AIM: To generate recombinant adenoviral vector con-taining calreticulin (CRT)-hepatitis B surface antigen (HBsAg) fusion gene for developing a safe, effective and HBsAg-specific therapeutic ***: CRT and HBsAg gene were fused using polymerase chain reaction (PCR), endonuclease diges-tion and ligation methods. The fusion gene was cloned into pENTR/D-TOPO transfer vector after the base pairs of DNA (CACC) sequence was added to the 5′ end. Adenoviral expression vector containing CRT-HBsAg fusion gene was constructed by homologous recombinan-tion. The human embryo kidney (HEK) 293A cells were transfected with linearized DNA plasmid of the recombi-nant adenoviral vector to package and amplify recombi-nant adenovirus. The recombinant adenovirus titer was characterized using the end-dilution assay. The expres-sion of the CRT/HBsAg fusion protein in Ad-CRT/HBsAg infected 293A cells was detected by Western ***: The CRT-HBsAg fusion gene was char-acterized by PCR and sequencing and its length and sequence were confirmed to be accurate. The CRT-HB-sAg fusion gene recombinant pENTR/D-TOPO transfer vector was constructed. The recombinant adenoviral vector, Ad-CRT/HBsAg, was generated successfully. The titer of Ad-CRT/HBsAg was characterized as 3.9 × 1011 pfu/mL. The CRT-HBsAg fusion protein was ex-pressed by HEK 293A cells correctly. CONCLUSION: CRT/HBsAg fusion gene recombinant replication-defective adenovirus expression vector is constructed successfully and this study has provided an experimental basis for further studies of Hepatitis B vi-rus gene therapy.

Background This research aims at identifying relative interventions on trachoma and testing the effectiveness of control measures adopted by assessing its prevalence and related risk factors in shandong province of China. Methods Trachoma rapid assessment （TRA） was conducted in 6 sub-districts selected from shandong province based on primary high risk assessment. Active trachoma in children aged 1-9 years and environmental risk factors of trachoma （unclean faces, absence of running water, and absence of flush toilets） were assessed （TRA 1）. Control measures were taken in endemic areas. A second TRA （TRA 2） was conducted after 12 months in the same 6 districts and findings of the two TRAs were compared. Results In TRA 1, we found trachoma in 3 sub-districts and the detection rate was 4% （95% CI： 0.39%-11.12%）, 6% （95% CI： 1.18%-14.17%）, and 6% （95% CI： 1.18%-14.17%） respectively. We could not find trachoma cases in TRA 2. Research data supports that children living with environmental risk factors face an increased risk to active trachoma. However, we could not find statistical evidence for this association, which may be caused by the limited data on prevalence. Conclusions This research indicates that the TRA methodology is easy to assess trachoma and its related risk factors. Based on the results of this study, we have already achieved the goal of ＂elimination of trachoma＂ in shandong province, as the detection rate of trachomatous infiamation follicular/trachomatous inflammation intense in 1-9-year-old children was less than 5%. Nerve cell metabolism in post brain ischemia depends on increased microcirculation perfusion and transport function of microvascular endothelial cells. In the present study, a rat model of middle cerebral artery occlusion was established to investigate the influence of electroacupuncture （EA） on hippocampal CA1 cerebral blood flow and glucose transporter 1 （GLUT1） expression in the microvascular endothelial cells. Following EA at Neiguan （PC 6）, the cerebral blood flow in the ischemic hippocampal CA1 region was significantly elevated, the number and microvascular integrated absorbance of the GLUTl-positive cells were significantly increased, nerve cell damage was ameliorated, and GLUT1 protein expression in the ischemic hippocampus was significantly increased. Results demonstrate that EA increased the cerebral blood flow of the hippocampal CA1 region and improved the glucose transport function, thereby attenuating neuronal injuries.

AIM:To explore the expression and function of slug,a transcriptional repressor,in human intrahepatic cholangiocarcinoma(IHCC)and identify its role in IHCC ***:Expression of slug was detected in 36 cases of IHCC and 12 cases of normal intrahepatic bile ducts and liver parenchyma by *** patients were divided into low slug expression group(< 20%of carcinoma cells stained)and high slug expression group(≥20%of carcinoma cells stained).Slug expression was correlated with clinicopathological parameters of IHCC *** patients were defined as short-term survivors if their survival time was<12 mo and as longterm survivors if their survival time was≥12 ***:Slug was not expressed in normal liver epi-thelium samples,lowly expressed in 15 tissue samples (10-,5+)and highly expressed in 21 tissue samples (16++;5+++)from IHCC *** survival rate of patients with a low slug expression was 33.3%(n =5)and 66.7%(n=10),*** survival rate of patients with a high slug expression was 61.9% (n=13)and 38.1%(n=8),respectively(P=0.02).Lymph node metastasis was found in 4(26.7%)out of the 15 patients with a low slug expression and in 14(66.7%)out of the 21 patients with a high slug expression,*** incidence rate of lymph node metastasis increased with the increasing slug expression level(P=0.003),and higher in patients with a high slug expression than in those with a low slug *** expression did not significantly correlate with the tumor size and stage or histologic grade,or with the gender and age of ***:Slug expression is a novel prognostic marker for IHCC with lymph node metastasis.

Loss of p120 catenin aggravates alveolar edema of ventilation induced lung injury

Background p120 catenin （p120ctn） is an adheren junction protein that regulates barrier function, but its role has not been explored in alveolar edema induced by ventilation. We measured stretch-induced cell gap formation in MLE 12 cells due to the loss of p120. We hypothesized that alveolar permeability was increased by high tung inflation associated with alveolar epithelia cell tight junctions being destroyed, which resulted from the loss of p120. Methods Cultured MLE12 cells were subjected to being stretched or un-stretched （control） and some cells were pretreated with pp2 （c-src inhibitor）. After the end of stretching for 0, 1, 2, and 4 hours, the cells were lysed, and p120 expression and c-src activation was determined by Western blotting analysis. In vivo, SD rats were taken to different tidal volumes （Vt 7 ml/kg or 40 ml/kg, PEEP=0, respiratory rate 30-40 betas/min） for 0, 1, 2, and 4 hour and some were pretreated with pp2, and alveolar edema was calculated. Rerults It was found that p120 expression was reduced and c-src activation increased in a time-dependent and strain- dependent manner due to cyclic-stretch of the alveolar epithelial cells. These changes could be reversed by inhibition of c-src. We obtained similar changes in rats when they were subjected to large tidal volumes and the alveolar edema increased more than in rats in the low Vt group. Pretreated the rats with inhibition of c-src had less pulmonary edema induced by the high tidal volume ventilation. Conclusions Cyclic stretch MLE 12 cells induced the loss of p120 and may be the same reason by high tidal volume ventilation in rats can aggravate alveolar edema. Maintenance of p120 expression may be a novel therapeutic strategy for the prevention and treatment of ventilation induced lung injury （VILI）.
