were collected preoperatively and postoperatively at 2 weeks,1 month,3 months,6 months,and 12 ***:No significant difference in the variables such as age,IOP and perimetry was found between the groups of Phaco-ExPRESS and *** the one-year postoperative visit for filtering blebs,Phaco-ExPRESS increased the mean area of epithelial microcysts significantly from 0.10±0.05 to 0.20±0.09μm^(2)perμm^(2),while Phaco-Trab decreased the mean area significantly from 0.08±0.04 to 0.04±0.06μm^(2)perμm^(2).Notably,the hyperreflective dots detected by IVCM decreased by 84.9%in Phaco-ExPRESS but increased by 36.3%in *** hyperreflective dots were further identified as neutrophil-and monocyte-like *** number of these cells were negatively correlated with the microcysts area(r=−0.7,P<0.01)but positively associated with the grade of connective tissue(r=0.5,P<0.01).By creating different microstructural changes in the filtering blebs,Phaco-ExPRESS produced a higher complete success rate(84.9%vs.41.2%,P<0.01)and significant decrease in the number of anti-glaucoma medications(P<0.01)when compared with those in ***,the qualified success showed no significant difference between the two groups(
