Simultaneous analysis of the intestinal parasites and diet through eDnA metabarcoding

Agricultural expansion and intensiﬁcation are having a huge impact on plant and arthropod diversity and abun-dance,affecting food availability for farmland birds.Difﬁcult food access,in turn,can lead to immunosuppression and a higher incidence of parasites.In the studies designed to examine changes in the diet of birds and their par-asites,metabarcoding is proving particularly useful.This technique requires mini-barcodes capable of amplifying the DnA of target organisms from fecal environmental DnA.To help to understand the impact of agricultural expansion on biodiversity,this study sought to design and identify mini-barcodes that might simultaneously as-sess diet and intestinal parasites from the feces of farmland birds.The capacity to identify diet and parasites of 2 existing and 3 newly developed mini-barcodes was tested“in silico”in relation to the behavior of a reference eukaryotic barcode.Among the newly designed mini-barcodes,MiniB18S_81 showed the higher taxonomic cover-age of eukaryotic taxa and a greater ampliﬁcation and identiﬁcation capacity for diet and parasite taxa.Moreover,when it was tested on fecal samples from 5 different steppe bird species,MiniB18S_81 showed high taxonomic resolution of the most relevant diet and parasite phyla,Arthropoda,nematoda,Platyhelminthes,and Apicomplexa at the order level.Thus,the mini-barcode developed emerges as an excellent tool to simultaneously provide detailed information regarding the diet and parasites of birds,essential for conservation and management.

Do non-target species visit feeders and water troughs targeting small game?A study from farmland Spain using camera-trapping

Provision of food and water is a widespread tool implemented around the world for the benefit of game and other wildlife,but factors affecting the use of food and water by non-target species are poorly known.We evaluated visits to feeders and water troughs by non-game species using camera-traps in two separate areas of Spain.Feeders and water troughs were either“protected”(when surrounded by more than 50%of shrubs/forest)or“open”(in the opposite case).A total of 18948 photos from 5344 camera-trapping days depicted animals,and 75 species were identified.Feeders and water troughs were visited by target species(partridges and lagomorphs,55.3%of visits)and non-target species(44.7%of visits).Among the latter,corvids were themost common(46.1%of visits),followed by rodents(26.8%),other birds(23.6%,mainly passerines),columbids(1.9%),and other species at minor percentages.The highest proportion of visiting days to feeders and water troughs was from corvids(0.173)followed by otherbirds(0.109)and rodents(0.083);the lowest proportion was recorded for columbids(0.016).Use intensity and visit frequency of water troughs tripled that recorded in feeders,and visits to open feeders/troughs were approximately twice those to protected ones.In summary:feeders and water troughs targeting small game species are also used regularly by non-target ones;they should be set close to cover to optimize their use by non-target species that are not competitors of target species(though corvids may visit them);water availability should be prioritized where drought periods are expected.

Avian malaria and related haemosporidian parasites can negatively impact fitness in many songbirds.Research on the malaria infection and its physiological costs on their avian hosts is heavily skewed toward native passerines,with exotic species underrepresented.However,introduced species may carry on and spread new pathogens to native species,and play a role on parasite transmission cycle in invaded bird communities as pathogen reservoir.Here,we molecularly assess the prevalence and diversity of haemosporidian parasites in three introduced wetland passerines(the Red Avadavat Amandava amandava,the Yellow-crowned Bishop Euplectes afer,and the Common Waxbill Estrilda astrild)captured during the same season in southwestern Spain.We also explored the relation between parasite infection,body condition,haematocrit,and uropygial gland volume.We detected an overall parasite prevalence of 3.55%,where Common Waxbills showed higher prevalence(6.94%)than Red Avadavats(1.51%).none Yellow-crowned Bishops were infected with haemosporidians.Almost 60%of infections were caused by Leucocytozoon,and about 40%by Plasmodium.We identified four unique lineages of Plasmodium and three of Leucocytozoon.Moreover,91%of the identified host-parasite interactions represented new host records for these haemosporidian parasites.Parasite infection was not related to body condition,haematocrit,and uropygial gland volume of the wetland passerines.Haematocrit values varied seasonally among bird species.Additionally,haematocrit was positively related to body condition in the Yellow-crowned Bishops,but not in the other species.Red Avadavats had higher haematocrit levels than Yellow-crowned Bishops,whereas Common Waxbills showed the lower haematocrit values.The uropygial gland volume was positively correlated with body condition in all bird species.Common Waxbills showed higher uropygial gland volumes related to their body size than birds from other two species.These outcomes highlight the importance of exotic invasive species in the transmission dynam

Brucella suis is responsible for swine brucellosis worldwide. Of the five different B. suis biovars （by.）, bv. 2 appears restricted to Europe where it is frequently isolated from wild boar and hares, can infect pigs and can cause human brucellosis. In this study, the differ- ential gene expression profile was characterized in spleens of Eurasian wild boar naturally infected with B. suis bv. 2. Of the 20,201 genes analyzed in the microarray, 633 and 1,373 were significantly （fold change 〉 1.8; P 〈 0.01） upregulated and downregulated, respectively, in infected wild boar. The analysis was focused on genes that were over represented after conditional test for biological process gene ontology. Upregulated genes suggested that B. suis bv. 2 infection induced cell maturation, migration and/or proliferation in infected animals. The genes downregulated in infected wild boar impaired the activity of several important cellular metabolic pathways such as metabolism, cytoskeleton organization and biogenesis, immune response and lysosomal function and vesicle-mediated transport. In addition, the response to stress, sperm fertility, muscle development and apoptosis seemed to be also impaired in infected animals. These results suggested that B. suis bv. 2 may use strategies similar to other smooth brucellae to facilitate intracellular multiplication and the development of chronic infections. To our knowledge, this is the first report of the analysis of gene expression profile in hosts infected with B. suis bv. 2, which is important to understand the molecular mechanisms at the host-pathogen interface in the main reservoir species with possible implications in the zoonotic cycle of the pathogen.

Aims Forest fragmentation and reduced tree population densities can poten-tially have negative impacts on mating patterns,offspring genetic diversity and reproductive performance.the aim of the present study is to test these hypotheses comparing an extremely fragmented,low tree density(~0.02 trees/ha)holm oak(Quercus ilex l.)stand from Central spain with a nearby high tree density stand(~50 trees/ha).Methods We genotyped adult trees and seeds from the low-density stand(436 seeds from 15 families)and the high-density stand(404 seeds from 11 families)using nine microsatellite markers.With these data,we performed paternity analyses,determined pollen flow,mating patterns and pollen pool structure,and estimated progeny genetic diversity in both stands.We also studied seed set and production and performed a pollen supplementation experiment to determine whether reduced tree density has limited foreign pollen availability.Important Findings We have found extensive pollen immigration(>75%)into the low tree density stand and monte Carlo simulations revealed that pollen moves larger distances than expected from null models of random dispersal.mating patterns and differentiation of pollen pools were similar in the high-density stand and the low-density stand but we found higher inter-annual differentiation of pollen pools in the for-mer.Progeny genetic diversity and self-fertilization rates did not dif-fer between the low-density stand and the high-density stand.seed set rates were significantly lower in the low-density stand than in the high-density stand and experimental cross-pollen supplementation evidenced that foreign pollen availability is indeed a limiting factor in the former.However,seed crops did not differ between the low-density stand and the high-density stand,indicating that limitation of foreign pollen is not likely to be of great concern in terms of reduced seed production and potential recruitment.Poor forest regeneration due to other ecological and human factors is probably a more important threat for the

Conflictive management of small mammals considered as pests： A long way to evidence-based policy making

This paper discusses the controversial management decisions made by policy-makers worldwide regarding poisoning campaigns aimed to control small mammal populations, often considered harmful economic pests. Aside from considerations regarding the biological consequences of these campaigns, we argue that when society rejects all values of science and expertise then only badly supported and negligent decisions will be made about conservation and management issues. The extermination of small mammal species, some of which play crucial ecological roles in several regions of the world, is just an example of such discredit and misinformation. Without a strong commitment towards evidence-based policy-making, economic investments in research and development could be entirely compromised.

Resource manipulation reveals flexible allocation rules to growth and reproduction in a Mediterranean evergreen oak

Aims In plants,resource allocation to growth and reproduction may depart from trade-off expectations if(i)investment in growth and reproduction relies on different resource pools,(ii)allocation to reproduction is dependent upon reaching some growth threshold or(iii)reproduction is developmentally linked to growth,both functions relying on the same resource pool.We examined the effects of enhanced resource level on patterns of resource allocation to growth and reproduction in holm oak(Quercus ilex sbsp.ballota),a Mediterranean evergreen tree.Methods In the experimental year(2003),we manipulated the amount of soil nutrients in autumn(to increase nutrient uptake during shoot elongation in the following spring)and soil water in summer(to increase water uptake during acorn growth).Indicators of growth and male and female reproduction were estimated in the preexperimental(2002),experimental(2003)and post-experimental(2004)years.Important Findings Fertilized trees produced significantly longer shoots,but the number of female flowers per shoot was not affected by treatments.The production of male catkins was also enhanced by fertilization.Irrigation did not affect the production of female flowers or abortion rates.Growth and female reproduction showed no consistent relationship in untreated trees,but resource addition elicited a growth-female reproduction trade-off in the experimental year.The sign of this significant relationship changed in the post-experimental year,indicating the existence of lagged effects of resource manipulation on acorn production.Overall,patterns of allocation to growth and reproduction varied as a function of sex,resource availability and year,a result consistent with extreme allocational plasticity in holm oak.

Mater nal effects occur whe n the phe no type of the mother in flue nces that of the young to the detriment of her survival, growth or fitn ess. The in vestment of the mother can be affected by mater nal body condition and/or experience. Trivers-Willard Hypothesis (TWH) and Local Resource Competition Hypothesis (LRCH) are the main hypotheses used to explain bias in birth sex-ratios in mammals, as well as for sex-biased maternal investment. Both hypotheses suggest that a different amount of investment must be expected according to the sex of the young. However, recent studies suggest that these differences are not in quantity but in the strategies: mechanisms and objectives may differ for each sex. We studied how maternal characteristics (age, body mass, body condition, and domi nance status) influence re leva nt aspects of the birth and early growth of the calf (birth date, birth body mass, body mass at weaning, and body condition at weaning) separately for each sex;and how that investment is mediated by milk production and composition (lactose, fat, and protein). One hundred eighty-eight newborns from 75 captive red deer hinds aged from 2 to 19 years were analyzed. The main differential investment observed was related to birth date: when producing a female, hinds give birth earlier in the season only if they have a good body condition;however, when gestating a male it is the older hinds those which deliver earlier. Subsequently, milk production and composition are correlated with birth body mass in female calves, but to weaning body mass in males. Thus, only hind body mass affects the weaning body mass of female calves, compared with age and hind body mass in males. These results suggest that while TWH fits the maternal investment strategy found for male calves, it is LRCH which correlates with the maternal investment patterns observed for females.
