Impact of comorbidity in elderly prostate cancer patients treated with brachytherapy

Objective： To analyze the correlations among comorbidity and overall survival （OS）, biochemical progression-free survival （b-PFS） and toxicity in elderly patients with localized prostate cancer treated with 125I brachytherapy. Methods： Elderly men, aged ≥65 years, with low-intermediate risk prostate cancer, were treated with permanent 125I brachytherapy as monotherapy. Comorbidity data were obtained from medical reports using age-adjusted Charlson comorbidity index （a-CCI）. The patients were categorized into two age groups （〈75 and 〉75 years old）, and two comorbidity score groups （a-CCI ≤3 and 〉3）. Toxicity was scored with radiation Therapy Oncology Group （RTOG） scale. Results： From June 2003 to October 2009, a total of 92 elderly patients underwent prostate brachytherapy, including 57 men （62%） with low-risk prostate cancer, and 35 men （38%） with intermediate-risk prostate cancer. The median age of patients was 75 years （range, 65-87 years）. Forty-seven patients （51%） bad a-CCI _〈3 and 45 patients （49%） a-CCI 〉3. With a median follow-up period of 56 months （range, 24-103 months）, the 5-year actuarial OS and b-PFS were 91.3% and 92.4% respectively, without statistical significance between two Charlson score groups. Toxicity was mild. None of the patients experienced gastrointestinal （GI） toxicity, and only 4 patiens （4%） experienced late genitourinary （GU） grade-3 （G3） toxicity. No correlation between acute GU and GI toxicity and eomorbidity was showed （P=0.50 and P=0.70, respectively）. Conclusions： Our data suggest that elderly men with low-intermediate risk prostate cancer and comorbidity can be considered for a radical treatment as 12Sl low-dose rate brachytherapy.

Neo-adjuvant radiotherapy in rectal cancer

Correlation of microRNAs responding to high dose γ-irradiation with predicted target mRNAs in HeLa cells using microarray analyses

An increasing data indicates that altered microRNAs(miRNAs)participate in the radiation-induced DNA damage ***,a correlation of mRNA and miRNA profiles across the entire genome and in response to irradiation has not been thoroughly *** analyzed miRNA microarray data collected from HeLa cells after ionizing radiation(IR),quantified the expression profiles of mRNAs and performed comparative analysis of the data sets using target prediction algorithms,Gene Ontology(GO)analysis,pathway analysis,and gene network *** results showed that the altered miRNAs were involved in regulation of various cellular ***-gene network analyses revealed that miR-186,miR-106b,miR-15a/b,CCND1and CDK6 played vital role in the cellular radiation *** qRT-PCR,we confirmed that twenty-two miRNAs showed differential expression in HeLa cells treated with IR and some of these miRNAs affected cell cycle *** study demonstrated that miRNAs influence gene expression in the entire genome during the cellular radiation response and suggested vital pathways for further research.

PIMD:An Integrative Approach for Drug Repositioning Using Multiple Characterization Fusion

Promoter of (Ce-Zr)O_2 Solid Solution Modified by Praseodymia in Three-Way Catalysts

The three way catalysts (TWCs) promoters (Ce Zr)O 2, (Pr Ce Zr)O 2 and (Pr Zr)O 2 were prepared by sol gel like method. They were characterized by XRD, EXAFS and BET surface area determination. The reduction features of the promoters were measured by temperature programmed reduction (TPR) of H 2 to access the potential for the promoters containing praseodymia as oxygen storage component in three way catalyst. The (Pr Zr)O 2 cubic solid solution is formed at high temperature up to 800 ℃, which makes it more reducible than the (Ce Zr)O 2 solid solution. For the (Pr Ce Zr)O 2 samples, the ternary solid solution plays an important role in the reduction process. The performance of the three way catalysts with fully formulated Pt, Pd and Rh is proceeded by using both light off temperature under a stoichiometric gas composition and the conversion of CO, C 3H 6 and NO under changing air/fuel ratio at a constant reaction temperature 400 ℃ . The results indicate that a small amount of praseodymia doping into (Ce Zr)O 2 favors the light off temperature of C 3H 6 and NO, and all the catalysts containing praseodymia obviously exhibits enhanced width of S value for NO conversion at lean region ( S ≥1.00).

Selenium as a chemopreventive agent in experimentally induced colon carcinogenesis

AIM:To elucidate the chemopreventive efficacy of selenium during experimentally induced colon ***:Thirty-two male wistar rats were divided into four groups:group □(normal control);group □[1,2-dimethylhydrazine(DMH) treated];group □ (selenium treated);and group □ (DMH + selenium treated).Groups □ and □ were given subcutaneous injections of DMH(30 mg/kg body weight) every week for 20 ***,in the form of sodium selenite,was given to groups □ and at 1 ppm in drinking water ad libitum for 20 *** the end of the study,rats were sacrificed and their colons were analyzed for the development of tumors,antioxidant enzyme levels and histological ***:100 of the DMH treated rats developed tumors,which was reduced to 60 upon simultaneous selenium ***,tumor multiplicity decreased to 1.1 following selenium supplementation to DMH treated *** of lipid peroxidation,glutathione-S-transferase,superoxide dismutase(SOD),catalase,and glutathione peroxidase(GPx) decreased following DMH treatment,whereas levels of glutathione(GSH) and glutathione reductase(GR) significantly increased in DMH treated *** administration to DMH treated rats led to an increase in the levels of lipid peroxidation,SOD,catalase,glutathione-S-transferase and GPx,but decreased the levels of GSH and *** studies on DMH treated rats revealed dysplasia of the colonic histoarchitecture,which showed signs of improvement following selenium ***:The study suggests the antioxidative potential of selenium is a major factor in providing protection from development of experimentally induced colon carcinogenesis.

Mitsugumin 29 (MG29) is a transmembrane protein that is normally found in the triad junction of skeletal muscle. Our previous studies have shown that targeted deletion of mg29 from the skeletal muscle resulted in abnormality of the triad junction structure, and also increased susceptibility to muscle fatigue. To elucidate the basis of these effects, we investigated the properties of Ca2+-uptake and -release in toxin-skinned Extensor Digitorium Longus (EDL) muscle fibers from control and mg29 knockout mice. Compared with the control muscle, submaximal Ca2+-uptake into the sarcoplasmic reticulum (SR) was slower and the storage of Ca2+ inside the SR was less in the mutant muscle, due to increased leakage process of Ca2+ movement across the SR. The leakage pathway is associated with the increased sensitivity of Ca2+/caffeine -induced Ca2+ release to myoplasmic Ca2+. Therefore, the increased fatigability of mutant EDL muscles can result from a combination of a slowing of Ca2+ uptake, modification of Ca2+-induced Ca2+ release (CICR), and a reduction in total SR Ca2+ content.
