Astragali Radix-Notoginseng Radix et Rhizoma medicine pair prevents cardiac remodeling by improving mitochondrial dynamic balance

Astragali Radix(AR) and Notoginseng Radix et Rhizoma(NR) are frequently employed in cardiovascular disease treatment. However, the efficacy of the AR-NR medicine pair(AN) in improving cardiac remodeling and its underlying mechanism remains unclear. This study aimed to evaluate AN's cardioprotective effect and potential mechanism on cardiac remodeling using transverse aortic constriction(TAC) in mice and angiotensin II(Ang II)-induced neonatal rat cardiomyocytes(NRCMs) and fibroblasts in vitro. High-performance liquid chromatography-quadrupole-time of flight tandem mass spectrometry(HPLC-Q-TOF-MS/MS) characterized 23 main components of AN. AN significantly improved cardiac function in the TAC-induced mice. Furthermore, AN considerably reduced the serum levels of N-terminal pro-B-type natriuretic peptide(NT-pro BNP), cardiac troponin T(CTn-T), and interleukin-6(IL-6) and mitigated inflammatory cell infiltration. Post-AN treatment, TAC-induced heart size approached normal. AN decreased cardiomyocyte cross-sectional area and attenuated the upregulation of cardiac hypertrophy marker genes(ANP, BNP, and MYH7) in vivo and in ***, AN alleviated collagen deposition in TAC-induced mice. AN also reduced the expression of fibrosis-related indicators(COL1A1 and COL3A1) and inhibited the activation of the transforming growth factor-β1(TGF-β1)/mothers against decapentaplegic homolog 3(Smad3) pathway. Thus, AN improved TAC-induced cardiac remodeling. Moreover, AN downregulated p-dynamin-related protein(Drp1)(Ser616) expression and upregulated mitogen 2(MFN-2) and optic atrophy 1(OPA1) expression in vivo and in vitro, thereby restoring mitochondrial fusion and fission balance. In conclusion, AN improves cardiac remodeling by regulating mitochondrial dynamic balance, providing experimental data for the rational application of Chinese medicine prescriptions with AN as the main component in clinical practice.

Herein,we report the dynamic kinetic resolution asymmetric acylation ofγ-hydroxy-γ-perfluoroalkyl butenolides/phthalides catalyzed by amino acid-derived bifunctional organocatalysts,and a series of ketals were obtained in high yields(up to 95%)and excellent enantioselectivities(up to 99%).In terms of synthetic utility,the reaction can be performed on a gram scale,and the product can be converted into potential biological nucleoside analog.

Interannual variability of boreal summer intraseasonal oscillation over the northwestern Pacific influenced by the Pacific Meridional Mode

During the boreal summer,intraseasonal oscillations exhibit significant interannual variations in intensity over two key regions:the central-western equatorial Pacific(5°S-5°N,150°E-150°W)and the subtropical Northwestern Pacific(10°-20°N,130°E-175°W).The former is well-documented and considered to be influenced by the ENSO,while the latter has received comparatively less attention and is likely influenced by the Pacific Meridional Mode(PMM),as suggested by partial correlation analysis *** elucidate the physical processes responsible for the enhanced(weakened)intraseasonal convection over the subtropical northwestern Pacific during warm(cold)PMM years,the authors employed a moisture budget *** findings reveal that during warm PMM years,there is an increase in summer-mean moisture over the subtropical northwestern *** increase interacts with intensified vertical motion perturbations in the region,leading to greater vertical moisture advection in the lower troposphere and consequently resulting in convective *** a process is pivotal in amplifying intraseasonal convection *** observational findings were further verified by model experiments forced by PMM-like sea surface temperature patterns.

The gut microbiota and the associated metabolism play a pivotal role in maintaining human health,yet their assessment is challenging due to their inherent diversity,variety,and ***,we developed a multichannel sensor array based on a single fluorescent probe,allowing for rapid and robust profiling gut microbiota and their key metabolites including cysteine(Cys),glutathione(GSH),and homocysteine(Hcy).The assay leverages the single fluorescent probe that is with multiple binding sites and the cross-reactive sensing principle to specifically recognize different biological *** analyzing the pattern of biological thiols,the assay is capable of rapidly identifying six gut-derived bacteria including probiotics,neutral,and pathogenic strains based on fluorescent fingerprints within 5 min,and also discriminating bacteria and their mixtures with different *** the assay that enables the simultaneous measurement of multiple gut-derived bacteria and their metabolites,the designed array achieved an accuracy of 0.99 when discriminating colorectal cancer(CRC)patient feces samples from healthy ***,the as-prepared sensor array can also be used to identify various stages of *** simplicity,rapidness,and cost-effectiveness of our approach render it a robust platform for the analysis of gut microbiota.

research progress of small-molecule natural medicines for the treatment of ischemic stroke

Stroke is the second leading cause of disability and mortality worldwide, imposing a substantial socioeconomic burden on individuals and healthcare systems. Annually, approximately 14 million people experience stroke, with ischemic stroke comprising nearly 85% of cases, of which 10% to 20% involve large vessel occlusions. Currently, recombinant tissue plasminogen activator(tPA) remains the only approved pharmacological intervention. However, its utility is limited due to a narrow therapeutic window and low recanalization rates, making it applicable to only a minority of patients. Therefore, there is an urgent need for novel therapeutic strategies, including pharmacological advancements and combinatory ***-molecule natural medicines, particularly those derived from traditional Chinese herbs,have demonstrated significant therapeutic potential in ischemic stroke management. These compounds exert multiple neuroprotective effects, such as antioxidation, anti-inflammatory action, and inhibition of apoptosis, all of which are critical in mitigating stroke-induced cerebral damage. This review comprehensively examines the pathophysiology of acute ischemic stroke(AIS) and highlights the recent progress in the development of small-molecule natural medicines as promising therapeutic agents for cerebral ischemic stroke.

The CmCYC2 factors regulated the different biosynthesis and distribution of phenolic compounds in the capitulum of Chrysanthemum morifolium‘Hangju’

Chrysanthemum morifolium *** an important industrial crop with a high medicinal value,whose capitulum consists of ray and disc florets containing numerous bioactive components,such as flavonoids and phenolic ***,the distribution and accumulations of the bioactive metabolites in florets of Chrysanthemum morifolium were ***,the ray and disc florets of Chrysanthemum morifolium were separated for comparing the bioactive *** contents of phenolic compounds and antioxidant activities were significantly different between the two *** metabolomic analysis identified 203 and 241 differential metabolites between ray and disc florets at two flowering *** derivatives of quercetin,isorhamnetin,tricin,and caffeoylquinic acid were enriched in the disc florets,while apigenin,kaempferol,acacetin,and their glycosides mainly accumulated in the ray ***,the results of transcriptome and qRT-PCR suggested that the F3H,F3'H,and FLS genes highly expressed in disc florets,while the FNS gene was well expressed in ray florets,which explained the different distribution of phenolic compounds between ray and disc ***,the expression of CmCYC2 genes was found to be significantly related to the distribution of flavones by correlation *** was proved by the luciferase reporter assay that CmCYC2a and CmCYC2d factors could bind and activate the CmFNS promoter to regulate the flavones biosynthesis in ray *** results revealed the significant difference of metabolite contents and gene expression profile between ray and disc floret in Chrysanthemum morifolium and demonstrated that the quality of Chrysanthemum morifolium‘Hangju'was closely relevant to its flower morphology.

Role of the Greenland Sea ice anomaly in the late-spring drought over Northwest China

Drought across Northwest China in late spring has exerted a vital effect on the local climate and agricultural production,and has been alleviated during the past *** study explored the influence of the preceding Arctic sea ice on the May drought in Northwest China caused by the precipitation *** analysis indicated that when the Greenland Sea ice concentration is abnormally high during February to April,the dry conditions in Northwest China tend to be *** increase of sea ice in the Greenland Sea can excite a meridional circulation,which causes sea surface temperature(SST)anomalies in the North Atlantic via the sea-air interaction,manifested as significant warm SST anomalies over the south of Greenland and the subtropical North Atlantic,but negative SST anomalies over the west of the *** abnormal SST pattern maintains to May and triggers a zonal wave train from the North Atlantic through Scandinavia and Central Asia to Northwest China,leading to abnormal cyclones in Northwest ***,Northwest China experiences a more humid climate than usual.

A robust statistical prediction model for late-summer heavy precipitation days in North China

Recently,heavy precipitation(HP)events have occurred frequently in North China(NC),causing devastating economic losses and human ***,the short-term climate prediction of HP is quite *** year-to-year increment(DY)method and sliding correlations,we developed a robust seasonal prediction model for late-summer HP days(HPDs)in NC during 1982–2022,utilizing three independent predictors—February sea surface temperature(SST)in the Indian Ocean(SST_IO),February snow depth over North Asia(SDE_NA),and May melted snow depth in NC(MSDE_NC).The SST_IO anomalies affect NC's precipitation through the Pacific-Japan *** SDE_NA anomalies are associated with East Asian anomalous anticyclone by southeastern propagation of Rossby wave at *** MSDE_NC anomalies are followed by vertical motion and moisture anomalies in situ and thereby cause precipitation *** prediction model can well simulate the variations of the HPDs,with a correlation coefficient(CC)of 0.81(0.65)between the observed and predicted HPDs_DY(HPDs_anomaly).The percentage with the same sign for 15 extreme HPDs_anomaly years(PSSE)is 100%.Moreover,in the cross-validation test during 1982–2022,the PSSE for HPDs_anomaly is as high as 100%,along with a low rootmean-square error of *** independent hindcasts during 2013–2022,the CC between the observed and predicted HPDs_DY(HPDs_anomaly)is 0.93(0.83),together with high Nash-Sutcliffe efficiency(0.82)and agreement index(0.89).Specifically,the predictions are broadly consistent with the observations for 2015,2016,2017,2021,and 2022,reflecting excellent performance of this prediction model of HPDs in NC.

Atractylenolide Ⅰ ameliorates post-infectious irritable bowel syndrome by inhibiting the polymerase Ⅰ and transcript release factor and c-Jun N-terminal kinase/inducible nitric oxide synthase pathway

OBJECTIVE: To explore the therapeutic effect and target of atractylenolide I(AT-I) on post-infectious irritable bowel syndrome(PI-IBS) rats. METHODS: Therefore, the preliminarily mechanism of AT-I in anti-PI-IBS were first predicted by network pharmacology and molecular docking, then the possible signaling pathways were systematically analyzed. Finally, the potential therapeutic targets and possible signaling pathways of AT-I on PI-IBS in Sprague-Dawley(SD) rat model were verified by experiments. RESULTS: AT-I could alleviate PI-IBS symptoms and reduce the expression of tumor necrosis factor α, interleukin-6 and Interferon-gamma in PI-IBS SD rat model and inhibit the c-Jun N-terminal kinase/inducible nitric oxide synthase(JNK/iNOS) pathway. Notably, AT-I treatment could inhibit the overexpression of polymerase Ⅰ and transcript release factor(PTRF). CONCLUSION: AT-I could alleviate PI-IBS symptoms through downregulation of PTRF and inhibiting the JNK/iNOS pathway. This study not only provides a scientific basis to clarify the anti-PI-IBS effect of AT-I and its mechanism but also suggests a novel promising therapeutic strategy to treat the PI-IBS.
