Roles of neuronal lysosomes in the etiology of Parkinson’s disease

Therapeutic progress in neurodegenerative conditions such as Parkinson’s disease has been hampered by a lack of detailed knowledge of its molecular *** advancements in genetics and genomics have provided fundamental insights into specific protein players and the cellular processes involved in the onset of *** this respect,the autophagy-lysosome system has emerged in recent years as a strong point of convergence for genetics,genomics,and pathologic indications,spanning both familial and idiopathic Parkinson’s ***,if not all,genes linked to familial disease are involved,in a regulatory capacity,in lysosome function(e.g.,LRRK2,alpha-synuclein,VPS35,Parkin,and PINK1).Moreover,the majority of genomic loci associated with increased risk of idiopathic Parkinson’s cluster in lysosome biology and regulation(GBA as the prime example).Lastly,neuropathologic evidence showed alterations in lysosome markers in autoptic material that,coupled to the alpha-synuclein proteinopathy that defines the disease,strongly indicate an alteration in *** this Brief Review article,I present a personal perspective on the molecular and cellular involvement of lysosome biology in Parkinson’s pathogenesis,aiming at a larger vision on the events underlying the onset of the *** attempts at targeting autophagy for therapeutic purposes in Parkinson’s have been mostly aimed at“indiscriminately”enhancing its activity to promote the degradation and elimination of aggregate protein accumulations,such as alpha-synuclein Lewy ***,this approach is based on the assumption that protein pathology is the root cause of disease,while pre-pathology and pre-degeneration dysfunctions have been largely observed in clinical and pre-clinical *** addition,it has been reported that unspecific boosting of autophagy can be ***,it is important to understand the mechanisms of specific autophagy forms and,even more,the adjustment of specific lysosome f

Effect of Air Distribution on the Transport Characteristics of Solid Particles in the Thermal Storage and Release System of Circulating Fluidized Bed

Solid particle heat storage technology offers a potential solution to the challenges posed by the significant growth of renewable energy sources,particularly in terms of grid security and ***,it has the capability to optimize the energy utilization efficiency of the power *** order to investigate the transport regulation characteristics of solid particles in the thermal storage and release system of a circulating fluidized bed(CFB),a test rig with a capacity of 0.1 MW(th)was *** rig was utilized to systematically study the transport regulation characteristics of solid particles under the double U-type valve feed structure and U-type valve discharge *** experimental findings indicate that the system's design enables efficient and rapid storage and release of solid particles in the *** air distribution mode,specifically the double U-type valve feed structure and the U-type valve discharge structure,significantly influence the feed and discharge characteristics of the ash storage *** was observed that the impact of loose air on these characteristics is more substantial than that of the return air,irrespective of the feed structure or the return *** adjusting the feed and discharge rate,it is recommended to adopt a scheme that involves coarse adjustment through loose air and fine adjustment through return air.

Intelligent responsive self-assembled micro-nanocapsules: Used to delay gel gelation time

In the application of polymer gels to profile control and water shutoff,the gelation time will directly determine whether the gel can "go further" in the formation,but the most of the methods for delaying gel gelation time are complicated or have low *** is an urgent need for an effective method for delaying gel gelation time with intelligent *** by the slow-release effect of drug capsules,this paper uses the self-assembly effect of gas-phase hydrophobic SiO2in aqueous solution as a capsule to prepare an intelligent responsive self-assembled *** capsule slowly releases the cross-linking agent under the stimulation of external conditions such as temperature and pH value,thus delaying gel gelation *** the pH value is 2 and the concentration of gas-phase hydrophobic SiO2particles is 10%,the gelation time of the capsule gel system at 30,60,90,and 120℃ is12.5,13.2,15.2,and 21.1 times longer than that of the gel system without containing capsule,*** with other methods,the yield stress of the gel without containing capsules was 78 Pa,and the yield stress after the addition of capsules was 322 *** intelligent responsive self-assembled micronanocapsules prepared by gas-phase hydrophobic silica nanoparticles can not only delay the gel gelation time,but also increase the gel *** slow release of cross-linking agent from capsule provides an effective method for prolongating the gelation time of polymer gels.

Developing a multi-method approach to identifying e-scooter hazard hotspots

In recent years, e-scooters have been introduced in many European cities. In several places we have witnessed a rapid uptake of this new mode of transport mainly as a result of public sharing schemes. A number of incidents, injuries and even fatalities have given rise to questions regarding the safety of these vehicles. These questions are being researched mainly using official crash data and data specifying injuries and hospital treatment. Until now, the research has focused on investigating typical injury patterns and estimating risk levels. Very little is known about exactly where conflicts and crashes occur. Knowledge of hazard hotspots is crucial when investigating risk levels and improving safety for all road ***, this paper develops an approach to investigating locations with potentially dangerous interactions within the active mobility system in the city of Berlin. The approach consists of explorative expert interviews, an online poll, and quantitative analyses. For the latter we combine three datasets. First, we research crash hotspots using official data. Second, we use data based on acceleration sensors from cyclists' smartphones to find locations of sudden movements. Third, we use trip data from the operators of escooter sharing systems. The information gathered is used in a conclusive expert workshop to identify hazard *** show that many of the conflicts with pedestrians are caused by parked escooters. Second, e-scooter trips are concentrated in the inner city and along specific routes. In moving traffic, various data sources are used to identify hotspots at intersections and in areas between *** present research lays the foundation for important further studies to investigate interactions at hotspots in detail by determining nine specific locations in the city of Berlin.

Comparative study between three carbonaceous nanoblades and nanodarts for antimicrobial applications

The design of nanostructured materials occupies a privileged position in the development and management of affordable and effective technology in the antibacterial ***,we discuss the antimicrobial properties of three carbonaceous nanoblades and nanodarts materials of graphene oxide(GO),reduced graphene oxide(RGO),and single-wall carbon nanotubes(SWCNTs)that have a mechano-bactericidal effect,and the ability to piercing or slicing bacterial *** demonstrate the significance of size,morphology and composition on the antibacterial activity mechanism,the designed nanomaterials have been *** minimum inhibitory concentration(MIC),standard agar well diffusion,and transmission electron microscopy were utilized to evaluate the antibacterial activity of GO,RGO,and *** on the evidence obtained,the three carbonaceous materials exhibit activity against all microbial strains tested by completely encapsulating bacterial cells and causing morphological disruption by degrading the microbial cell membrane in the order of RGO>GO>*** of the external cell wall structure and outer membrane proteins,the synthesized carbonaceous nanomaterials exhibited higher antibacterial activity against Gram-positive bacterial strains than Gram-negative and fungal *** had the lowest MIC values(0.062,0.125,and 0.25 mg/mL against ***,***,and ***,respectively),as well as minimum fungal concentrations(0.5 mg/mL for both *** and ***).At 12 hr,the cell viability values against tested microbial strains were completely *** lysis and death occurred as a result of severe membrane damage caused by microorganisms perched on RGO *** work gives an insight into the design of effective graphene-based antimicrobial materials for water treatment and remediation.

Electrolytic water splitting(EWS)is an attractive and promising technique for the production of hydrogen ***,the sluggish kinetic rate of hydrogen/oxygen evolution reactions leads to a high overpotential and low energy *** to date,Pt/Ir-based nanocatalysts have become the state-of-the-art EWS catalysts,but disadvantages such as high cost and low earth abundance greatly limit their applications in EWS *** an attractive candidate for the Pt/Ir catalysts,series of Ru-based nanomaterials have aroused much attention for their low price,Pt-like hydrogen bond strength,and high EWS *** particular,Ru-doped functional porous materials have been becoming one of the most representative EWS catalysts,which can not only achieve the dispersion and adjustment for active Ru sites,but also simultaneously solve the problems of mass transfer and catalytic conversion in *** this review,the design and preparation strategies of Ru-doped functional porous materials toward EWS in recent years are summarized,including Ru-doped metal organic frameworks(MOFs),Ru-doped porous organic polymers(POPs),and their ***,detailed structure–activity relationships induced by the tuned geometric/electronic structures of Ru-doped functional porous materials are further depicted in this *** but not least,the challenges and perspectives of Ru-doped functional porous materials catalysts are reasonably proposed to provide fresh ideas for the design of Ru-based EWS catalysts.

Nonlinear Characteristic Analysis of Gas-Interconnected Quasi-Zero Stiffness Pneumatic Suspension System:A Theoretical and Experimental Study

Because of significantly changed load and complex and variable driving road conditions of commercial vehicles,pneumatic suspension with lower natural frequencies is widely used in commercial vehicle suspension *** ever,traditional pneumatic suspension system is hardly to respond the greatly changed load of commercial vehicles To address this issue,a new Gas-Interconnected Quasi-Zero Stiffness Pneumatic Suspension(GIQZSPS)is presented in this paper to improve the vibration isolation performance of commercial vehicle suspension systems under frequent load *** new structure adds negative stiffness air chambers on traditional pneumatic suspension to reduce the natural frequency of the *** can adapt to different loads and road conditions by adjusting the solenoid valves between the negative stiffness air ***,a nonlinear mechanical model including the dimensionless stiffness characteristic and interconnected pipeline model is derived for GIQZSPS *** the nonlinear mechanical model of GIQZSPS system,the force transmissibility rate is chosen as the evaluation index to analyze ***,a testing bench simulating 1/4 GIQZSPS system is designed,and the testing analysis of the model validation and isolating performance is carried *** results show that compared to traditional pneumatic suspension,the GIQZSPS designed in the article has a lower natural *** the system can achieve better vibration isolation performance under different load states by switching the solenoid valves between air chambers.

Stability-Considered Lane Keeping Control of Commercial Vehicles Based on Improved APF Algorithm

Regarding the lane keeping system,path tracking accuracy and lateral stability at high speeds need to be taken into account especially for commercial vehicles due to the characteristics of larger mass,longer wheelbase and higher mass *** improve the performance mentioned above comprehensively,the control strategy based on improved artificial potential field(APF)algorithm is *** the paper,time to lane crossing(TLC)is introduced into the potential field function to enhance the accuracy of path tracking,meanwhile the vehicle dynamics parameters including yaw rate and lateral acceleration are chosen as the repulsive force field *** lane keeping controller based on improved APF algorithm is designed and the stability of the control system is proved based on Lyapunov *** addition,adaptive inertial weight particle swarm optimization algorithm(AIWPSO)is applied to optimize the gain of each potential field *** co-simulation results indicate that the comprehensive evaluation index respecting lane tracking accuracy and lateral stability is reduced ***,the proposed control strategy is verified by the HiL *** provides a beneficial reference for dynamics control of commercial vehicles and enriches the theoretical development and practical application of artificial potential field method in the field of intelligent driving.
