Chinese Assam tea(Camellia sinensis ***)is an important tea crop with a long history of cultivation in Yunnan,*** its potential value as a genetic resource,its genetic diversity and domestication/breeding history remain *** address this issue,we genotyped 469 ancient tea plant trees representing 26 *** *** populations,plus two of its wild relatives(six and three populations of *** and ***,respectively)using 16 nuclear microsatellite *** showed that Chinese Assam tea has a relatively high,but comparatively lower gene diversity(H_(S)=0.638)than the wild relative ***(H_S=0.658).Clustering in STRUCTURE indicated that Chinese Assam tea and its two wild relatives formed distinct genetic groups,with considerable interspecific *** Chinese Assam tea accessions clustered into three gene pools,corresponding well with their geographic ***,New Hybrids analysis indicated that 68.48%of ancient Chinese Assam tea plants from Xishuangbanna were genetic intermediates between the Puer and Lincang gene *** addition,10%of the ancient Chinese Assam tea individuals were found to be hybrids between Chinese Assam tea and *** results suggest that Chinese Assam tea was domesticated separately in three gene pools(Puer,Lincang and Xishuangbanna)in the Mekong River valley and that the hybrids were subsequently selected during the domestication *** the domestication history of Chinese Assam tea in southwestern Yunnan remains complex,our results will help to identify valuable genetic resources that may be useful in future tea breeding programs.

Online prediction of network-level public transport demand based on principle component analysis

Online demand prediction plays an important role in transport network services from operations,controls to management,and information ***,the online prediction models are impacted by streaming data quality issues with noise measurements and missing *** address these,we develop a robust prediction method for online network-level demand prediction in public *** consists of a PCA method to extract eigen demand images and an optimization-based pattern recognition model to predict the weights of eigen demand images by making use of the partially observed real-time data up to the prediction time in a *** prediction model is robust to data quality issues given that the eigen demand images are stable and the predicted weights of them are optimized using the network level data(less impacted by local data quality issues).In the case study,we validate the accuracy and transferability of the model by comparing it with benchmark models and evaluate the robustness in tolerating data quality issues of the proposed *** experimental results demonstrate that the proposed Pattern Recognition Prediction based on PCA(PRP-PCA)consistently outperforms other benchmark models in accuracy and ***,the model shows high robustness in accommodating data quality *** example,the PRP-PCA model is robust to missing data up to 50%regardless of the noise *** also discuss the hidden patterns behind the network level *** visualization analysis shows that eigen demand images are significantly connected to the network structure and station activity *** the demand changes dramatically before and after the pandemic,the eigen demand images are consistent over time in Stockholm.

Osteopontin increases hepatocellular carcinoma cell growth in a CD44 dependant manner

AIM:To investigate the role of osteopontin(OPN) and its splice variants in the proliferation of hepatocellular carcinoma(HCC).METHODS:The expression of OPN variants in HCC cell lines as well as HCC tissue samples and nontumour tissue was studied using polymerase chain *** variant cDNAs were cloned into a mammalian expression vector allowing both transient expression and the production of stable OPN expressing cell *** expression was studied in these cells using Western blotting,immunofluoresnce and enzyme linked immunosorbent assay.A CD44 blocking antibody and siRNA targeting of CD44 were used to examine the role of this receptor in the OPN stimulated cell growth observed in ***-7 cells stably expressing either OPN-A,-B or-C were injected subcutaneously into the flanks of nude mice to observe in vivo tumour *** of OPN mRNA and protein in these tumours was examined using reverse transcriptionpolymerase chain reaction and ***:OPN is expressed in HCC in 3 forms,the full length OPN-A and 2 splice variants OPN-B *** variant expression was noted in HCC tissue as well as cognate surrounding cirrhotic liver *** of these OPN variants in the HCC derived cell line Huh-7 resulted in secretion of OPN into the culture *** of OPN conditioned media to na ve Huh-7 and HepG2 cells resulted in significant cell growth suggesting that all OPN variants can modulate cell proliferation in a paracrine *** the OPN mediated increase in cellular proliferation was dependent on CD44 as only CD44 positive cell lines responded to OPN conditioned media while siRNA knockdown of CD44 blocked the proliferative *** expression also increased the proliferation of Huh-7 cells in a subcutaneous nude mouse tumour model,with Huh-7 cells expressing OPN-A showing the greatest proliferative ***:This study demonstrates that OPN plays a significant role in the proliferation of HCC thro

Background:Newly diagnosed breast cancer patients experience symptoms that may affect their quality of life,treatment outcomes,and *** and managing breast cancer-related symptoms soon after diagnosis is *** purpose of this study was to investigate the associations between health-related fitness(HRF)and patient-reported symptoms in newly diagnosed breast cancer ***:This study utilized baseline data from the Alberta Moving Beyond Breast Cancer Cohort Study that were collected within 90 days of *** measures included peak cardiopulmonary fitness(peak volume of oxygen consumption(VO_(2peak))),maximal muscular strength and endurance,flexibility,and body *** measures included depression,sleep quality,and *** multivariable logistic regression was performed for ***:Of 1458 participants,51.5%reported poor sleep quality,26.5%reported significant fatigue,and 10.4%reported moderate *** multivariable-adjusted models,lower relative VO_(2peak)was independently associated with a greater likelihood of all symptom measures,including moderate depression(p<0.001),poor sleep quality(p=0.009),significant fatigue(p=0.008),any symptom(p<0.001),and multiple symptoms(p<0.001).VO_(2peak)demonstrated threshold associations with all symptom measures such that all 3 lower quartiles exhibited similar elevated risk compared to the highest *** strength of the threshold associations varied by the symptom measure with odds ratios ranging from-1.5 for poor sleep quality to-3.0 for moderate depression and multiple ***,lower relative upper body muscular endurance was also independently associated with fatigue in a dose-response manner(p=0.001),and higher body weight was independently associated with poor sleep quality in an inverted U pattern(p=0.021).Conclusion:Relative VO_(2peak)appears to be a critical HRF component associated with multiple patient-reported symptoms in newly diag

Synchronous colorectal Iiver metastases: timing of resection and patterns of referral

There remain no consensus guidelines on the optimum surgical strategy for the management of synchronous colorectal liver metastases in spite of a number of published reviews and meta-analyses(1,2).In their paper,Howard et *** the‘real world’referral patterns and resection pathways of canadian surgeons for this challenging patient cohort(3).This is a timely and interesting area for *** was generated from an online survey sent out to all surgeons in *** were received from 58 surgeons but 6 responses were inadequate for analysis leaving 52 surgeon responses included for *** study does not indicate the total number of surgeons who were invited to participate and so the proportional response rate is *** all survey based-studies,the small number of respondents included does raise the question of inherent selection bias which cannot be *** addition,almost all included surgeons had access to hepatobiliary services,with over half reporting access to hepatobiliary surgeons within their own institutions,such availability would undoubtedly affect decision making in this *** information around the nature and scope of general surgical and surgical oncology training in Canada would have been useful in understanding the likely experience of *** this paper raises some interesting points in the management of patients with synchronous colorectal liver metastases.

Why Can’t Canada Spend More on Mental Health?

The World Health Organization (WHO) notes that mental illness accounts for 13% of the world’s disease burden, yet most countries under invest despite the social and economic costs of mental illness. It has been suggested that this lack of investment may be a result of stigma. A number of high income countries invest 10% or more in their mental health services. Although Canada is a high income country, its mental health spending is 7.2% according to the WHO Mental Health Atlas. This article will review the factors influencing Canada and its provinces’ under investment in mental health, compare its performance with other countries and make the case on why and how this could change.

Development and Evaluation of a Multiplex PCR for the Detection of Campylobacter concisus and Other Campylobacter spp. from Gastroenteritis Cases

We developed and evaluated a multiplex PCR (m-PCR) for application in routine diagnostic laboratories to detect Campylobacter spp. in stool samples including C. concisus, C. jejuni, and C. coli. When this m-PCR was applied on spiked faecal samples, C. concisus, C. jejuni, and C. coli were specifically identified at 105 cells/gm of faeces. To compare the sensitivity of the m-PCR with conventional culture techniques, the same spiked stool samples were cultured on an antibiotic free Columbia blood agar using the filtration technique. The detection limit of conventional culture method was 105 cells/gm of stool for C. concisus and 106 cells/gm of stool for C. jejuni and C. coli. The m-PCR was applied to test 127 faecal samples from children with gastroenteritis and the results were compared with the conventional bacterial cultures data. By this m-PCR technique, C. jejuni was detected in 7 samples, C. coli in 2 samples, and C. concisus in 7 samples. However, the conventional culture results for these samples were 6 for C. jejuni, 2 for C. coli and only one sample was positive for C. concisus. In total, 19 samples were positive for Campylobacter spp. by m-PCR while only 9 samples were positive for Campylobacter spp. by culture. In conclusion, m-PCR is more sensitive than the culture technique to detect C. concisus and other fastidious campylobacters in faeces.
