The aim of this study is to produce large quantities of yam tubers in the field from seed tubers derived from vitroplants regenerated from aerial stem cuttings and farmers’ tuber seeds. Seed tubers from ten yam cultivars, including five from Dioscorea alata (cDa083, cDa053, cDa115, cDa150 and cDa266) and five from Dioscorea cayenensis-rotundata (cDr015, cDr027, cDr150, cDr206 and cDr148) with distinct agro-morphological characteristics, were used as material. A completely randomized block design with 3 replications was used. In each block, all cultivars and seed types were represented. When put into cultivation, the tubers produced by the vitroplants all germinated and gave 100% healthy plants, compared with 86% to 100% healthy plants for the seed tubers produced by the farmers. Yields per hectare ranged from 10 to 25 tonnes for seed tubers produced by in vitro plants, and from 4 to 9 tons for seed tubers produced by farmers. Seed tubers from vitroplants can therefore be recommended to farmers as a solution to the problem of seed tuber unavailability.

A major challenge facing photodynamic therapy(PDT) is that the activity of the immuneinduced infiltrating CD8^(+)T cells is subject to the regulatory T lymphocytes(Tregs), leaving the tumor at risk of recurrence and metastasis after the initial ablation. To augment the antitumor response and reprogram the immunosuppressive tumor microenvironment(TME), a supramolecular photodynamic nanoparticle(DACss) is constructed by the host-guest interaction between demethylcantharidin-conjugated β-cyclodextrin(DMC-CD) and amantadine-terminated disulfide-conjugated FFVLGGGC peptide with chlorin e6 decoration(Ad-ss-pep-Ce6) to achieve intelligent delivery of photosensitizer and immunomodulator for breast cancer treatment. The acid-labile β-carboxamide bond of DMC-CD is hydrolyzed in response to the acidic TME, resulting in the localized release of DMC and subsequent inhibition of *** guest molecule Ad-ss-pep-Ce6 can be cleaved by a high level of intracellular GSH, reducing photosensitizer toxicity and increasing photosensitizer retention in the tumor. With a significant increase in the CTL/Treg ratio, the combination of Ce6-based PDT and DMC-mediated immunomodulation adequately achieved spatiotemporal regulation and remodeling of the TME, as well as improved primary tumor and in situ lung metastasis suppression with the aid of PD-1 antibody.

Development of Novel Antiviral Therapies for Hepatitis C Virus

Over 170 million people worldwide are infected with hepatitis C virus (HCV), a major cause of liver diseases. Current interferon-based therapy is of limited efficacy and has significant side effects and more effective and better tolerated therapies are urgently needed. HCV is a positive, single-stranded RNA virus with a 9.6 kb genome that encodes ten viral proteins. Among them, the NS3 protease and the NS5B polymerase are essential for viral replication and have been the main focus of drug discovery efforts. Aided by structure-based drug design, potent and specific inhibitors of NS3 and NS5B have been identified, some of which are in late stage clinical trials and may significantly improve current HCV treatment. Inhibitors of other viral targets such as NS5A are also being pursued. However, HCV is an RNA virus characterized by high replication and mutation rates and consequently, resistance emerges quickly in patients treated with specific antivirals as monotherapy. A complementary approach is to target host factors such as cyclophilins that are also essential for viral replication and may present a higher genetic barrier to resistance. Combinations of these inhibitors of different mechanism are likely to become the essential components of future HCV therapies in order to maximize antiviral efficacy and prevent the emergence of resistance.

Sizing and performance features of rotary and reciprocating positive displacement pumps

An overview of some popular rotary and reciprocating positive displacement( PD) pump types is given with the objective of presenting and comparing the respective sizing relationships and performance features. Reciprocating pumps discussed are the piston and plunger types. Rotary pumps addressed are gear( external and internal),vane,lobe,screw,and liquid ring pumps. To put the relative pump sizes in perspective,attention is fixed on the rotors or reciprocating elements of PD pumps,just as impellers indicate the sizes of rotodynamic pumps. The size of a PD pump is found from a dimensionless combination of displacement flow rate,rotative speed and diameter. The flow rate,head( or pressure rise) and power are related through the component efficiencies. The cavitation coefficient,often close to unity,connects the rotor tip speed or piston speed with the required NPSH,which can also be affected by the pressure rise of the pump due to leakage across the internal clearances. Operational effects due to cavitation,ingestion of gas or abrasives,and viscous and non-Newtonian fluids are discussed.

Development and validation of a GC–FID method for quantitative analysis of oleic acid and related fatty acids

Oleic acid is a common pharmaceutical excipient that has been widely used in various dosage forms. Gas chromatography （GC） has often been used as the quantitation method for fatty acids normally requiring a derivatization step. The aim of this study was to develop a simple, robust, and derivatization-free GC method that is suitable for routine analysis of all the major components in oleic acid USP-NF （United States Pharmacopeia-National Formulary） material. A gas chromatography-flame ionization detection （GC-FID） method was developed for direct quantitative analysis of oleic acid and related fatty acids in oleic acid USP-NF material. Fifteen fatty acids were separated using a DB-FFAP （nitroterephthalic acid modified polyethylene glycol） capillary GC column （30 m × 0.32 mm i.d.） with a total run time of 20 rain. The method was validated in terms of specificity, linearity, precision, accuracy, sensitivity, and robust- ness. The method can be routinely used for the purpose of oleic acid USP-NF material analysis.

Background:Patients with inflammatory bowel disease(IBD)are at increased risk of herpes zoster(HZ).We evaluated the incidence of HZ in ulcerative colitis(UC)and Crohn’s disease(CD)patients and compared this with HZ incidence in a non-IBD ***:We conducted a retrospective cohort study(GSK study identifier:VEO-000043)of adults aged18 years with UC and CD and without IBD who were identified from claims recorded in a US healthcare database between October 2015 and February *** HZ incidence rates/1,000 person-years(PY)were calculated,and comparisons of HZ incidence rates between UC or CD and non-IBD cohorts were made using adjusted generalized linear ***:The study population comprised a total of 29,928 UC,25,959 CD,and 11,839,329 non-IBD *** overall HZ incidence rates were 13.64/1,000 PY(UC),15.94/1,000 PY(CD),and 7.95/1,000 PY(non-IBD).UC and CD patients had increased HZ incidence rates,with adjusted incidence rate ratios of 1.35(95%confidence interval[CI],1.26–1.44)and 1.66(95%CI,1.56–1.77),respectively,compared with non-IBD *** analysis indicated increased relative rates of HZ in progressively younger age strata in the UC and CD patients compared with non-IBD *** incidence rates were higher in UC and CD patients who had previously received thiopurines or methotrexate,TNF-inhibitors,or corticosteroids than in UC and CD patients who did not take those ***:UC and CD patients had increased HZ incidence rates compared with patients without IBD,demonstrating the importance of HZ prevention in IBD patients.

Asbestos cement （AC） pipes were commonly installed in the drinking water distribution systems from the mid 1920s to the late 1980s. In recent years, an increase in the number of water main breaks has occurred in the AC portions of some pipe networks, which can be partially attributed to the corrosion of the aged pipes. This study evaluated the potential role that microorganisms may have played in the degeneration and failure of AC pipes. In this study, a fresh AC pipe section was collected from the distribution network of the City of Regina, Canada and examined for microbiological activities and growth on inside surfaces of pipe sample. Black slime bacterial growths were found to be attached to inner pipe surfaces and a distinctively fibrous internal coating （patina） with iron oxides was formed over the time. The microbial populations inside the patina and the black slime were tested with BART^TM testers. Heterotrophic aerobic bacteria （HAB） and slime forming bacteria （SLYM） dominated in both the black growths and inside the patina. Iron related bacteria, denitrification bacteria and sulfate reducing bacteria were also commonly present. Microbial challenge assays were conducted by submerging the cut segments of the AC pipe into selected bacterial cultures for a period of 10 days under both aerobic and anaerobic environments. Weight changes were determined and the surface morphology was examined for each of the assayed pipe segments. Results indicated that acid producing bacteria, SLYM and HAB could facilitate the pipe weight loss under anaerobic environments.

A novel compound, Delta-9-Tetrahydrocannabinol Nitrogen Mustard (THC-mustard), waschemically synthesized and characterized. The rationale was to target a known anti-tumoragent, nitrogen mustard, to tumor cells with 'THC Receptors' and/or 'Estrogen Recep-tots', A microtest by vitro bioassay was designed and developed to compare the ID50of the drug in cell culture against various tumor cell types. The ID50’s of the THC-mustardwere determined against several tumor cell types in culture, compared with MechlorethamineHCI, Delta-9-Tetrahydrocannabinol (THC), and an equimolar mixture of THC and nitrogennmstard. A preliminary toxicity study by the NCI of the THC-mustard (in vivo) in Swissmale mice bearing the P388 tumor (1 dose × 1day) by the I. P. route was also carried out.A deseription of the rationale, chemical synthesis and characterization, bioassay, and resultsis herewith presented.

Probiotic modulation of dendritic cells co-cultured with intestinal epithelial cells

AIM： To investigate cytokine production and cell surface phenotypes of dendritic cells （DC） in the presence of epithelial cells stimulated by probiotics.METHODS： Mouse DC were cultured alone or together with mouse epithelial cell monolayers in normal or in- verted systems and were stimulated with heat-killed probiotic bacteria, Bifidobacterium lactis ADO 11 （BL）, Bifidobacterium bilfidum BGN4 （BB）, Lactobacillus casei IBS041 （LC）, and Lactobacillus acidophilus AD031 （LA）, for 12 h. Cytokine levels in the culture supernatants were determined by enzyme-linked immunosorbent as say and phenotypic analysis of DC was investigated by flow cytometry.RESULTS： BB and LC in singlecultured DC increased the expression of I-Ad, CD86 and CD40 （I-Ad, 18.51 vs 30.88, 46.11, CD86, 62.74 vs 92.7, 104.12; CD40, 0.67 vs 6.39, 3.37, P 〈 0.05）. All of the experimental probiot-ics increased the production of inflammatory cytokines, interleukin （IL）-6 and tumor necrosis factor （TNF）-α. However, in the normal coculture systems, LC and LA decreased the expression of I-A^α （39.46 vs 30.32, 33.26, P 〈 0.05）, and none of the experimental probiotics increased the levels of IL-6 or TNF-α. In the inverted coculture systems, LC decreased the expression of CD40 （1.36 vs -2.27, P 〈 0.05）, and all of the experimental probiotics decreased the levels of IL-6. In addition, BL increased the production of IL-10 （103.8 vs 166.0, P 〈 0.05） and LC and LA increased transforming growth factor-13 secretion （235.9 vs 618.9, 607.6, P 〈 0.05）.CONCLUSION： These results suggest that specific pro- biotic strains exert differential immune modulation mediated by the interaction of dendritic cells and epithelial cells in the homeostasis of gastrointestinal tract.
