The power of“controllers”:Transposon-mediated duplicated genes evolve towards neofunctionalization

Since the discovery of the first transposon by *** McClintock,the prevalence and diversity of transposable elements(TEs)have been gradually *** fundamental genetic components,TEs drive organismal evolution not only by contributing functional sequences(e.g.,regulatory elements or“controllers”as phrased by ***)but also by shuffling genomic *** the latter respect,TE-mediated gene duplications have contributed to the origination of new genes and attracted extensive *** response to the development of this field,we herein attempt to provide an overview of TEmediated duplication by focusing on common rules emerging across duplications generated by different TE ***,despite the huge divergence of transposition machinery across TEs,we identify three common features of various TE-mediated duplication mechanisms,including end bypass,template switching,and recurrent *** three features lead to one common functional outcome,namely,TE-mediated duplicates tend to be subjected to exon shuffling and ***,the intrinsic properties of the mutational mechanism constrain the evolutionary trajectories of these *** finally discuss the future of this field including an in-depth characterization of both the duplication mechanisms and functions of TE-mediated duplicates.

Dear Editor,The fall army worm,Spodoptera frugiperda(Lepidoptera:Noctuidae),a highly agricultural destructive pest of cereals,is originally rampant in tropical and subtropical America(Kenis et al.2019).The caterpillars of this moth feed on more than 350 plant species(Harrison et al.2019),including several economically important cultivated grasses such as maize,rice,sorghum and sugarcane,severely threatening the food security(Cock et al.2017).Spodoptera frugiperda is recognized to comprise two morphologically identical but genetically distinct strains with different host ***"rice strain^R strain,RS)is thought to preferentially feed on rice and various pasture grasses,while the‘com strain,(C strain,CS)on maize,cotton and sorghum(Juarez et al.2012).Unable to tolerate freezing temperature,*** is seldom reported outside of ***,since its first detection in Nigeria in 2016,several congruent reports from African countries about sudden and severe upsurges of *** raised the alarm that *** is continuously expanding their range to become truly threat(Goergen et al.2016).

The number of elderly people living with HIV is increasing globally,and the condition of this population is relatively complicated due to the dual effects of aging and HIV ***,the impact of HIV infection combined with aging on the immune homeostasis of secondary lymphoid organs remains ***,we used the simian immunodeficiency virus mac239(SIVmac239)strain to infect six young and six old chinese rhesus macaques(ChRMs)and compared the infection characteristics of the two groups in the chronic stage through multiplex immunofluorescence staining of lymph *** results showed that the SIV production and CD4/CD8 ratio inversion in old ChRMs were more severe than those in young ChRMs in both the peripheral blood and the lymph nodes,especially when a large number of CD8+T cells infiltrated the follicles and germinal ***3 in these follicular CXCR5+CD8+T cells was highly activated,with high expression of granzyme B,which might be caused by the severe inflammatory milieu in the follicles of old *** study indicates that aging may be a cofactor involved in SIV-induced immune disorders in secondary lymphoid tissues,affecting the effective antiviral activity of highly enriched follicular CXCR5+CD8+cells.

As of June 2020, Coronavirus Disease 2019(COVID-19) has killed an estimated 440 000 people worldwide, 74% of whom were aged ≥65 years,making age the most significant risk factor for death caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2) infection. To examine the effect of age on death, we established a SARSCoV-2 infection model in chinese rhesus macaques(Macaca mulatta) of varied ages. Results indicated that infected young macaques manifested impaired respiratory function, active viral replication, severe lung damage, and infiltration of CD11b^+ and CD8^+ cells in lungs at one-week post infection(wpi), but also recovered rapidly at 2 wpi. In contrast, aged macaques demonstrated delayed immune responses with a more severe cytokine storm, increased infiltration of CD11b^+ cells, and persistent infiltration of CD8^+ cells in the lungs at 2 wpi. In addition,peripheral blood T cells from aged macaques showed greater inflammation and chemotaxis, but weaker antiviral functions than that in cells from young macaques. Thus, the delayed but more severe cytokine storm and higher immune cell infiltration may explain the poorer prognosis of older aged patients suffering SARS-CoV-2 infection.

Discovery of Cosmic Spherules from the Mesoproterozoic Strata and its Significance——in Case of the Ming Tombs Area, Beijing

This study covers cosmic spherules derived from the Mesoproterozoic Dahongyu Formation in the Ming Tombs area, Beijing. The cosmic spherules include iron oxide cosmic spherules, carbonaceous chondrites, and atomic iron "steely bead"-shaped cosmic spherules. The mineral assemblage of silicon carbide, forsterite, zircon, and glass spherules and fragments were picked from melt-silicified carbonate of the Mesoproterozoic Dahongyu Formation(ca. 1625 Ma). Cosmic spherule assemblages are solely discovered from sedimentary rocks in china. Platinum group elements(PGE) were determined for the first time in cosmic spherules and associated minerals. PGE comparative observation between meteorite and cosmic spherules is presented in this study. It is recognized that an extraterrestrial meteorite impact event might have occurred in the Dahongyu Stage. The main evidence is a large number of iron cosmic spherules in silicified oncolitic limestone, and associated cosmic silicon carbide, glass spherules, and fragments, as well as the presence of forsterite. The impact-volcanic crater is characteristic of a big black shale block dropped into the bended silicified limestone.

Brassica rapa ***(turnip)is an important crop in Qinghai-Tibet Plateau(QTP)with anti-hypoxic *** is rich in glucosinolates,isothiocyanates and phenolic compounds with diverse biological activities,involving anti-oxidant,anti-tumor,anti-diabetic,anti-inflammatory,anti-microbial,hypolipidemic,cardioprotective,hepatoprotective,nephroprotective and analgesic *** this study,the ethyl acetate(EtOAc)and butanol parts of Brassica rapa were first revealed with inhibitory effects onα-glucosidase,whereas the water part was *** bioassay-guided isolation on the EtOAc and butanol parts yielded 12 compounds,involving three indole derivatives,indole-3-acetonitrile(1)4-methoxyindole-3-acetonitrile(2)and indole-3-aldehyde(3)two flavonoids,liquiritin(4)and licochalcone A(5)two phenylpropanoids,sinapic acid(6)and caffeic acid(7)two phenylethanol glycosides,2-phenylethylβ-glucopyranoside(8)and salidroside(9)and three other compounds,syringic acid(10)adenosine(11)and(3β,20E)-ergosta-5,20(22)-dien-3-ol(12)Licochalcone A(5)and caffeic acid(7)showedα-glucosidase inhibitory activity with IC 50 values of 62.4±8.0μM and 162.6±3.2μM,comparable to the positive control,acarbose(IC 50=142±0.02μM).Docking study suggested that licochalcone A(5)could well align in the active site ofα-glucosidase(docking score=-52.88)by forming hydrogen bonds(Gln1372,Asp1420,Gln1372,Arg1510),hydrophobic effects(Tyr1251,Tyr1251,Trp1355,Phe1560,Ile1587,Trp1355,Phe1559,Phe1559)andπ-πstacking interaction(Trp1355).This study provides valuable information for turnip as a new resource in searching anti-diabetic candidates.

Melanocortin-1 receptor (MC1R) plays a major role in pigmentation in many *** investigate if the MC1R gene is associated with coat color in water buffalo,the coding region of MC1R gene of 216 buffalo samples was sequenced,which included 49 black river buffalo (Murrah and Nili-Ravi),136 swamp buffalo (Dehong,Diandongnan,Dechang,Guizhou,and Xilin) with white and gray body,and 31 hybrid offspring of river buffalo Nili-Ravi (or Murrah) and swamp *** the three variation sites found,SNP684 was synonymous,while SNP310 and SNP384 were nonsynonymous,leading to p.S104G and p.I128M changes,*** Individuals carrying homozygote EBR/EBR were *** genotype and phenotype analysis of the hybrid offspring of black river buffalo and gray swamp buffalo further revealed that the river buffalo type allele EBR or the allele carrying the amino acid p.104S was important for the full function of *** in silico functional analysis showed that the amino acid substitutions p.G104S and p.M128I had significant impact on the function of *** results indicate that the allele EBR or the allele carrying the amino acid p.104S was associated with the black coat color in buffalo.

Malania oleifera(Olacaceae)is a valued tree species,mostly because its seeds have high precious fatty acid content(particularly nervonic acid).However,seedling mortality rates are often high and regeneration of this tree has been problematic,which greatly hinders its utilization at a large *** difficulties of some tree species in the family Olacaceae have been attributed to their root hemiparasitic *** by field observations and the taxonomic proximity of *** to root hemiparasites in Olacaceae,we hypothesized that tuberous structures observed on the roots of *** are parasitic organs known as *** test this hypothesis,we collected root samples from *** plants of various ages and growth conditions,investigated the morphological and anatomical features of tuberous structures and their connections to neighboring *** analyses confirmed that *** are root *** the best of our knowledge,this is the first empirical report on root hemiparasitism in *** life strategies of root hemiparasitic plants differ greatly from autotrophic plants,the root hemiparasitic habit needs to be taken into account for successful seedling regeneration of *** study establishes the foundation for investigations into a long-neglected but essential aspect in research of these highly valued tree species.

Molecular evolution of stress-response gene Leptin in high-altitude chinese snub-nosed monkeys (Rhinopithecus genus)

chinese snub-nosed monkeys (genus Rhinopithecus, subfamily Colobinae), including R. bieti, R. brelichi and R. roxellana, are well-known as the non-human primates with the highest known altitudinal distribution. They represent an interesting model organism of adaptation to the extreme environmental stresses. However, no study at the molecular level has yet been reported for the high-altitude adaptation in chinese snub-nosed monkeys. Leptin, as an adipocyte-derived hormone, is believed to play an important role in energy homeostasis in adaptation to high altitude environments. In the present study, we sequenced and compared leptin sequences of the chinese snub-nosed monkeys (R. bieti and R. roxellana) with their lowland close relative R. avunculus and other Colobines. Unexpectedly, no amino acid changes were observed in the 7 Colobinae species examined, including the 2 chinese snub-nosed monkeys, indicating no difference in the evolutionary pattern of the Leptin gene between high-altitude monkeys and their lowland counterparts. In contrast to a previous finding of adaptive evolution of Leptin gene in plateau pikas, our study suggests that this gene may not have an important role in high-altitude adaptation of chinese snub-nosed monkeys. Other nuclear genes associated with energy metabolism, or mitochondrial genes, are most likely to be involved the molecular mechanism underlying adaptation of these monkeys to cold and hypoxia associated with the highland environment.
