Dear Editor,Epigenetic programming plays a critical role in response to environmental fluctuations in eukaryotes (Jaenisch and Bird,2003). From initial contact to successful colonization, pathogenic microorganisms adeptly adapt to environmental changes by orchestrating a cascade of gene expression. However, how plant pathogens adapt to the host environment during early infection at the epigenetic level is largely elusive.

＜正＞The National Academies of Sciences 2007 report,"Toxicity Testing in the 21st Century:A Vision and A Strategy" envisions toxicity testing using human cells in vitro to evaluate perturbations of cellular response networks and coupling results from these assays with computational systems biology and in vitro-in vivo extrapolations （IVIVE） to complete human safety assessments. To support this transition in toxicity testing,our laboratory is evaluating key cell response networks to develop proof of concept safety assessments for a series of prototype compounds and networks,including the DNA damage response *** DNA damage toxicity pathway project studies the p53-mediated DNA damage stress response in human cells to determine the underlying response circuitry for the p53 pathway and the dose response behavior for pathway activation after chemical induced DNA damage. This research effort has three overarching goals:（1） map the key determinants of cellular fate following DNA damage induced by chemicals with different mechanisms of action（indirect vs. direct DNA-damage）,（2） identify dose-dependent thresholds associated with cellular adaptation and toxicity（mutation） and（3） perform a risk assessment for a prototype chemical based on predicted regions of safety determined from in vitro *** work involved validation of the cell model and collection of a dense data stream comprised of dose response data at the gene, protein（p53,p-p53,p-H2AX,MDM2,p21, WIP1）,and cellular（cell cycle arrest,apoptosis, micronucleus;MN）level using prototype chemicals for DNA damage:etoposide（topoisomeraseⅡinhibition）,methylmethane sulfonate（DNA alkylation） and quercetin（oxidative damage）.18 point dose-response curves were generated for protein and cell fate endpoints in a p53 competent cell line（HT1080）using flow cytometry and high content *** genome transcriptomic analysis was also performed for each prototype chemical at several doses ranging from concentrations with no e

Laboratory animals maintained on a reduced calorie but nutritionally adequate diet have extended life spans and lowered incidences of spontaneous and chemically induced cancers compared to ad libitum- fed counterparts. Many of the effects of dietary restriction on laboratory animals have been suggested to be related to a deceleration of the aging process. The inhibition of age-related changes in xenobiotic metabolizing enzyme activities by dietary restriction has previously been reported. Alterations of these enzyme activities may cause changes in metabolic activation of carcinogens and, therefore, carcinogen-DNA binding. DNA-repair capability has also been reported to be enhanced in diet-restricted rats. Using AFB 1 as a model carcinogen, we have studied in vivo and in vitro hepatic AFB 1-DNA binding, demonstrating that dietary restriction (60% of ad libitum consumption) may decrease the metabolic activation of AFB1, and subsequently reduce AFBI-DNA binding. Objective. - This 1- year, open- label, multicenter study was designed to assess the long- term tolerability and efficacy of sumatriptan nasal spray 20 mg in adolescent patients with migraine. Methods. - A prospective, multicenter, open- label study was conducted in patients aged 12 to 17 years who were allowed to treat an unlimited number of migraines at severe, moderate, or mild pain intensity with sumatriptan nasal spray for up to 1 year. All patients started the study at the 20- mg dose of sumatriptan nasal spray. Dose could be adjusted downward to 5 mg at the discretion of the investigator to optimize therapy. Results. - A total of 484 adolescent migraineurs treated 4676 migraines with sumatriptan nasal spray 20 mg (3593 during the first 6 months and 1083 during the second 6 months). A total of 3940 migraines and 699 migraines were treated with one and two 20- mg doses of sumatriptan nasal spray, respectively. Only 10 patients (treating 42 migraines) took the 5- mg dose of sumatriptan nasal spray. The overall percentage of migraines treated with either one 20- mg dose or one, two, or three 20- mg doses with at least 1 drug- related adverse event was 19% . The most common specific drug- related adverse event was unpleasant taste, reported in 17% of migraines. No other single drug- related adverse event was reported in more than 1% of migraines over the 1- year treatment period. When unpleasant taste was excluded from the adverse- event tabulations, the percentages of migraines with at least 1 drug- related adverse event after one or one, two, or three 20- mg doses declined to 4% and 3% , respectively. No patient experienced any drug- related changes in 12- lead ECGs, vital signs, or nasal assessments; and no clinically meaningful changes in clinical laboratory values were observed. Across all migraines with evaluable efficacy data (n = 4334), headache relief was reported in 43% of migraines at 1 hour and in 59% at 2 hours after dosing with sumatriptan nasal spra

Introduction:A fundamental challenge to the integrity of tropical dry forest ecosystems is the invasion of nonnative grass *** grasses compete for resources and fuel anthropogenic *** 2012,a bulldozer from the Puerto Rico Electric Power Authority cleared a 570-m trail from a state road into a mature dry forest section of Guánica Forest to control a *** monitored colonization by a non-native invasive grass(Megathyrsus maximus),a highly invasive tree(Leucaena leucocephala),and a native grass(Uniola virgata),as well as natural regeneration,along the bulldozer *** determined whether bulldozing facilitated colonization by these species into the forest and the extent of ***:Distance from propagule source and temporal variations strongly influenced colonization by our three focal *** maximus invaded along the trail from source populations by the state *** establishment of new colonies of *** seedlings went as far as 570 m inside the forest(i.e.,at the end of the bulldozer trail),but we found most new colonies within 270 m of the *** leucocephala exhibited a similar spreading *** disturbance,Uniola virgata was distributed widely across the forest,but the highest densities were found in areas near the latter portion(>401 m)of the bulldozer ***,the species formed new clumps along more than half of the trail(250 to 570 m),apparently colonizing from undisturbed patches ***:Bulldozing facilitated the invasion of non-native *** projected community assemblage will be more fire-prone than before since *** carries fire across the landscape better than ***,emphasizing the capacity of invasive plant colonization to alter local ecological processes after only a single wildfire and bulldoze *** results provide a valuable baseline for short-term vegetation response to anthropogenic disturbances in tropical semi-deciduous dry forests.

Objective： To study the precise role of DOR in the regulation of sodium channels at present. Methods： With Xenopus oocytes co-expressing sodium channel subtype 2 （Nav1.2） and DOR. Results： 1） Nav1.2 expression induced tetrodotoxin-sensitive inward currents; 2） DOR expression reduced the inward currents; 3） activation of DOR reduced the amplitude of the current and rightly shifted the activation curve of the current in the oocytes with both Nav1.2 and DOR, but not in ones with Nav1.2 alone; 4） the DOR agonist-induced inhibition of Nay 1.2 currents was in a dose-dependent manner and saturable; 5） the DOR agonist had no effect on naive oocytes. Conclusion： These data represent the first demonstration that activation of DOR inhibits Na^＋ channel function by decreasing the amplitude of sodium currents and increasing its threshold of activation. This novel finding has far-reaching impacts on novel solutions of certain neurological disorders such as hypoxic/ischemic injury, epilepsy and pain. Also, our data may improve the understanding of the mechanisms underlying acupuncture since acupuncture is known to activate the brain opioid system.

Objective: The purpose of this study was to investigate whether switching HIV-infected patients stabilized on Trizivir (abacavir 300 mg/lamivudine 150 mg/zidovudine 300 mg) plus lopinavir/ritonavir 400 mg/100mg twice daily to Trizivir alone affects clinical efficacy and tolerability. Methods: This phase 4, open-label, pilot study was conducted over 96 weeks in 23 antiretroviral-na?ve, HIV-infected patients. Initially, these patients received induction therapy with Trizivir plus lopinavir/ritonavir 400 mg/100mg twice daily. Patients who achieved a viral load 3. Nineteen patients completed induction;of the four who did not, three were lost to follow-up and one withdrew due to gastrointestinal adverse events. In 14 induction completers who had viral load measurements taken at week 48, intent-to-treat: observed analysis showed a week 48 viral load 3 higher than the baseline count. Twelve patients completed the subsequent 48-week Trizivir-alone maintenance phase, of whom 11 (92%) achieved viral loads of both 3 above baseline. Trizivir-only maintenance was associated with fewer adverse events than the Trizivir-lopinavir/ritonavir induction phase and with improvement in total cholesterol, LDL-cholesterol, and triglycerides. Conclusions: Trizivir-alone maintenance after Trizivir-lopinavir/ritonavir induction maintained virologic and CD4+ cell response, and was associated with an improved adverse event and lipid profile.

The solvent selection methodology developed earlier by. Gani （tal. （Comp. Chem. Eng., 2005）has been extended to handle multi-step reaction systems. The solvent selection problem was formulated based on the methodology guidelines, and solved using ICAS software tool. A list with solvent candidates is generated so that it can be further investigated experimentally. Comments and clarifications from chemists have been incorporated into the problem forrnulations to clarify the role of the solvents in the chemistry and potential reactivity issues. Highly promising results were obtained, in accordance with. industrial process data.
