ISP and PAP4 peptides promote motor functional reco.ery after peripheral nerve injury

Both intracellular sigma peptide(ISP) and phosphatase and tensin homolog agonist protein(PAP4) promote nerve regeneration and motor functional reco.ery after spinal co.d injury. However, the role of these two small peptides in peripheral nerve injury remains unclear. A rat model of brachial plexus injury was established by crush of the C6 ventral root. The rats were then treated with subcutaneous injection of PAP4(497 μg/d, twice per day) or ISP(11 μg/d, once per day) near the injury site for 21 successive days. After ISP and PAP treatment, the survival of motoneurons was increased, the number of regenerated axons and neuromuscular junctions was increased, muscle atrophy was reduced, the electrical response of the motor units was enhanced and the motor function of the injured upper limbs was greatly improved in rats with brachial plexus injury. These findings suggest that ISP and PAP4 promote the reco.ery of motor function after peripheral nerve injury in rats. The animal care and experimental procedures were approved by the Laboratory Animal Ethics co.mittee of Jinan University of China(approval No. 20111008001) in 2011.

Rice（Oryza sativa L.） MADS3 transcription factor regulates the homeostasis of reactive oxygen species（ROS）during late anther development,and one MADS3 mutant,mads3-4,has defective anther walls,aborted microspores and co.plete male ***,we report the untargeted metabolomic analysis of both wild type and mads3-4 mature *** of MADS3 led to an unbalanced redox status and caused oxidative stress that damages lipid,protein,and *** co.e with oxidative stress in mads3-4 anthers,soluble sugars were mobilized and carbohydrate metabolism was shifted to amino acid and nucleic acid metabolism to provide substrates for the biosynthesis of antioxidant proteins and the repair of *** of MADS3 also affected other aspects of rice anther development such as seco.dary metabolites associated with cuticle,cell wall,and auxin *** of the disco.ered metabolic changes in mads3-4 anthers were co.roborated with changes of expression levels of co.responding metabolic pathway ***,this co.parative metabolomic analysis indicated that MADS3 gene affects rice anther development far beyond the ROS homeostasis regulation.

Because of their strong proliferative capacity and multi-potency, placenta-derived mesenchymal stem cells have gained interest as a cell source in the field of nerve damage repair. In the present study, human placenta-derived mesenchymal stem ceils were induced to differentiate into neural stem cells, which were then transplanted into the spinal co.d after local spinal co.d injury in rats. The motor functional reco.ery and pathological changes in the injured spinal co.d were observed for 3 successive weeks. The results showed that human placenta-derived mesenchymal stem cells can differentiate into neuron-like cells and that induced neural stem cells co.tribute to the restoration of injured spinal co.d without causing transplant rejection. Thus, these cells promote the reco.ery of motor and sensory functions in a rat model of spinal co.d injury. Therefore, human placenta-derived mesenchymal stem cells may be useful as seed cells during the repair of spinal co.d injury.

Programmed cell death receptor 1(PD-1)and its ligand,PD-L1,are important immune checkpoint *** antibodies that block PD-1/PD-L1 have shown promising clinical efficacy in a subset of cancer patients,the detailed cellular and molecular mechanisms behind anti-PD-1 and anti-PD-L1 immunotherapy are not well ***,the way in which PD-L1 co.tributes to immune suppression on tumor and non-tumor cells remains *** selectively blocking PD-L1 on either tumor or non-tumor cells,we demonstrated that PD-L1 from both sources suppressed the anti-tumor T-cell *** PD-L1 on either tumor cells or non-tumor cells inhibited tumor growth and enhanced immune cell infiltration,as well as the tumor-specific T-cell ***,simultaneously blocking tumor-and non-tumor-derived PD-L1 maximized anti-tumor T-cell responses and demonstrated *** addition,the relative co.tribution of PD-L1 on tumor and non-tumor cells to immune suppression depended on the PD-L1 expression ***,we found that the F4/80 receptor was involved in the anti-tumor effect of PD-L1 *** together,our data indicate that PD-L1 on both tumor and non-tumor cells is critical for T-cell inhibition,which provides new directions for the optimization of PD-L1-blocking antibodies and the development of clinical biomarker strategies.

Piriformospora indica, a root-co.onizing endophytic fungus of Sebacinales, promotes plant growth and co.fers resistance against biotic and abiotic stresses. In order to co.firm the influence of P. indica on growth, proline, malondialdehyde（MDA）, chlorophyll, and cadmium（Cd） amounts in Nico.iana tabacum under Cd stress, hydroponics, pot and field trials were co.ducted. The results showed that P. indica can store Cd in plant roots and reduce leaf Cd co.tent, reduce the co.centration of MDA, and increase the proline and chlorophyll co.tent and the activities of catalase, peroxidase, and superoxide dismutase under hydroponic Cd stress. RT-PCR analysis showed that the relative expression level of genes Gsh2, Ta PCS1, oas1, GPX, and Hsp70 in co.onized plants was 4.3, 1.4, 2.9, 1.7, and 6.9fold higher than in un-co.onized plants respectively. Cd exposure significantly reduced un-co.onized plants＇ agronomic traits co.pared to P. indica-co.onized ones. Our results suggested that P. indica can sequester Cd in roots, so that much less cadmium was transported to leaves, and the increased co.centrations of antioxidant enzymes, pigments and proline co.tents, as well as the higher expression of stress-related phytochelatin biosynthesis genes in P. indica-inoculated plants, may also serve to protect N. tabacum plants against oxidative damage, enhancing Cd tolerance.

Exact solutions for free vibration frequencies and modes are obtained for thickness- shear and thickness-twist vibrations of unelectroded circular AT-cut quartz plates governed by the two-dimensional scalar differential equation derived by Tiersten and Smythe. co.parisons are made with experimental results and the widely-used perturbation solution by Tiersten and Smythe under the assumption of weak in-plane anisotropy. Our solution is found to be much closer to the experimental results than the perturbation solution. For the frequency of the fundamental thickness-shear mode, the error of the perturbation method is 0.4549%, significant in resonator applications.
