Compared to small molecule process analytical technology （PAT） applications, biotechnology product PAT applications have certain unique challenges and opportunities. Understanding process dynamics of bioreactor cell culture process is essential to establish an appropriate process control strategy for biotechnology product PAT applications. Inline spectroscopic techniques for real time monitoring of bioreactor cell culture process have the distinct potential to develop PAT approaches in manufac- turing biotechnology drug products. However, the use of inline Fourier transform infrared （FTIR） spectroscopic techniques for bioreactor cell culture process monitoring has not been reported. In this work, real time inline FTIR Spectroscopy was applied to a lab scale bioreactor mAb IgG3 cell culture fluid biomolecular dynamic model. The technical feasibility of using FTIR Spectroscopy for real time tracking and monitoring four key cell culture metabolites （including glucose, glutamine, lactate, and ammonia） and protein yield at increasing levels of complexity （simple binary system, fully formulated media, actual bioreactor cell culture process） was evaluated via a stepwise approach. The FTIR fingerprints of the key metabolites were identified. The multivariate partial least squares （PLS） calibration models were established to correlate the process FTIR spectra with the concentrations of key metabolites and protein yield of in-process samples, either individually for each metabolite and protein or globally for all four metabolites simultaneously. Applying the 2＇ld derivative pre-processing algorithm to the FTIR spectra helps to reduce the number of PLS latent variables needed significantly and thus simplify the interpretation of the PLS models. The validated PLS models show promise in predicting the concentration profiles of glucose, glutamine, lactate, and ammonia and protein yield over the course of the bioreactor cell culture process. Ozone Profile Retrieval from Satellite Observation Using High Spectral Resolution Infrared Sounding Instrument

This paper presents a preliminary result on the retrieval of atmospheric ozone profiles using an im- proved regression technique and utilizing the data from the Atmospheric InfraRed Sounder (AIRS), a hyper-spectral Instrument expected to be flown on the EOS-AQUA platform in 20D2. Simulated AIRS spectra were used to study the sensitivity of AIRS radiance on the tropospheric and stratospheric ozone changes, and to study the impact of various channel combinations on the ozone profile retrieval. Sensitivity study results indicate that the AIRS high resolution spectral channels between the wavenumber 650- 800 cm-1 provide very useful information to accurately retrieve tropospheric and stratospheric ozone pro- files Eigenvector decomposition of AIRS spectra indicate that no more than 100 eigenvectors are needed to retrieve very accurate ozone profiles. The accuracy of the retrieved atmospheric ozone profile from the pres- ent technique and utilizing the AIRS data was compared with the accuracy obtained from current Advanced TIROS Operational Vertical Sounder (ATOVS) data aboard National Oceanic and Atmospheric Admini- stration (NOAA) satellites As expected, a comparison of retrieval results confirms that the ozone profile re- trieved with the AIRS data is superior to that of ATOVS

Analysis of roof and pillar failure associated with weak floor at a limestone mine

A limestone mine in Ohio has had instability problems that have led to massive roof falls extending to the surface. This study focuses on the role that weak, moisture-sensitive floor has in the instability *** NIOSH research related to this subject did not include analysis for weak floor or weak bands and recommended that when such issues arise they should be investigated further using a more advanced analysis. Therefore, to further investigate the observed instability occurring on a large scale at the Ohio mine, FLAC3 D numerical models were employed to demonstrate the effect that a weak floor has on roof and pillar stability. This case study will provide important information to limestone mine operators regarding the impact of weak floor causing the potential for roof collapse, pillar failure, and subsequent subsidence of the ground surface.

Abstract Polyaniline film was prepared by electrochemical method in an acidic solution of aniline. The micromor- phology of the polyaniline film was transformed to three-dimensional network structure instead of little particles while the deposition time was extended. The peak wavelength of the photoluminescence spectrum was 491 nm. The luminous intensity increased with the extension of deposition time, and so did the electrochemical activity.

Derivation of Regression Coefficients for Sea Surface Temperature Retrieval over East Asia

Among the regression-based algorithms for deriving SST from satellite measurements, regionally optimized algorithms normally perform better than the corresponding global algorithm. In this paper, three algorithms are considered for SST retrieval over the East Asia region （15°-55°N, 105°-170°E）, including the multi-channel algorithm （MCSST）, the quadratic algorithm （QSST）, and the Pathfinder algorithm （PFSST）. All algorithms are derived and validated using collocated buoy and Geostationary Meteorological Satellite （GMS-5） observations from 1997 to 2001. An important part of the derivation and validation of the algorithms is the quality control procedure for the buoy SST data and an improved cloud screening method for the satellite brightness temperature measurements. The regionally optimized MCSST algorithm shows an overall improvement over the global algorithm, removing the bias of about -0.13℃ and reducing the root-mean-square difference （rmsd） from 1.36℃ to 1.26℃. The QSST is only slightly better than the MCSST. For both algorithms, a seasonal dependence of the remaining error statistics is still evident. The Pathfinder approach for deriving a season-specific set of coefficients, one for August to October and one for the rest of the year, provides the smallest rmsd overall that is also stable over time.

Corrosion behavior of 907 steel under thin electrolyte layers of artificial seawater

The corrosion behavior of 907 steel under thin electrolyte layer(TEL) has been investigated by means of cathodic polarization curve measurement, electrochemical impedance spectroscopy(EIS) and scanning electron microscopy(SEM). The results show that the cathodic diffusion current density presents the variation trend of initial increase and subsequent decrease with the decrease of TEL thickness, and the maximum deposits at 58 μm. The cotangent-hyperbolic impedance(O) is rationally first introduced to study the diffusion process of the reactants through the corrosion products layer with many permeable holes. The initial corrosion rate of 907 steel under different TEL thickness increases with the decrease of TEL thickness except that of 104 μm,whereas the corrosion rate after long time corrosion can be ranked as 104 μm﹥402 μm﹥198 μm﹥301 μm﹥bulk solution.

CD4 and CD8 T cells mediate distinct lethal meningoencephalitis in mice challenged with Tacaribe arenavirus

Neonates are at increased risk of viral encephalopathies that can result in neurological dysfunction, seizures, permanent disability and even death. The neurological damage results from the combined effect of the virus and the immune response it elicits, thus finding tools to facilitate viral clearance from central nervous system （CNS） while minimizing neuron damage remains a critical challenge. Neonatal mice inoculated intraperitoneally with Tacaribe virus （TCRV） develop seizures, hindlimb paralysis and death within 15 days of inoculation. TCRV localizes to the CNS within days of challenge, primarily infecting astrocytes in the cerebellum and brain stem. We show that infection leads to inflammation, T cell and monocyte infiltration into the cerebellar parenchyma, apoptosis of astrocytes, neuronal degeneration and loss of Purkinje cells. Infiltrating antigen-specific T cells fail to clear the virus but drive the disease, as T-cell-deficient CD3ε KO mice survive TCRV infection with minimal inflammation or clinical manifestations despite no difference in CNS viral loads in comparison with T-cell sufficient mice. CD8＋ T cells drive the pathology, which even in the absence of CD4＋ T-cell help, infiltrate the parenchyma and mediate the apoptotic loss of cerebellar astrocytes, neurodegeneration and loss of Purkinje cells resulting in loss of balance, paralysis and death. CD4＋ T cells are also pathogenic inducing gliosis and inflammation in the cerebellum and cerebrum that are associated with wasting and death several weeks after CD4＋ T-cell transfer. These data demonstrate distinct pathogenic effects of CD4＋ and CD8＋ T cells and identify them as possible therapeutic targets.

CFD modeling of methane distribution at a continuous miner face with various curtain setback distances

Knowledge of the airflow patterns and methane distributions at a continuous miner face under different ventilation conditions can minimize the risks of explosion and injury to miners by accurately forecasting potentially hazardous face methane levels. This study focused on validating a series of computational fluid dynamics(CFD) models using full-scale ventilation gallery data that assessed how curtain setback distance impacted airflow patterns and methane distributions at an empty mining face(no continuous miner present). Three CFD models of face ventilation with 4.6, 7.6 and 10.7 m(15, 25, and 35 ft) blowing curtain setback distances were constructed and validated with experimental data collected in a full-scale ventilation test facility. Good agreement was obtained between the CFD simulation results and this *** airflow and methane distribution information are provided. Elevated methane zones at the working faces were identified with the three curtain setback distances. Visualization of the setback distance impact on the face methane distribution was performed by utilizing the post-processing capability of the CFD software.

It crystallizes in monoclinic, space group C2/c with a = 21.374（14）, b = 10.430（7）, c = 10.741（7） A, β= 114.184（8）°, V= 2184（2） A^3, Z= 4, Dc = 1.573 g/cm^3, F（000） = 1064, μ（MoKα） = 1.355 mm ^-1, T= 291（2） K, the final R = 0.0548 and wR = 0.1437 for 1524 observed reflections with 1 〉 2σ（I）. In this compound, the nitrogen atom links two ferrocenyl groups and one phenyl group forming a novel imido derivative.
