The introduction of alleles into commercial crop breeding pipelines is both time consuming and *** technologies that are disrupting traditional breeding processes are doubled haploid(DH)breeding and genome editing(GE).Recently,these techniques were combined into a GE trait delivery system called HI-Edit(Haploid Inducer-Edit).In HI-Edit,the pollen of a haploid inducer line is reprogrammed to deliver GE traits to any variety,obviating recurrent *** HI-Edit to operate at scale,an efficient transformable HI line is needed,but most maize varieties are recalcitrant to transformation,and haploid inducers are especially difficult to transform given their aberrant reproductive *** marker assisted selection and a three-tiered testing scheme,we report the development of new Iodent and Stiff Stalk maize germplasm that are transformable,have high haploid induction rates,and exhibit a robust,genetically-dominant anthocyanin native trait that may be used for rapid haploid *** show that transformation of these elite‘‘HI-Edit”lines is enhanced using the BABYBOOM and WUSCHEL morphogenetic ***,we evaluate the HI-Edit performance of one of the lines against both Stiff Stalk and non-Stiff Stalk *** strategy and results of this study should facilitate the development of commercially scalable HI-Edit systems in diverse crops.

Host-induced gene silencing of the Verticillium dahliae thiamine transporter protein gene(VdThit)confers resistance to Verticillium wilt in cotton

Verticillium wilt(VW),induced by the soil-borne fungus Verticillium dahliae(Vd),poses a substantial threat to a diverse array of plant *** molecular breeding technology for the development of cotton varieties with heightened resistance to VW stands out as one of the most efficacious protective *** this study,we successfully generated two stable transgenic lines of cotton(Gossypium hirsutum L.),VdThitRNAi-1 and VdThit-RNAi-2,using host-induced gene silencing(HIGS)technology to introduce double-stranded RNA(dsRNA)targeting the thiamine transporter protein gene(VdThit).Southern blot analysis confirmed the presence of a single-copy insertion in each *** examination showed marked reductions in the colonization and spread of Vd-mCherry in the roots of VdThit-RNAi cotton compared to wild type(WT).The corresponding disease index and fungal biomass of VdThit-RNAi-1/2 also exhibited significant ***-time quantitative PCR(qRT-PCR)analysis demonstrated a substantial inhibition of VdThit expression following prolonged inoculation of VdThit-RNAi *** RNA sequencing(sRNA-Seq)analysis revealed the generation of a substantial number of VdThit-specific siRNAs in the VdThit-RNAi transgenic ***,the silencing of VdThit by the siVdThit produced by VdThit-RNAi-1/2 resulted in the elevated expression of multiple genes involved in the thiamine biosynthesis pathway in *** field conditions,VdThit-RNAi transgenic cotton exhibited significantly enhanced disease resistance and yield compared with *** summary,our findings underscore the efficacy of HIGS targeting VdThit in restraining the infection and spread of Vd in cotton,thereby potentially enabling the development of cotton breeding as a promising strategy for managing VW.

Using Aspen HYSYS to Develop a Custom Molecule-based Reformer Model

A new continuous catalytic reforming model was configured by using a molecule-based reactor module. Themodel was based on the Sinopec research Institute of Petroleum Processing Co., Ltd. continuous catalytic reformer fullmodel, and was reduced to a size of 157 naphtha molecules (C1−C12) that underwent 764 reactions. The new model inheritedthe advantages of the original model, and had better solving performance and flexibility owing to support by the AspenHYSYS environment. Typical commercial plant data were selected for model validation, which showed advantages in theaccuracy of detailed predictions and the range of its application. In addition, the solving time was reduced from minutes toseconds. Therefore, the simplified model proved to be feasible for industrial application.

Variation in Yellow Root Cassava (Manihot esculentus Crantz) Genotypes and Phenotypic Relationship for Selected Postharvest and Morphological Traits

This study evaluated the variation in yellow root cassava (Manihot esculentus Crantz) genotypes and phenotypic relationship for selected postharvest and morphological traits. The trial was established at the Njala Agricultural research Centre experimental site, Njala, during 2017/2018 cropping season in a randomized complete block design with three replications. Findings showed that the higher the total carotene content (TCC) in yellow flesh cassava genotypes, the longer the rate of postharvest physiological deterioration (PPD). Genotypes TR-0051-TCC/17 and TR-0012-TCC/17 recorded higher TCC (18.9 µg/g and 13.6 µg/g) and longer rate of PPD (4.29 and 3.14), respectively. Genotypes TR-0051-TCC/17, TR-0016-TCC/17, TR-0028-TCC/17, TR-0012-TCC/17 and TR-0020-TCC/17 had the highest TCC values of 18.9 µg/g, 16.09 µg/g, 14.72 µg/g, 13.6 µg/g and 11.23 µg/g with corresponding higher color chart values of 6, 6, 6, 5, and 6, respectively. This suggests the direct dependence of TCC on the root parenchyma color intensity in yellow flesh cassava genotypes. Findings also show a direct relationship between morphological and postharvest traits in yellow flesh cassava genotypes that could be exploited for the genetic improvement of cassava for increased shelf life, nutrition and related quality traits, as well as conservation and utilization of the crop.

Derivation of a Revised Tsiolkovsky Rocket Equation That Predicts Combustion Oscillations

Our study identifies a subtle deviation from Newton’s third law in the derivation of the ideal rocket equation, also known as the Tsiolkovsky Rocket Equation (TRE). TRE can be derived using a 1D elastic collision model of the momentum exchange between the differential propellant mass element (dm) and the rocket final mass (m1), in which dm initially travels forward to collide with m1 and rebounds to exit through the exhaust nozzle with a velocity that is known as the effective exhaust velocity ve. We observe that such a model does not explain how dm was able to acquire its initial forward velocity without the support of a reactive mass traveling in the opposite direction. We show instead that the initial kinetic energy of dm is generated from dm itself by a process of self-combustion and expansion. In our ideal rocket with a single particle dm confined inside a hollow tube with one closed end, we show that the process of self-combustion and expansion of dm will result in a pair of differential particles each with a mass dm/2, and each traveling away from one another along the tube axis, from the center of combustion. These two identical particles represent the active and reactive sub-components of dm, co-generated in compliance with Newton’s third law of equal action and reaction. Building on this model, we derive a linear momentum ODE of the system, the solution of which yields what we call the Revised Tsiolkovsky Rocket Equation (RTRE). We show that RTRE has a mathematical form that is similar to TRE, with the exception of the effective exhaust velocity (ve) term. The ve term in TRE is replaced in RTRE by the average of two distinct exhaust velocities that we refer to as fast-jet, vx1, and slow-jet, vx2. These two velocities correspond, respectively, to the velocities of the detonation pressure wave that is vectored directly towards the exhaust nozzle, and the retonation wave that is initially vectored in the direction of rocket propagation,
