NIR driven catalytic enhanced acute lung injury therapy by using polydopamine@Co nanozyme via scavenging ROS

Acute lung injury(ALI) was characterized by excessive reactive oxygen species(ROS) levels and inflammatory response in the lung. Scavenging ROS could inhibit the excessive inflammatory response, further treating ALI. Herein, we designed a novel nanozyme(P@Co) comprised of polydopamine(PDA) nanoparticles(NPs) loading with ultra-small Co, combining with near infrared(NIR) irradiation, which could efficiently scavenge intracellular ROS and suppress inflammatory responses against ALI. For lipopolysaccharide(LPS) induced macrophages, P@Co + NIR presented excellent antioxidant and anti-inflammatory capacities through lowering intracellular ROS levels, decreasing the expression levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α) as well as inducing macrophage M2 directional polarization. Significantly, it displayed the outstanding activities of lowering acute lung inflammation, relieving diffuse alveolar damage, and up-regulating heat shock protein 70(HSP70) expression, resulting in synergistic enhanced ALI therapy effect. It offers a novel strategy for the clinical treatment of ROS related diseases.

Exploring the mechanism of Xiaoaiping Injection inhibiting autophagy in prostate cancer based on proteomics

Xiaoaiping(XAP) Injection demonstrates the anti-prostate cancer(PCa) effects, yet the underlying mechanism remains unclear. This study aims to investigate the impact of XAP on PCa and elucidate its mechanism of action. PCa cell proliferation was evaluated using a cell counting kit-8(CCK-8) assay. Cell apoptosis was assessed through Hoechst staining and Western blotting assays. Proteomics technology was employed to identify key molecules and significant signaling pathways modulated by XAP in PCa cells. To further validate potential key genes and important pathways, a series of assays were conducted, including acridine orange(AO)staining, transmission electron microscopy, and immunofluorescence assays. The molecular mechanism of XAP against PCa in vivo was examined using a PC3 xenograft mouse model. Results demonstrated that XAP significantly inhibited cell proliferation in multiple PCa cell *** C4-2 and prostate cancer cell line-3(PC3) cells, XAP induced cellular apoptosis, evidenced by reduced B-cell lymphoma 2(Bcl-2) levels and elevated Bcl-2-associated X(Bax) levels. Proteomic, immunofluorescence, and quantitative reverse transcription-polymerase chain reaction(q RT-PCR) investigations revealed a strong correlation between forkhead box O3a(Fox O3a) autophagic degradation and the anti-PCa action of XAP. XAP hindered autophagy by reducing the expression levels of autophagy-related protein 5(Atg5)/autophagy-related protein 12(Atg12) and enhancing Fox O3a expression and nuclear translocation. Furthermore,XAP exhibited potent anti-PCa action in PC3 xenograft mice and triggered Fox O3a nuclear translocation in tumor tissue. These findings suggest that XAP induces PCa apoptosis via inhibition of Fox O3a autophagic degradation, potentially offering a novel perspective on XAP injection as an effective anticancer therapy for PCa.

Study on the diagnostic criteria of syndrome of dampness obstruction in idiopathic membranous nephropathy based on expert consensus

OBJECTIVE: To reach consensus on the diagnostic criteria of syndrome of dampness obstruction in idiopathic membranous nephropathy(IMN) patients by literature research and expert investigation(interviews and a Delphi method). METHODS: Our study was consistent with T/CACM 1336-2020. We searched the monographs and references published in the past 40 years(1983-2022), and established the diagnostic criteria pool of waterdampness syndrome and dampness-turbidity syndrome in Traditional chinese medicine(TCM) based on literature by using frequency statistics and correlation analysis. Expert investigation(interview method and two rounds of Delphi method) was used to form the diagnostic criteria of water-dampness syndrome and dampnessturbidity syndrome of idiopathic membranous nephropathy. Clinical diagnostic test research was carried out, and compared with “Diagnostic Criteria for dampness syndrome”(T/CACM 1454-2023) to evaluate the authenticity, reliability and clinical application value of the standard. RESULTS: A total of 122 relevant guides, standards, monographs and documents were included through searching books and chinese databases. Four experts were interviewed and two rounds of delphi method(75 experts nationwide) were carried out. The experts' opinions are relatively concentrated and the differences are small. Based on the weight of each index, the diagnostic criteria indexes of water-dampness syndrome and dampness-turbidity syndrome were selected. After discussion by the core group members, the diagnostic model of "necessary symptoms and optional symptoms" was established, and the final diagnostic criteria of waterdampness syndrome and dampness-turbidity syndrome were established. One hundred and ninety-one inpatients and outpatients of Guangdong Provincial hospital of chinese medicine from January 2021 to February 2023 were included in Diagnostic test study. Electroacupuncture enhances the mitophagy of granulosa cells in premature ovarian insufficiency model mice by inactivating the hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein,drosophila disc large tumor suppresso

OBJECTIVE: To investigate the potential mechanism of electroacupuncture（EA） in alleviating premature ovarian insufficiency（POI） and to provide a theoretical basis for EA treatment of POI. METHODS: For this purpose, a POI mice model was developed by injecting 12 mg/kg busulfan and 120 mg/kg cyclophosphamide intraperitoneally to induce POI. It was then proceeded by EA intervention at Guanyuan（CV4） acupoint on the second day following modeling. Similarly, apoptosis in ovarian granulosa cells was detected by terminal deoxynucleotidyl transferase d UTP nick end labeling staining, while enzyme-linked immunosorbent assay was employed for measuring serum folliclestimulating hormone（FSH）, luteinizing hormone（LH）, estrogen（E2）, and anti-müllerian hormone（AMH） levels. Moreover, transmission electron microscopy（TEM） was employed for examining mitochondrial morphology, while autophagy and hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein, drosophila disc large tumor suppressor, and zonula occludens-1 protein binding motif（YAP/TAZ） pathway related protein levels in ovarian tissue were detected via Western blotting. RESULTS: Analysis of serum levels of various hormones indicated that serum FSH and LH were reduced in EA compared to the POI group, while E2and AMH levels were found to be elevated in EA compared to the POI group. The EA was found to inhibit apoptosis in granulosa cells in POI model mice, in addition to improved mitochondrial damage and significantly improved mitophagy. Pathway analysis revealed that EA was involved in activating the hippo-YAP/TAZ pathway, followed by reversing EA effects on granulosa cell apoptosis and mitophagy with the use of verteporfin, an autophagy and YAP-T-cell factor/enhancer of split and activator of transcription domain family member interaction inhibitor. Objective:To explore the efficacy and safety of Juan Bi Pill(JBP)in treatment of active rheumatoid arthritis(RA).Methods:From February 2017 to May 2018,115 participants from 4 centers were randomly divided into JBP group(57 cases)and placebo group(58 cases)in a 1:1 ratio using a random number table *** received a dose of JBP(4 g,twice a day,orally)combined with methotrexate(MTX,10 mg per week)or placebo(4 g,twice a day,orally)combined with MTX for 12 *** were required with follow-up visits at 24 and 48 weeks,attending 7 assessment *** were undergo disease activity assessment 7 times(at baseline and 2,4,8,12,24,48 weeks)and safety assessments 6 times(at baseline and 4,8,12,24,48 weeks).The primary endpoint was 28-joint Disease Activity Score(DAS28-ESR and DAS28-CRP).The secondary endpoints included American College of Rheumatology(ACR)criteria for 20%and 50%improvement(ACR20/50),Health Assessment Questionnaire Disability Index(HAQ-DI),clinical disease activity index(CDAI),visual analog scale(VAS),Short Form-36(SF-36)score,Medial Outcomes Study(MOS)sleep scale score,serum erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),tender joint count,swollen joint count,and morning *** adverse reactions were observed during the ***:After 12 weeks of treatment,DAS28-ESR and DAS28-CRP scores in both groups were lower than before treatment(both P<0.01),while the remission rate of DAS28-ESR and DAS28-CRP and low disease activity of JBP group were higher than those in the placebo group(both P<0.01).JBP demonstrated better efficacy on ACR20 and ACR50 compliance rate at 12 and 48 weeks comparing to placebo(all P<0.05).The CDAI and HAQ-DI score,pain VAS and global VAS change of RA patients and physicians,the serum ESR and CRP levels,and the number of tenderness and swelling joints were lower than before treatment at 4,8,12,24,48 weeks in both groups(P<0.05 or P<0.01),while the reduction of above indices in

Gut microbial characteristics of the damp-heat constitution:a population-based multicenter cross-sectional study

OBJECTIVE: To investigate the difference in gut microbiota between population with damp-heat constitution(DHC) and balanced constitution(BC). METHODS: A multi-centered cross-sectional casecontrol study was conducted, which included 249 participants with damp-heat constitution or balanced constitution. Baseline information of participants was collected, and stool samples were collected for gut microbiota analysis. Principal coordinate analysis, linear discriminant analysis effect size analysis, receiver operating characteristic, random forest model, and phylogenetic investigation of communities by reconstruction of unobserved states methods were used to reveal the relationship between gut microbiota and the damp-heat constitution. RESULTS: Compared to those in the BC group, the richness and diversity of the microbiota, specifically those of several short-chain fatty acid producing genera such as Barnesiella, Coprobacter, and Butyricimonas, were significantly decreased in the DHC group. Regarding biological functions, flavonoid biosynthesis, propanoate metabolism, and nucleotide sugar metabolism were suppressed, while arachidonic acid metabolism and glutathione metabolism were enriched in the DHC group. Finally, a classifier based on the microbiota was constructed to discriminate between the DHC and BC populations. CONCLUSION: The gut microbiota of the DHC population exhibits significantly reduced diversity and is closely related to inflammation, metabolic disorders, and liver steatosis, which is consistent with clinical observations, thus serving as a potential diagnostic tool for traditional chinese medicine constitution discrimination.

Different Acupuncture Therapies for Postherpetic Neuralgia:An Overview of Systematic Reviews and Meta-analysis

Background: Postherpetic neuralgia(PHN) is the most common complication of herpes zoster infection and affects patients' quality of life. Acupuncture therapy is regarded as a competitive method of treatment for analgesia. Objective: To summarize evidence from systematic reviews(SRs) and evaluate the effectiveness and safety of different acupuncture therapies for treating PHN. Methods: Eight electronic databases were searched from their inception to August 5, 2022, including 4 international electronic databases(PubMed, EMBASE, the Cochrane Library, and Web of Science) and 4 chinese databases(chinese Biomedical Database, China National Knowledge Infrastructure, VIP Database and Wanfang Database).Methodological quality was assessed by A Measurement Tool to Assess Systematic Reviews 2(AMSTAR 2).The Risk of Bias in Systematic Review(ROBIS) tool was used to assess the risk of bias in SRs. Evidence level was assessed by the Grading of Recommendations Assessment, Development, and Evaluation(GRADE)approach. Results: Totally, 7 SRs were included, including 128 studies and 9,792 patients. In AMSTAR 2, most of the SRs were of low or critically low levels since they had more than 1 critical deficiency. In ROBIS, 1 SR(14.29%)was rated as high risk, and the other 6(85.71%) were rated as low risk. In the GRADE system, 9 outcomes(28.13%)were valued as high level, 5(15.63%) as moderate level, 1(3.13%) as low, and 17(53.13%) as very low. In the effectiveness of acupuncture therapy, the group "moxibustion vs. original medical treatment" [mean difference(MD)=–1.44, 95% confidence interval(CI): –1.80 to –1.08, I2=99%, P<0.00001] was of the highest heterogeneity and the group "bloodletting vs. original medical treatment"(MD=–2.80, 95% CI: –3.14 to –2.46, I2=0, P<0.00001)was of the lowest heterogeneity.
