Objective：To investigate the cellular effects of Pien Tze Huang（片仔癀,PZH） in the HT-29 human colon carcinoma cell ***：The viability of HT-29 cells was determined by MTT assay.A fluorescence-activated cell sorting（FACS） analysis with annexin-V/propidium iodide（PI） and JC-1 staining were performed to determine cell apoptosis and the loss of mitochondrial membrane potential,*** of caspase 3 was evaluated by a colorimetric *** mRNA expression levels of Bcl-2 and Bax were measured by reverse transcription polymerase chain reaction（RT-PCR）.Results：PZH,in a dose- and time-dependent manner,reduced viability and induced apoptosis of HT-29 ***,PZH treatment resulted in the collapse of the mitochondrial membrane potential,activation of caspase 3,and an increase in the Bax/Bcl-2 ***：PZH inhibits the growth of HT-29 cells by inducing cancer cell apoptosis via regulation of the Bcl-2 family and activation of caspase 3,which may,in part,explain its anticancer activity.

促排卵是多囊卵巢综合征(PCOS)不孕症的一线治疗方案。卵巢对促排卵治疗的排卵应答差被认为与胰岛素抵抗和高雄激素血症相关。在一个包含1000名PCOS不孕妇女(PCOSAct)的前瞻性队列中，我们开展了一项全外显子联合靶向单核苷酸多态性(SNP)测序以及代谢组学研究。在全基因组水平找出与无排卵显著相关的常见变异和罕见突变，并通过机器学习算法构建排卵预测模型。研究发现，ZNF438基因中标记为rs2994652 (p=2.47×10–8)的常见变异和REC114基因中的一个罕见功能突变(rs182542888,p=5.79×10–6)与促排卵治疗失败显著相关。携带rs2994652 A等位基因和REC114 ***101Leu (rs182542888)的PCOS不孕妇女进行促排卵治疗的总排卵率更低(分别为比值比(OR)=1.96,95%置信区间(CI)[1.55～2.49];OR=11.52,95%CI [3.08～43.05])，出现排卵的间隔时间更长(平均56.7天vs.49.0天，p<0.001;78.1天vs.68.6天，p=0.014)。对于rs2994652突变者，L-苯丙氨酸水平升高并与胰岛素抵抗稳态模型(HOMA-IR)指数(r=0.22, p=0.05)和空腹血糖(r=0.33, p=0.003)呈正相关；对于rs182542888突变者，花生四烯酸代谢产物水平下降并与升高的抗苗勒管激素(r=-0.51, p=0.01)和总睾酮(r=-0.71, p=0.02)呈负相关。整合基因变异位点、代谢产物及临床特征的联合预测模型可提高对排卵的预测能力[曲线下面积(AUC)=76.7%]。ZNF438基因的一个常见变异和REC114基因的一个罕见功能突变，以及与二者相关的苯丙氨酸和花生四烯酸代谢物改变，与PCOS女性不孕症的促排卵治疗失败相关。

Detailed characterizations of genomic alterations have not identified subtype-specific vulnerabilities in adult gliomas. Mapping gliomas into developmental programs may uncover new vulnerabilities that are not strictly related to genomic alterations. After identifying conserved gene modules co-expressed with EGFR or PDGFRA (EM or PM), we recently proposed an EM/PM classification scheme for adult gliomas in a histological subtype- and grade-independent manner. By using cohorts of bulk samples, paired primary and recurrent samples, multi-region samples from the same glioma, single-cell RNA-seq samples, and clinical samples, we here demonstrate the temporal and spatial stability of the EM and PM subtypes. The EM and PM subtypes, which progress in a subtype-specific mode, are robustly maintained in paired longitudinal samples. Elevated activities of cell proliferation, genomic instability and microenvironment, rather than subtype switching, mark recurrent gliomas. Within individual gliomas, the EM/PM subtype was preserved across regions and single cells. Malignant cells in the EM and PM gliomas were correlated to neural stem cell and oligodendrocyte progenitor cell compartment, respectively. Thus, while genetic makeup may change during progression and/or within different tumor areas, adult gliomas evolve within a neurodevelopmental framework of the EM and PM molecular subtypes. The dysregulated developmental pathways embedded in these molecular subtypes may contain subtype-specific vulnerabilities.

目的观察褪黑素受体激动剂(Neu-P11)对眼压的影响以及水通道蛋白1在正常眼压兔眼组织中的表达。方法新西兰兔30只随机分为对照组、褪黑素组和Neu-P11组。用局部点眼法,1个月后处死动物,取眼前段组织;用免疫组化法、RT-PCR法和Western blot法检测。结果 50μmol·L-1Neu-P11点眼,可使兔眼压降低,1 h达到最低(P<0.01)。Neu-P11组和褪黑素组,角膜内皮、虹膜基质血管和色素上皮细胞、睫状体非色素上皮以及小梁网组织细胞的胞膜和胞浆均可见阳性表达,且阳性反应数明显增多。Neu-P11组和褪黑素组AQP1mRNA和蛋白质水平较对照组上调,且Neu-P11组比褪黑素组更高,2组差异有统计学意义(P<0.05)。结论 Neu-P11能够降低兔眼压,并能够使兔眼组织中AQP1的表达增高。

The fuel reforming system for a fuel cell vehicle requires a compact and low-cost static mixer that can uniformly mix the feed gas regardless of the flow rate. For that reason, the mixer is frequently designed with a simple structure of parallel distribution tubes. Such a gas mixer has multiple tubes arranged in parallel in the cross section of the main duct. Each tube has multiple tiny holes arranged in a row on its wall face through which gas is blown out. However, it is very difficult to achieve uniform flow rates from all the holes under both high and low flow rate *** fluid analysis is effective in solving this problem. Under a low flow rate condition, the flow through the tubes is laminar because of the small Reynolds number associated with the tube diameter, whereas a high flow rate results in turbulent flow because of the large Reynolds ***, at intermediate flow rates, the Reynolds number is in a transitional state. Previously,when a transitional state was present in the calculation area, sufficient prediction accuracy was not obtained because the turbulent model could not be adapted suitably to different parts. Consequently, we adopted a new calculation method that determines the Reynolds number in each flow during a calculation and automatically switches between use and non-use of the turbulent model. The calculated results were compared with experimental measurements obtained by 3-D stereo particle image velocimetry (PIV). Good agreement was seen between the two sets of results, which confirmed the validity of the new calculation method.

The goal of this study was to evaluate the effects of a mixture of yeast culture, cell wall hydrolysates, and yeast extracts（collectively ＂yeast products,＂ YP） on the performance, intestinal physiology, and health of weaned piglets. A total of 90 piglets weaned at 21 d of age were blocked by body weight, sex, and litter and randomly assigned to one of three treatments for a 14-d feeding experiment, including（1） a basal diet（control）,（2） 1.2 g/kg of YP, and（3） 20 mg/kg of colistin sulfate（CSE）. No statistically significant differences were observed in average daily feed intake, average daily weight gain, or gain-to-feed ratio among CSE, YP, and control piglets. Increased prevalence of diarrhea was observed among piglets fed the YP diet, whereas diarrhea was less prevalent among those fed CSE. Duodenal and jejunal villus height and duodenal crypt depth were greater in the control group than they were in the YP or CSE groups. Intraepithelial lymphocytes（IEL） in the duodenal and jejunal villi were enhanced by YP, whereas IEL in the ileal villi were reduced in weaned piglets fed YP. Secretion of jejunal and ileal interleukin-10（IL-10） was higher and intestinal and serum antioxidant indexes were affected by YP and CSE. In YP- and CSE-supplemented animals, serum D-lactate concentration and diamine oxidase（DAO） activity were both increased, and intestinal mR NA expressions of occludin and ZO-1 were reduced as compared to the control animals. In conclusion, YP supplementation in the diets of weaned piglets appears to increase the incidence of diarrhea and has adverse effects on intestinal morphology and barrier function.

We reported here the fabrication of the elec- trically tunable infrared （IR） reflectors based on the polymer stabilized cholesteric liquid crystal （PSCLC） with negative dielectric anisotropy. A systematic study of the influence of cell gap on the electrically tunable reflection bandwidth was performed. When a direct current （DC） electric field was ap- plied, the reflection bandwidth red shifted in the cells with small cell gap, whereas the bandwidth broadening was ob- served in the cells with large cell gap. It is therefore reasonable to deduct that the reflection is dictated by the pitch gradient steepness which strongly relies on the cell thickness. The re- sults reveal that for making PSCLC based IR reflector windows with electrically induced bandwidth broadening, a minimal cell gap thickness is required. The resulted IR reflectors pos- sess a short native switching time and long-term operation stability, and are potentially applicable as smart energy saving windows in buildings and automobiles.

Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid *** aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical ***:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib ***,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent *** points included safety,tolerability and anti-tumour ***:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 *** most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease *** the famitinib monotherapy stage,three patients(15%)showed partial *** complete response rate was 65%at the completion of treatment and 95%3 months after the treatment *** a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,*** with a decrease of perfusion p
