EGFR and HER2 expression in advanced biliary tract cancer

AIM:To analyze the pathogenetic role and potential clinical usefulness of the epidermal growth factor receptor(EGFR)and the human epidermal growth factor receptor 2(HER2)in patients with advanced biliary tract cancer(BTC). METHODS:EGFR and HER2 expression was studied in biopsy samples from 124 patients(51%women; median age 64.8 years),with advanced BTC diagnosed between 1997 and *** micrometers sections of paraffin embedded tissue were examined by standard, FDA approved *** with scores of 2+or 3+for HER2 expression on immunochemistry were additionally tested for HER2 gene amplification by fluorescence in situ hybridisation(FISH).RESULTS:34/124 patients(27.4%)had gallbladder cancer,47(37.9%)had intrahepatic BTC and 43(34.7%)had extrahepatic or perihilar *** expression was examined in a subset of 56 samples. EGFR expression was absent in 22/56 tumors(39.3%). Of the remaining samples expression was scored as 1+in 12(21.5%),2+in 13(23.2%)and 3+in 9(16%), ***2 expression was as follows:score 0 73/124(58.8%),score 1+27/124(21.8%),score 2+ 21/124(17%)and score 3+4/124(3.2%).HER2 gene amplification was present in 6/124,resulting in an overall amplification rate of 5%. CONCLUSION:Our data suggest that routine testing and therapeutic targeting of HER2 does not seem to be useful in patients with BTC,while targeting EGFR may be promising.

Comprehensive Transcriptome Analysis of Auxin Responses in Arabidopsis

In plants, the hormone auxin shapes gene expression to regulate growth and development. Despite the detailed characterization of auxin-inducible genes, a comprehensive overview of the temporal and spatial dynamics of auxinregulated gene expression is lacking. Here, we analyze transcriptome data from many publicly available Arabidopsis profiling experiments and assess tissue-specific gene expression both in response to auxin concentration and exposure time and in relation to other plant growth regulators. Our analysis shows that the primary response to auxin over a wide range of auxin application conditions and in specific tissues comprises almost exclusively the up-regulation of genes and iden- tifies the most robust auxin marker genes. Tissue-specific auxin responses correlate with differential expression of Aux/ IAA genes and the subsequent regulation of context- and sequence-specific patterns of gene expression. Changes in transcript levels were consistent with a distinct sequence of conjugation, increased transport capacity and down-regulation of biosynthesis in the temperance of high cellular auxin concentrations. Our data show that auxin regulates genes associated with the biosynthesis, catabolism and signaling pathways of other phytohormones. We present a transcriptional overview of the auxin response. Specific interactions between auxin and other phytohormones are highlighted, particularly the regulation of their metabolism. Our analysis provides a roadmap for auxin-dependent processes that underpins the concept of an ＇auxin code＇-a tissue-specific fingerprint of gene expression that initiates specific developmental processes.

PCH1 and PCHL Directly Interact with PIF1,Promote Its Degradation,and Inhibit Its Transcriptional Function during Photomorphogenesis

Impact of remote ischemic preconditioning on wound healing in small bowel anastomoses

AIM： To investigate the influence of remote ischemic preconditioning （RIPC） on anastomotic integrity. METHODS： Sixty male Wistar rats were randomized to six groups. The control group （n = 10） had an end- to-end ileal anastomosis without RIPC. The preconditioned groups （n = 34） varied in time of ischemia and time of reperfusion. One group received the amino acid L-arginine before constructing the anastomosis （n = 9）. On postoperative day 4, the rats were re-laparotomized, and bursting pressure, hydroxyproline concentration, intra-abdominal adhesions, and a histological score concerning the mucosat ischemic injury were collected. The data are given as median （range）.RESULTS： On postoperative day 4, median bursting pressure was 124 mmHg （60-146 mmHg） in the control group. The experimental groups did not show a statistically significant difference （P 〉 0.05）. Regarding the hydroxyproline concentration, we did not find any significant variation in the experimental groups. We detected significantly less mucosal injury in the RIPC groups. Furthermore, we assessed more extensive intra-abdominal adhesions in the preconditioned groups than in the control group. CONCLUSION： RIPC directly before performing small bowel anastomosis does not affect anastomotic stability in the early period, as seen in ischemic preconditioning.

Above-ground woody biomass allocation and within tree carbon and nutrient distribution of wild cherry(Prunus avium L.)-a case study

Background： The global search for new ways to sequester carbon has already reached agricultural lands. Such land constitutes a major potential carbon sink. The production of high value timber within agroforestry systems can facilitate an in-situ carbon storage function. This is followed by a potential long term ex- situ carbon sinkwithin long lasting products such as veneer and furniture. For this purpose wild cherry （Prunus avium L.） is an interesting option for middle Europe, yielding high prices on the timber market. Methods： A total number of 39 wild cherry were sampled in 2012 and 2013 to assess the leafless above ground biomass. The complete trees including stem and branches were separated into 1 cm diameter classes. Wood and bark from sub-samples were analysed separately and nutrient content was derived. Models for biomass estimation were constructed for all tree compartments. Results： The smallest diameter classes possess the highest proportion of bark due to smaller cross sectional area. Tree boles with a greater amount of stem wood above 10 cm in diameter will have a more constant bark proportion. Total branch bark proportion also remains relatively constant above *** measurements of 8 cm. A balance is evident between the production of new branches with a low diameter and high bark proportion offset by the thickening and a relative reduction in bark proportion in larger branches. The results show that a single tree with an age of 17 and 18 years can store up to 85 kg of carbon within the aboveground biomass portion, an amount that will increase as the tree matures. Branches display greater nutrient content than stem sections per volume unit which can be attributed to a greater bark proportion. Conclusions： Using the derived models the carbon and the nutrient content of above-ground woody biomass of whole trees can be calculated. Modelling stratified forest attributes using optical/LiDAR features in a central European landscape

The transjugular intrahepatic portosystemic shunt: Smaller stent diameters are required to optimize pressure response

Background and aims:The present treatment goal of the transjugular intrahepatic portosystemic shunt(TIPS)is a portosystemic pressure gradient of12 mmHg or its reduction by>50%.This study relates the stent diameter to the reduction of the pressure gradient and attempts to predict the appropriate stent diameter necessary to reach the treatment ***:Pressure response,super response,and poor response were investigated in 208 de-novo TIPS patients and defined as post-TIPS gradients between>6 and 12 mmHg,6 mmHg,or not reaching the goal(>12 mmHg,reduction<50%),*** were related to the smallest stent diameters measured by planimetry of the radiographic ***:Responders(65%),super responders(26%),or poor responders(9%)had comparable stent diameters of 7.21.0 mm,but different post-TIPS gradients(9.71.9 mmHg,4.51.5 mmHg,and 14.21.4 mmHg,p<0.001),relative reduction of pre-TIPS gradients(51.711.4%,73.611.1%,and 34.09.1%,p<0.001),and specific reduction per mm of stent diameter(7.52.0%/mm,10.12.0%/mm,and 4.81.4%/mm,p<0.001).Prediction of the stent diameter required to reach response was not *** two super responders had a stent diameter of<6 *** and poor responders differed by the increase in the right atrial pressure(+5.0 mmHg vs.+3.1 mmHg,p=0.026)and reduction in the portal vein pressure(-8.6 mmHg vs.-4.6 mmHg,p<0.001).Conclusion:Most patients reached the treatment goal with stent diameters of<8 ***(super response,gradient ≤6 mmHg)can be prevented by stent diameters as small as 6 *** individual response was not related to the stent diameter and not *** dysfunction may play an important role by its effect on the right atrial(preload)and portal pressure(afterload).
