Alternative Exposure Parameters and Post Process Noise Reduction Expect Considerable Dose Reduction in Single Mammography Views—Initial Experience on Mastectomy Specimens

Objective: To experimentally indicate a lower limit of dose in mammographic imaging yielding adequate image quality for complementary diagnostic views, by evaluation of image series with different exposure parameters and additional image processing on mastectomy specimens with diverse pathology. Methods: Image series were obtained on seven specimens with different target-filter combinations at different exposure values. Three experienced radiologists assessed the lowest acceptable dose level per specimen using a relative grading technique. With the standard image as reference, fibroglandular tissue and pathological structures, including microcalcifications, were evaluated. Subsequently, a series of pixel binning processes was tested and subjectively assessed on the selected images. Results: The lowest dose level at which image quality was acceptable, and achieved with a W/Ag target-filter combination at 32 kV and 4 mAs. These images can be acquired with 10.4% to 22.4% of the average glandular dose in standard images. Post process pixel binning added to the interpretability of such low dose images. Conclusion: This specimen study suggests that dose level of mammography images might be reduced substantially by general application of a W/Ag spectrum, particularly when combined with post process noise reduction. Future studies should focus on the feasibility of this technique in clinical mammography.

What Are the Chances? fMRI Correlates of Observing High and Low-Probability Actions

Cognitive scientists often use probabilistic equations to model human behavior in ambiguous situations. How, where, and even if such probabilities are represented in the human brain remains largely unknown. Here, we manipulated the probability of simple bottle-pouring action based on two considerations, the relative fullness of two glasses and the relative distance between the two glasses and the bottle. Whole brain functional magnetic resonance imaging was used to measure brain activity while participants viewed probable and improbable pouring actions. Improbable actions elicited increased activity in the theory of mind (ToM) network, commonly found active when trying to grasp the intentions of others, whereas probable actions elicited increased activity in the human mirror neuron system (hMNS) and areas associated with mental imagery and memory. These data provide novel insight into the brain mechanisms humans use to distinguish between high and low-probability actions.

Target-activated and ratiometric photochromic probe for “double-check” detection of toxic thiols in live cells

Photochromic molecules can achieve reversible isomerization upon alternate light irradiations, which offers a great opportunity to improve the precision of analytes detection and imaging in complicated biological environments. Previous reported photochromic probe exhibited only mono-color switching and an initially fluorescence-ON state that may cause high background signal and impose an adverse impact on the desired sensing precision. To overcome this set-back, we developed a novel photochromic probe with an analyte-activation mode for ratiometric sensing of toxic thiols in both real water samples and live cells. The dynamic dual-fluorescence signal is released only after the fast and selective cleavage of the 2,4-dinitrophenyl sulfonate by the targeted thiophenol derivatives. Consequently, a "double-check" with synchronized dual-fluorescence blinking for analyte detection is successfully employed upon alternate light triggers with rapid response(k=7.2×10^(-2) s^(-1)), high sensitivity(LOD=6.1 nM) as well as selectivity of thiophenol derivatives over other common thiol species(e.g., GSH, Cys and Hcy). The photochromic probe was successfully introduced to the fast and on-site detection of highly toxic thiophenols in real waste water samples. Moreover, by using confocal laser-scanning microscopy(CLSM) and flow cytometric analysis, the potential applications of this ratiometric photochromic probe for trace toxic thiol sensing in live cells are examined.

Cytomegalovirus in inflammatory bowel disease： Asystematic review

AIM: To identify definitions of cytomegalovirus(CMV) infection and intestinal disease, in inflammatory bowel disease(IBD), to determine the prevalence associated with these ***: We conducted a systematic review and interrogated Pub Med, EMBASE and Cochrane for literature on prevalence and diagnostics of CMV infection and intestinal disease in IBD patients. As medical headings we used "cytomegalovirus" OR "CMV" OR "cytomegalo virus" AND "inflammatory bowel disease" OR "IBD" OR "ulcerative colitis" OR "colitis ulcerosa" OR "Crohn's disease". Both Me SH-terms and free searches were performed. We included all types of English-language(clinical) trials concerning diagnostics and prevalence of CMV in ***: The search strategy identified 924 citations, and 52 articles were eligible for inclusion. We identified 21 different definitions for CMV infection, 8 definitions for CMV intestinal disease and 3 definitions for CMV reactivation. Prevalence numbers depend on used definition, studied population and region. The highest prevalence for CMV infection was found when using positive serum PCR as a definition, whereas for CMV intestinal disease this applies to the use of tissue PCR > 10 copies/mg tissue. Most patients with CMV infection and intestinal disease had steroid refractory disease and came from East ***: We detected multiple different definitions used for CMV infection and intestinal disease in IBD patients, which has an effect on prevalence numbers and eventually on outcome in different trials.

Candidate colorectal cancer predisposing gene variants in Chinese early-onset and familial cases

AIM: To investigate whether whole-exome sequencing may serve as an efficient method to identify known or novel colorectal cancer(CRC) predisposing genes in early-onset or familial CRC ***: We performed whole-exome sequencing in 23 Chinese patients from 21 families with nonpolyposis CRC diagnosed at ≤ 40 years of age, or from multiple affected CRC families with at least 1 firstdegree relative diagnosed with CRC at ≤ 55 years of *** DNA from blood was enriched for exome sequences using the Sure Select Human All Exon Kit, version 2(Agilent Technologies) and sequencing was performed on an Illumina Hi Seq 2000 *** were processed through an analytical pipeline to search for rare germline variants in known or novel CRC predisposing ***: In total, 32 germline variants in 23 genes were identified and confirmed by Sanger *** 6 of the 21 families(29%), we identified 7 mutations in 3 known CRC predisposing genes including MLH1(5 patients), MSH2(1 patient), and MUTYH(biallelic, 1 patient), five of which were reported as *** remaining 15 families, we identified 20 rare and novel potentially deleterious variants in 19 genes, six of which were truncating *** previously unreported variant identified in a conserved region of EIF2AK4(***738_Asp739insA rgA rg) was found to represent a local Chinese variant, which was significantly enriched in our early-onset CRC patient cohort compared to a control cohort of 100 healthy Chinese individuals scored negative by colonoscopy(33.3% vs 7%, P < 0.001).CONCLUSION: Whole-exome sequencing of early-onset or familial CRC cases serves as an efficient method to identify known and potential pathogenic variants in established and novel candidate CRC predisposing genes.

NPHS2 Gene Mutation and Polymorphisms in Indonesian Children with Steroid-Resistant Nephrotic Syndrome

Objective:?Although several NPHS2 gene mutations and polymorphisms were described and?associated with clinical manifestation of steroid-resistant nephrotic syndrome (SRNS),?the?occurrence of these genetic abnormalities or variants appeared?to be influenced by race and ethnic group. We have investigated?probable mutations and variants in NPHS2 gene involved in SRNS and their association with clinical manifestations. Methods: We examined 28 children?with primary SRNS?whovisited?the pediatric nephrology division of 10 teaching hospitals in Indonesia. Molecular genetic studies of the NPHS2 gene were?conducted through screenings?for the exon 1, exon 2, and exon 8. The mutational analysis of NPHS2 was performed by DNA sequencing.?Fisher’s Exact Test was used to determine?the?correlation?between?NPHS2 polymorphisms and clinical ***:?Seven?females (25%) and 21 males (75%)?participated in the study.?The mean age of the subjects with 95% CI is: 7.6 (6.1 - 9.0) years while the mean age at onset of disease with 95% CI is: 5.4 (3.9 - 7.0) years. Sixteen patients (57.14%) were younger than 6 years at the onset of disease. Seventeen (60.7%) subjects had normal eGFR, while 11 (39.3%) had chronic renal insufficiency. The mean eGFR of the subjects with 95% CI is: 111.4 (87.7 - 135.1) ml/min/1.73 m2. The mean systolic blood pressure with 95% CI is: 117.0 (108.9 - 125.1) mmHg and the mean diastolic blood pressure with 95% CI is: 77.0 (70.3 - 83.7) mmHg.?We identified 6 NPHS2polymorphisms,?i.e.?g.-52G>T, c.101A>G, g.-117C>T, c.288C>T, c.954C>T, and c.1038A>G and no mutation?was found. There was?no correlation?between?NPHS2 polymorphisms and clinical manifestations (p > 0.05). Conclusion: The?results demonstrate no mutation of NPHS2 gene, and the 6 NPHS2 gene polymorphisms that were identified have no correlation with the clinical manifestation in Indonesian children with SRNS.

Dispersal of invasive species by drifting

Drifting can be an effective way for aquatic organisms to disperse and colonise new areas. Increasing connectivity between European large rivers facilitates invasion by drifting aquatic macroinvertebrates. The present study shows that high abundances of invasive species drift in the headstream of the river *** villosusandChelicorophium curvispinumrepresented up to 90% of the total of drifting macroinvertebrates. Drift activity shows seasonal and diel patterns. Most species started drifting in spring and were most abundant in the water column during the summer period. Drift activity was very low during the winter period. Diel patterns were apparent; most species, includingD. villosus, drifted during the night. Drifting macroinvertebrates colonised stony substrate directly from the water column.D. villosusgenerally colonised the substrate at night, while higher numbers ofC. curvispinumcolonised the substrate during the day. It is very likely that drifting functions as a dispersal mechanism for crustacean invaders. Once waterways are connected, these species are no longer necessarily dependent on dispersal vectors other than drift for extending their distribution range.

Cavity enhanced absorption spectroscopy for N_2O detection at 2.86 μm using a continuous tunable color center laser

The cavity enhanced absorption technique is applied to N2O detection around 2.86 μm using a continuous-wave color center laser. A high-finesse triangular ring cavity is used in this technology. Transmission through the cavity is obtained by jittering the cavity-length with a piezo on one of the cavity mirrors. A minimum detectable absorption coefficient of 2 × 10-6 cm-1 is achieved with a mirror reflectivity of 99.24%, corresponding to a N2O detection limit of 600 parts per billion.
