Cardiac resident macrophages (CRMs) are the main population of cardiac immune cells. The role of these cells in regeneration, functional remodeling, and repair after cardiac injury is always the focus of research. However, in recent years, their dynamic changes and contributions in physiological states have a significant attention. CRMs have specific phenotypes and functions in different cardiac chambers or locations of the heart and at different stages. They further show specific differentiation and development processes. The present review will summarize the new progress about the spatiotemporal distribution, potential developmental regulation, and their roles in cardiac development and aging as well as the translational potential of CRMs on cardiac diseases. Of course, the research tools for CRMs, their respective advantages and disadvantages, and key issues on CRMs will further be discussed.

Key epidemiological parameters,including the effective reproduction number,,and the instantaneous growth rate,,generated from an ensemble of models,have been informing public health policy throughout the COVID-19 pandemic in the four nations of the United Kingdom of Great Britain and Northern Ireland(UK).However,estimation of these quantities became challenging with the scaling down of surveillance systems as part of the transition from the“emergency”to“endemic”phase of the *** Office for National Statistics(ONS)COVID-19 Infection Survey(CIS)provided an opportunity to continue estimating these parameters in the absence of other data *** used a penalised spline model fitted to the publicly-available ONS CIS test positivity estimates to produce a smoothed estimate of the prevalence of SARS-CoV-2 positivity over *** resulting fitted curve was used to estimate the“ONS-based”and across the four nations of the *** produced under this model are compared to government-published estimates with particular consideration given to the contribution that this single data stream can offer in the estimation of these *** on the nation and parameter,we found that up to 77%of the variance in the government-published estimates can be explained by the ONS-based estimates,demonstrating the value of this singular data stream to track the epidemic in each of the four *** additionally find that the ONS-based estimates uncover epidemic trends earlier than the corresponding government-published *** work shows that the ONS CIS can be used to generate key COVID-19 epidemiological parameters across the four UK nations,further underlining the enormous value of such population-level studies of *** is not intended as an alternative to ensemble modelling,rather it is intended as a potential solution to the aforementioned challenge faced by public health officials in the UK in early 2022.

High-throughput preparation of plasmid DNA libraries for next-generation sequencing(NGS)is an important capability for molecular biology *** particular,it is an essential quality control(QC)check when large numbers of plasmid variants are being ***,we describe the use of the Design of Experiments(DOE)methodology to optimise the miniaturised preparation of plasmid DNA libraries for NGS,using the Illumina®Nextera XT technology and the Labcyte Echo®acoustic liquid dispensing ***,we describe methods which can be implemented as a QC check for identifying the presence of genomic DNA(gDNA)in plasmid DNA samples and the subsequent shearing of the gDNA,which otherwise prevents the acoustic transfer of plasmid *** workflow enables the preparation of plasmid DNA libraries which yield high-quality sequencing data.

In view of its potent microbicidal actions, ozone (O3) offers much potential for application as a therapeutic agent in oral health, e.g. in the treatment of dental caries. This oxidant is extremely reactive towards biomolecules present in the oral environment, and in this study we have employed high-resolution proton (1H) nuclear magnetic resonance (NMR) spectroscopy to determine the nature and extent of the oxidation of biomolecules known to be present in carious dentin, plaque and saliva. Phosphate-buffered (pH 7.00) aqueous solutions containing sodium pyruvate, α-D-glucose, L-cys teine and L-methionine (5.00 mM) were treated with gaseous O3 (4.48 mmol.) delivered by a therapeutic O3 generating device. Attack of O3 on methionine and cysteine generated the corresponding primary oxidation products of these substrates, specifically methionine sulphoxide [98% ± 4% (mean ± SEM) yield] and cystine (95% ± 6% yield) respectively, and treatment of pyruvate with this oxidant produced acetate and CO2 via an oxidative decarboxylation process (93% ± 4% yield). Reaction of O3 with α-D-glucose gave rise to formate as a major product (24% ± 2% yield). In conclusion, multicomponent 1H NMR analysis of appropriate chemical model systems provides valuable molecular information regarding the reactivity of O3 towards biomolecules present in the oral environment, information which is of much relevance to its therapeutic mechanisms of action. Moreover, in view of the much higher concentrations of these O3-scavenging biomolecules in oral fluid and/or soft tissue environments than that of O3 applied, they may also serve to offer protection against putative adverse effects inducible by any of this oxidant which escapes from its site of therapeutic application (e.g., at primary root carious lesions).

The article examines the tradition of the writing of Encyclopaedias and historical dictionaries in seventeenth and eighteenth century France. The main ones--by Bayle, chambers, and by the group assembled by d＇Alembert and Diderot--are all connected with unorthodoxy in religion. The massive collection of knowledge that all three dictionaries compiled sometimes seems to allow a juxtaposition of ideas which cannot be properly reconciled--a situation which leaves it to the reader to create a coherent whole. But Diderot goes the farthest in this direction, and causes even the possibility of such a whole to be questioned.

Electrocatalytic splitting of water by means of renewable energy as the electricity supply is one of the most promising methods for storing green renewable energy as hydrogen. Although two-thirds of the earth’s surface is covered with water, there is inadequacy of freshwater in most parts of the world. Hence, splitting seawater instead of freshwater could be a truly sustainable alternative. However, direct seawater splitting faces challenges because of the complex composition of seawater. The composition, and hence, the local chemistry of seawater may vary depending on its origin, and in most cases, tracking of the side reactions and standardizing and customizing the catalytic process will be an extra challenge. The corrosion of catalysts and competitive side reactions due to the presence of various inorganic and organic pollutants create challenges for developing stable electro-catalysts. Hence, seawater splitting generally involves a two-step process, i.e., purification of seawater using reverse osmosis and then subsequent fresh water splitting. However, this demands two separate chambers and larger space, and increases complexity of the reactor design. Recently, there have been efforts to directly split seawater without the reverse osmosis step. Herein, we represent the most recent innovative approaches to avoid the two-step process, and compare the potential application of membrane-assisted and membrane-less electrolyzers in direct seawater splitting(DSS). We particularly discuss the device engineering, and propose a novel electrolyzer design strategies for concentration gradient based membrane-less microfluidic electrolyzer.

Acute myeloid leukaemia(AML)patients harbouring certain chromosome abnormalities have particularly adverse *** these patients,targeted therapies have not yet made a significant clinical *** understand the molecular landscape of poor prognosis AML we profiled 74 patients from two different centres(in UK and Finland)at the proteomic,phosphoproteomic and drug response phenotypic *** data were complemented with transcriptomics analysis for 39 *** integration highlighted a phosphoproteomics signature that define two biologically distinct groups of KMT2A rearranged leukaemia,which we term MLLGA and *** presented increased DOT1L phosphorylation,HOXA gene expression,CDK1 activity and phosphorylation of proteins involved in RNA metabolism,replication and DNA damage when compared to MLLGB and no KMT2A rearranged *** was particularly sensitive to 15 compounds including genotoxic drugs and inhibitors of mitotic kinases and inosine-5-monosphosphate dehydrogenase(IMPDH)relative to other ***-risk KMT2A-MLLT3 cases were mainly represented in a third group closer to MLLGA than to *** expression of IMPDH2 and multiple nucleolar proteins was higher in MLLGA and correlated with the response to IMPDH inhibition in KMT2A rearranged leukaemia,suggesting a role of the nucleolar activity in sensitivity to *** summary,our multilayer molecular profiling of AML with poor prognosis and KMT2A-MLLT3 karyotypes identified a phosphoproteomics signature that defines two biologically and phenotypically distinct groups of KMT2A rearranged *** data provide a rationale for the potential development of specific therapies for AML patients characterised by the MLLGA phosphoproteomics signature identified in this study.
