Objective:To explore the effect of AIBL on Oncomelania hupensis,the intermediate snail host of Schistosoma ***:The enzyme histochemical profiles of cholineslerase, cytochrome oxidase,lactate dehydrogenase,nitric oxide synthase,and succinate dehydrogenase in the soft tissues of Oncomelania hupensis,the intermediate host snail of Schistosoma japonicum, were analyzed before and after treatment with the active ingredient of Buddleia lindleyana(AIBL), a potent and safe plant ***:Treatment with AIBL induced a notable decrease in the activities of the five enzymes(P<0.01).Conclusions:The results indicate that AIBL impairs the activities of the enzymes,thereby influencing the transfer of neurotransmitter and energy supply in Oncomelania hupensis and ultimately harming their various physiological functions, which are considered to cause death of the species.

A suspended cell line from Trichoplusia ni embryos was established, and its susceptibility to Autographa californica multiple nuclear polyhedrosis virus （AcMNPV） infection was investigated. This cell line had characteristics distinct from the BTI-Tn5B 1- 4 cell line （Tn5B 1-4） from T. ni in growth, and showed approximately the same responses to AcMNPV infection, production of occlusion bodies, and levels of recombinant protein expression. No clumps were observed at maximum cell density at late-log phase in shake-flask or T-flask cultures, and thus the cells represent a useful new contribution for baculovirus research. The cells consist of two major morphological types： approximately 70% spindle-shaped cells and 30% round cells. The cell line was highly susceptible to virus infection and produced around 107 AcMNPV occlusion bodies per cell, on average. Production of β-galactosidase and secreted alkaline phosphatase was high with 3.97 ± 0.13 × 10^4 IU/mL and 3.48 ± 0.40IU/mL, respectively. This cell line may be applicable for studies of scale-up production of viruses or baculovirus-insect cell expression. We also believe the new line can be a source for cell clones with higher production of virus and recombinant proteins compared to the parent or other existing cell lines such as Tn5B 1-4.

Collective behaviors efficiently impart benefits to a diversity of species ranging from bacteria to *** larvae tend to cluster and form coordinated digging groups under crowded conditions,yet understanding the rules governing this behavior is in its *** primarily took advantage of the Drosophila model to investigate cooperative foraging ***,we report that Drosophila-related species and the black soldier fly have evolved a conserved strategy of cluster digging in food ***,we investigated relative factors,including larval stage,population density,and food stiffness and quality,that affect the cluster digging ***,oxygen supply through the posterior breathing spiracles is necessary for the organization of digging *** importantly,we theoretically devise a mathematical model to accurately calculate how the cluster digging behavior expands food resources by diving depth,cross-section area,and food *** found that cluster digging behavior approximately increases 2.2 fold depth,1.7-fold cross-section area,and 1.9 fold volume than control groups,*** of food sources significantly facilitates survival,larval development,and reproductive success of Drosophila challenged with competition for limited food resources,thereby conferring trophic benefits to fitness in ***,our findings highlight that the cluster digging behavior is a pivotal behavior for their adaptation to food scarcity,advancing a better understanding of how this cooperative behavior confers fitness benefits in the animal kingdom.

Abscisic acid(ABA)-based chemically induced proximity(CIP) is primarily mediated by the interaction of the ABA receptor pyrabactin resistance 1-like 1(PYL1) and the 2C-type protein phosphatase ABI1, which confers ABA-induced proximity to their fusion proteins, and offers precise temporal control of a wide array of biological processes. However, broad application of ABA-based CIP has been limited by ABA response intensity. In this study, we demonstrated that ABA-induced interaction between another ABA receptor pyrabactin resistance 1(PYR1) and ABI1 exhibited higher ABA response intensity than that between PYL1 and ABI1 in HEK293T cells. We engineered PYR1-ABI1and PYL1-ABI1 into ABA-induced transcriptional activation tools in mammalian cells by integration with CRISPR/d Cas9 and found that the tool based on PYR1-ABI1 demonstrated better ABA response intensity than that based on PYL1-ABI1 for both exogenous and endogenous genes in mammalian cells. We further achieved ABA-induced RNA m6A modification installation and erasure by combining ABA-induced PYR1-ABI1 interaction with CRISPR/d Cas13, successfully inhibiting tumor cell proliferation. We subsequently improved the interaction of PYR1-ABI1 through phage-assisted continuous evolution(PACE), successfully generating a PYR1 mutant(PYR1m) whose interaction with ABI1 exhibited a higher ABA response intensity than that of the wild-type. In addition, we tested the transcriptional activation tool based on PYRm-ABI1 and found that it also showed a higher ABA response intensity than that of the wild type. These results demonstrate that we have developed a novel ABA-based CIP and further improved upon it using PACE, providing a new approach for the modification of other CIP systems.

A curcumin derivative ligand,1,7-bis(3-methoxyl-4-acetoxyl)phenyl-1,6-heptadiene-3,5-diketone (diacetylcurcumin,abbreviated as HL),and its Cu and Ni complexes have been synthesized and fully characterized by elemental analyses,IR,1 HNMR and molar *** resulting complexes exhibit two-photon excited fluorescence (TPEF) in DMF,and have been proven to be potentially useful for two-photon microscopy imaging in living *** addition,cytotoxicity tests showed that the low-micromolar concentrations of ML 2 did not cause significant reduction in cell viability over a period of at least 24 h and should be safe for further biological studies.

The synthesis of atomically ordered Pt-based intermetallic electrocatalysts for the direct alcohol fuel cells generally requires the addition of surfactants or the high-temperature ***,some residual surfactants on the surface of the assynthesized catalysts would prevent the exposure of catalytic active sites,the high-temperature annealing process is easy to accelerate the sintering of the metal,which both lead to the decline of electrocatalytic ***,we construct the atomically ordered bimetallic PtBi intermetallics with clean surfaces and unique three-dimensional hollow acorn-shell-like structure(3D PtBi HASL)by a simple,low-temperature,surfactant-free one-pot synthetic *** from the special hollow structures,the obtained 3D PtBi HASL intermetallics expose abundant accessible active ***,the introduction of oxophilic metal Bi can enhance adsorption of OHads,thereby significantly facilitating removal of poisoned *** functional theory(DFT)simulations further indicate that formation of the PtBi intermetallic phase with the downshift of the Pt d-band center endows 3D Pt49.4Bi50.6 HASL intermetallics with significantly attenuated COads and enhanced OHads adsorption,bringing about the boosting electrocatalytic *** mass activity of the 3D Pt49.4Bi50.6 HASL intermetallics for ethylene glycol oxidation reaction is as high as 24.67 A·mgPt^(−1),which is 12.98 times higher than that of commercial Pt/C(1.90 A·mgPt^(−1)).This work may inspire the design of Pt-based intermetallics as high-efficiency anode electrocatalysts for fuel cell applications.

In recent years, adenosine tri-phosphate （ATP） has been reported to exist in apoplasts of plant cells as a signal molecule. Extracellular ATP （eATP） plays important roles in plant growth, development, and stress tolerance. Here, extra- cellular ATP was found to promote stomatal opening of Arabidopsis thaliana in light and darkness. ADP, GTP, and weakly hydrolyzable ATP analogs （ATPγS, Bz-ATP, and 2meATP） showed similar effects, whereas AMP and adenosine did not affect stomatal movement. Apyrase inhibited stomatal opening. ATP-promoted stomatal opening was blocked by an NADPH oxidase inhibitor （diphenylene iodonium） or deoxidizer （dithiothreitol）, and was impaired in null mutant of NADPH ox- idase （atrbohD/F）. Added ATP triggered ROS generation in guard cells via NADPH oxidase. ATP also induced Ca^2＋ influx and H ＋ efflux in guard cells. In atrbohD/F, ATP-induced ion flux was strongly suppressed. In null mutants of the heterotrimeric G protein α subunit, ATP-promoted stomatal opening, cytoplasmic ROS generation, Ca^2＋ influx, and ^H＋ efflux were all sup- pressed. These results indicated that eATP-promoted stomatal opening possibly involves the heterotrimeric G protein, ROS, cytosolic Ca^2＋, and plasma membrane H＋-ATPase.
