Efficacy of granulocyte-colony stimulating factor treatment in a rat model of anterior ischemic optic neuropathy

Non-arteritic anterior ischemic optic neuropathy （NA-AION） is the most common cause of acute ischemic damage to the optic nerve （ON）, and the leading cause of seriously impaired vision in people over 55 years of age. It demonstrated that subcutaneous administration of Granulocyte colony-stimulating factor （G-CSF） reduces RGC death in an ON crush model in rats, and that the neuroprotective effects may involve both anti-apoptotic and anti-inflammatory processes. Our recent work shows that the protective actions of G-CSF in rAION models may involve both anti-apoptotic and anti-inflammatory processes. However, the exact rescuing mech- anisms involved in the administration of G-CSF in rAION models need further investigation. In addition, further studies on the administration of G-CSF at different time intervals after the induction of rAION may be able to illustrate whether treatment given at a later time is still neu- roprotective. Further, it is unknown whether treatment using G-CSF combined with other drugs will result in a synergistic effect in a rAION model. Inflammation induced by ischemia plays an essential role on the ON head in NA-A1ON, which can result in disc edema and compartment changes. Therefore, it is reasonable that adding an anti-inflammatory drug may enhance the therapeutic effects of G-CSF. An ongoing goal is to evaluate the novel sites of action of both G-CSF and other anti-inflammatory drugs, and to identify the functionally protective pathways to enhance RGC survival. These investigations may open up new therapeutic avenues for the treatment of ischemic optic neuropathy.

Epigenetic regulation of genes during development: A conserved themefrom flies to mammals

Eukaryotic genome is organized in form of chromatin within the nucleus. This organization is important for compaction of DNA as well as for the proper expression of the genes. During early embryonic development, genomic packaging receives variety of signals to eventually set up cell type specific expression patterns of genes. This process of regulated chromatinization leads to ＂cell type specific epigenomes＂. The expression states attained during differentiation process need to be maintained subsequently throughout the life of the organism. Epigenetic modifications are responsible for chromatin dependent regulatory mechanism and play a key role in maintenance of the expression state-a process referred to as cellular memory. Another key feature in the packaging of the genome is formation of chromatin domains that are thought to be structural as well as functional units of the higher order chromatin organization. Boundary elements that function to define such domains set the limits of regulatory elements and that of epigenetic modifications. This connection of epigenetic modification, chromatin structure and genome organization has emerged from several studies. Hox genes are among the best studied in this context and have led to the significant understanding of the epigenetic regulation during development. Here we discuss the evolutionarily conserved features of epigenetic mechanisms emerged from studies on homeotic gene clusters.

Representations of coherent and squeezed states in an extended two-parameter Fock space

Recently an f -deformed Fock space which is spanned by ｜ n λ was introduced. These bases are the eigenstates of a deformed non-Hermitian Hamiltonian. In this contribution, we will use rather new nonorthogonal basis vectors for the construction of coherent and squeezed states, which in special case lead to the earlier known states. For this purpose, we first generalize the previously introduced Fock space spanned by ｜ n λ bases, to a new one, spanned by extended two-parameters bases ｜ n λ 1 ,λ 2 . These bases are now the eigenstates of a non-Hermitian Hamiltonian H λ 1 ,λ 2 = a λ 1 ,λ 2 a ＋ 1 2 , where a λ 1 ,λ 2 = a ＋ λ 1 a ＋ λ 2 and a are, respectively, the deformed creation and ordinary bosonic annihilation operators. The bases ｜ n λ 1 ,λ 2 are nonorthogonal （squeezed states）, but normalizable. Then, we deduce the new representations of coherent and squeezed states in our two-parameter Fock space. Finally, we discuss the quantum statistical properties, as well as the non-classical properties of the obtained states numerically.

The pathogenesis of intrahepatic cholestasis of pregnancy (ICP), a disorder that adversely affects maternal wellbeing and fetal outcome, is unclear. However, multiple factors probably interact along with a genetic predisposition. We would like to add some comments on a paper recently published concerning the role of ABCB11 and ABCC2 polymorphisms in both ICP and contraceptive-induced cholestasis, especially in the light of our recently published findings about a positive association between ICP and ABCC2 common variants.

Atherosclerotic cardiovascular disease is the main cause of mortality and morbidity in the world. Plasma levels of low density lipoprotein cholesterol （LDL-C） are positively correlated with the risk of atherosclerosis. High plasma LDL concentrations in patients with hypercholesterolemia lead to build-up of LDL in the inner walls of the arteries, which becomes oxidized and promotes the formation of foam cells, consequently initiating atherosclerosis. Plasma LDL is mainly cleared through the LDL receptor （LDLR） pathway. Mutations in the LDLR cause familiar hyperch- olesterolemia and increase the risk of premature coronary heart disease. The expression of LDLR is regulated at the transcriptional level via the sterol regulatory element binding protein 2 （SREBP-2） and at the posttranslational levels mainly through proprotein convertase subtilisin/kexin-type 9 （PCSK9） and inducible degrader of the LDLR （IDOL）. In this review, we summarize the latest advances in the studies of PCSK9.

ELECTRON THEORY ON THERMAL EXPANSION OF SOLIDS AND ITS APPLICATION TO LIGHT METALS

In first-order approximation, the interaction potential function among atoms in solidsmay be expressed aswhere g_α depends on the electron structures of solids; r_α and r_(αo) express the distancesbetweell two atoms in nonequilibrium and equilibrium respectively ; α≡A, B, C, …, de-noting the quantities related to the nearest, next-near and the third near neighbours withrespect to the reference atom, and the quantity related to the lattice electron; λ_D beingthe average Debye wavelength; n is the bond strength parameter determined by the theo-retical formula derived by the author. Using the Debye model of solids, the total ener-gy E_(Tτ) of the solid at T(K) can be obtained easily. Then the functional relation ofthe lattice constant γ_(AOT) (T) with T(K) can be found out by solving the equation V(p_α,γA) - E_(Tτ) = 0 for γ_A and that of the linear thermal expansion coefficient α(T) and T(K)can be derived further. The theory has been applied to the alkali and alkaline earthmetals and the results agree well with the experimental. In the paper, we have also calculated the Gruneisen constants using the theoreticalvalues of the linear thermal expansion coefficient α(T) and that of the bulk modulus obtain-ed previously by the author, and made their comparison with the values calculated fromthe experimental data. Furthermore, the product of the melting point T_m and α(T) approx-imates the Hindnert-Sonder rule. These results demonst rate that the new type of theinteraction potential function among atoms in a solid and the electron theory of thermalexpan sion established in the paper are practical and worth further developing.

Steroid hormones are one of the major bioactive lipid species that regulate a large number of physiological processes including developmental transitions, growth, metabolism, inflammation, immune response and reproduction in animals. In insects, steroid hormones ecdysone and 20-hydroxyecdysone (20E) trigger the major transitional events, molting and metamorphosis (Thummel, 1996). In Drosophila, ecdysone synthesis primarily occurs in the prothoracic gland, an endocrine gland expressing a large number of ecdysone biosynthetic enzymes required for a series of hydroxylation and oxidation steps.

G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation

The G-quadruplex （G4） elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix. This （34 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA. The reversibility and structural diversity of G4s make them highly versatile genetic structures, as demonstrated by their roles in various functions including telomere metabolism, genome maintenance, immunoglobulin gene diversification, transcription, and translation. Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci. To investigate their potential roles in a large-genome model plant species, we computationaily identified 149,988 non-telomeric G4 motifs in maize （Zea mays L., B73 AGPv2）, 29% of which were in non-repetitive genomic regions. G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand, one in the 5~ UTR and the other at the 5~ end of the first intron. Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro. The G4 motifs were prevalent in key regulatory genes associated with hypoxia （group VII ERFs）, oxidative stress （D J-1/GATasel）, and energy status （AMPK/ SnRK） pathways. They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis, sugar degradation, inositol metabolism, and base excision repair. Collectively, the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses. Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize, a model genetic system and major world crop species for feed, food, and fuel.
