Belonechitina capitata, a typically middle to late Ordovician chitinozoan index taxon was for the first time recovered from the northeastern Kumaon region, a part of Garhwal-Kumaon Tethys basin of the Himalaya, India. This species is of great biostratigraphic importance and has already been reported from Avalonia, Baltica and northern Gondwana. The study area was during Ordovician, part of a lowpalaeolatitudinal Gondwana region. The vesicles of recovered forms are black and fragmentary. This is principally attributed to intense tectonic activity during the Himalayan orogenic movement which resulted into high thermal alteration. The chitinozoans are found along with melanosclerites.

In the present study,we redescribed Gymnodinitm renplendens through incubation of process bearing cysts extracted from sediment collected in the northern Gulf of *** morphology and eltrastructure of the motile stage and cyst stage were examined using light microscopy,scanning election microacopy,and transmission electron microscopy and this revealed that the species should be transferred to the genus *** genus differs from other gymnodinioid genera in posaeaaing a Smurf-cap apical structure complex(ASC)and currenfly encompasses only one species,Barrufeta *** shows a Smurf-cap ASC that consists of three rows of elongated vesicles with small knobs in the middle *** is very similar to *** in cell morphology,but can be separated using the ultrastructure such as the shape and location of nucleus and pyrenoids,which highlights the importance of ultrastructure at inter-specific level in the genus *** unique cysts of *** are brown and process bearing,and have a tremic archeopyle with a zigrag margin on the dorsal side of the epicyat,and not polar as in cysts of *** cysts do not survive the palynological treatment used here and probably have a wide ***-likelihood and Bayesian inference were carried out based on partial large subunit ribosomal DNA(LSU rDNA)*** phylogeny supports that thegenus Barrafetu is monophyletic,and that the genus Gymnodinitm is *** results suggest that details of the ASC together with ultrastructure are potential features to subdivide the genus Gymnodinium.

Mammalian hair, as a key feature related to endothermy, physical protection, and camouflage, has a long geohistorical record that dates back even to the Mesozoic. However, the limited information on the taphonomy of hair hinders the understanding of its evolutionary significance in deep time. To address this, we conducted taphonomic experiments to simulate the transformation process of black hair. The changes in the morphological structure, microscopic characteristics, and chemical composition of hair during the simulated fossilization process were documented by scanning electron microscopy, Fourier transform infrared(FTIR) spectroscopy, and Raman spectroscopy. Our results revealed that damage to the morphology of hair began at a mild temperature(150–175°C), and hair transformed into a fluid-like mass at a relatively high temperature(200°C). In addition, the morphological structure of melanosomes had relatively greater preservation potential than keratins did, and the microscopic features of melanosomes were persistently stable. The Raman and FTIR spectra indicated that the characteristic melanin and keratin peaks changed as the temperature increased. Although the chemical signals were not lost completely, the variations in the characteristic amide bands in the FTIR spectra indicate that the keratins severely degraded at high temperatures(>200°C), whereas the characteristic melanin bands in the Raman spectra suggested that melanin signals were still recognizable, even at high temperatures(>200°C). These results were confirmed by principal component analysis(PCA) and principal component analysis linear discriminant analysis(PCA–LDA). In addition, the Raman signals of all the samples in the simulated experiments were distinguished from the signals of common geological carbonaceous materials(humic acid), which suggested that Raman spectroscopy can be used to detect melanin in fossils.

Efficient and safe cell engineering by transfection of nucleic acids remains one of the long-standing hurdles for fundamental biomedical research and many new therapeutic applications,such as CAR T cell-based *** has recently gained increasing attention as a more safe and versatile alternative tool over viral-or DNA transposon-based approaches for the generation of adoptive T ***,limitations associated with existing nonviral mRNA delivery approaches hamper progress on genetic engineering of these hard-to-transfect immune *** this study,we demonstrate that gold nanoparticle-mediated vapor nanobubble(VNB)photoporation is a promising upcoming physical transfection method capable of delivering mRNA in both adherent and suspension *** transfection experiments on HeLa cells showed the importance of transfection buffer and cargo concentration,while the technology was furthermore shown to be effective for mRNA delivery in Jurkat T cells with transfection efficiencies up to 45%.Importantly,compared to electroporation,which is the reference technology for nonviral transfection of T cells,a fivefold increase in the number of transfected viable Jurkat T cells was ***,our results point toward the use of VNB photoporation as a more gentle and efficient technology for intracellular mRNA delivery in adherent and suspension cells,with promising potential for the future engineering of cells in therapeutic and fundamental research applications.

Biallelic loss-of-function variants in ATM (Ataxia Telangiectasia Mutated) cause Ataxia Telangiectasia (AT), a rare disorder associated with cerebellar degeneration and ataxia, cancer predisposition, infertility, growth retardation, etc. ATM is a phosphoinositide 3-kinase-related kinase (PIKK) with a role in DNA repair and maintenance of genome stability. Studying a multisystem genetic disease like AT requires animal models to ascertain its pathogenesis at the level of tissues, organs and the organism. Due to its small size, cheap maintenance, large progeny, rapid development and initial transparency, zebrafish (Danio rerio) is an increasingly popular vertebrate model organism, suitable for genetic modifications and large-scale in vivo therapeutic screens as embryos are chemically permeable to small compounds. Currently, no zebrafish model for AT exists.1 We generated atm knock-outs through CRIPSR-Cas9 mutagenesis. We show that atm conserved its function as a tumour suppressor gene and is involved in gametogenesis and fertility. Therefore, this mutant is of great value for further studies investigating the role of atm in reproduction and tumorigenesis.

The Carboniferous period lasted about 60 Myr, from ～358.9 Ma to ～298.9 Ma. According to the International Commission on Stratigraphy, the Carboniferous System is subdivided into two subsystems, i.e., Mississippian and Pennsylvanian, including 6 series and 7 stages. The Global Stratotype Sections and Points(GSSPs) of three stages have been ratified, the Tournaisian, Visean, and Bashkirian stages. The GSSPs of the remaining four stages(i.e., the Serpukhovian, Moscovian,Kasimovian, and Gzhelian) have not been ratified so far. This paper outlines Carboniferous stratigraphic subdivision and correlation on the basis of detailed biostratigraphy mainly from South China, and summarizes the Carboniferous chronostratigraphic framework of China. High-resolution biostratigraphic study reveals 37 conodont zones, 24 foraminiferal(including fusulinid) zones, 13 ammonoid zones, 10 brachiopod zones, and 10 rugose coral zones in the Carboniferous of China. The biostratigraphic framework based on these biozones warrants the precise correlation of regional stratigraphy of China(including2 subsystems, 4 series, and 8 stages) to that of the other regions globally. Meanwhile, the Carboniferous chemo-, sequence-,cyclo-, and event-stratigraphy of China have been intensively studied and can also be correlated worldwide. Future studies on the Carboniferous in China should focus on(1) the correlation between shallow-and deep-water facies and between marine and continental facies,(2) high-resolution astronomical cyclostratigraphy, and(3) paleoenvironment and paleoclimate analysis based on geochemical proxies such as strontium and oxygen isotopes, as well as stomatal indices of fossil plants.

The European Society for Organ Transplantation(ESOT)has created a consensus platform for evidence-based and best-practice *** the 2022 ESOT consensus conference,nine topics were chosen for further exploration based on their potential for impact on healthcare,existing research gaps,and incomplete coverage of the topic in current scientific literature(1).Among these topics,molecular biology testing for non-invasive diagnosis of allograft rejection was a focal point,as detailed in the ESOT consensus statement on biomarkers in liver transplantation(LT)(2).We extend our congratulations to the authors of the liver subgroup on their dedicated effort and rigorous approach in translating major unmet clinical needs into four well-defined questions of interest regarding biomarkers for prediction and/or diagnosis of longer-term complications after LT.

Boundary layer influence on ship model tests in extremely shallow and confined water

Ships sailing in shallow and/or confined water(when calling a harbour or other berthing areas),will experience a different behaviour due to the interaction with vertical and/or horizontal *** other hydrodynamic changes induced in confined water,the lateral ship-bank interaction force changes its sign at a critical distance between ship and bank or ***,this distance and its effects on model test results have not been quantified in the *** investigate the shallow water hydrodynamics coupled with bank effects,systematic model tests were carried out at Flanders Hydraulics research(FHR)with different ship *** following parameters were systematically varied:water depth,lateral position,speed,and propeller *** change of the ship-bank induced lateral force from an attraction force in medium-deep and shallow water to a repulsion force in extremely shallow water conditions,can be ascribed to the interaction of the boundary layers of the ship model and the environment(tank and installed banks).In this article,a mathematical model is proposed for the critical distance in terms of boundary layer influence *** indicates the range where the model tests are influenced by the horizontal or vertical restrictions combined with the propeller’s dynamic ***,the expression has also been extended to describe the relationship between full-scale ship length and water depth with the boundary layer influence *** to lower Reynolds numbers and relatively thicker boundary layers at model scale,upscaling of the model test results,according to Froude’s law,may provide erroneous *** influence of the boundary layer initiates at a relatively higher under keel clearance(UKC)for a smaller ship model compared with a larger ship ***,the boundary layer’s influence with respect to ship model length should be considered during model testing.
