Effect of miR-513a-5p on Etoposide-stimulating B7-H1 Expression in Retinoblastoma Cells

This study investigated the effect of etoposide,an anticancer chemotherapy drug,on B7-H1 expression in retinoblastoma(Rb) cells and the role of miR-513a-5p in the *** cells were divided into control and etoposide *** the etoposide group,cells were treated with etoposide at different concentrations(2.5,5,10,20 and 40 μg/mL) for 24 *** given no treatment of etopside served as *** transcription polymerase chain reaction(RT-PCR),fluorescence quantitative PCR and flow cytometry were performed to measure the mRNA and protein expression of B7-H1 in Rb *** mRNA expression of miR-513a-5p in Rb cells before and after etoposide treatment was also detected by fluorescence quantitative *** miR-513a-5p mimics and the miR-513a-5p inhibitor were transfected into Rb cells separately,and fluorescence quantitative PCR and flow cytometry were used to detect the effect of the miR-513a-5p mimics or inhibitor on B7-H1 ***5.2 was employed to predict the miR-513a-5p binding sites in the 3’-untranslated region of B7-H1 *** reporter plasmids carrying this site were prepared and transfected into Rb cells and luciferase activity *** results showed that etoposide stimulated the mRNA and protein expression of B7-H1 in Rb cells,which reached a maximal level after treatment with 5 μg/mL etoposide(P<0.05).However,miR-513a-5p expression was decreased in Rb cells after etoposide *** the miR-513a-5p inhibitor was added,B7-H1 expression was increased with the concentration of the miR-513a-5p inhibitor(P<0.05).Moreover,B7-H1 expression was decreased gradually with the concentration of the miR-513a-5p mimics increased(P<0.01).Additionally,the miR-513a-5p mimics were found to inhibit the luciferase *** was concluded that etoposide can promote B7-H1 expression in Rb cells,which may be associated with *** promoting effect of etoposide on B7-H1 expression can be reversed by miR-513a-5p ***-

Comparison of two different methods of preoperative marking for toric intraocular lens implantation： bubble marker versus pendulum marker

AIM： To compare the accuracy of two different methods of preoperative marking for toric intraocular lens（IOL） implantation, bubble marker versus pendulum marker, as a means of establishing the reference point for the final alignment of the toric IOL to achieve an outcome as close as possible to emmetropia.·METHODS： Toric IOLs were implanted in 180 eyes of110 patients. One group（55 patients） had preoperative marking of both eyes done with bubble marker（ASICO AE-2791TBL） and the other group（55 patients） with pendulum marker（Rumex 誖 3-193）. Reference marks were placed at 3-, 6-, and 9-o＇clock positions on the limbus. Slit-lamp photographs were analyzed using Adobe Photoshop（version 7.0）. Amount of alignment error（in degrees） induced in each group was measured.· RESULTS： Mean absolute rotation error in the preoperative marking in the horizontal axis was 2.42±1.71 in the bubble marker group and 2.83 ±2.31 in the pendulum marker group（P =0.501）. Sixty percent of the pendulum group and 70% of the bubble group had rotation error ≤3（P =0.589）, and 90% eyes of the pendulum group and 96.7% of the bubble group had rotation error ≤5（P =0.612）.·CONCLUSION： Both preoperative marking techniques result in approximately 3 of alignment error. Both marking techniques are simple, predictable, reproducible and easy to perform.

Quantitative analysis of retinal vasculature in normal eyes using ultra-widefield fluorescein angiography

AIM:To quantify the area and density of retinal vascularity by ultra-widefield fluorescein angiography(UWFA).METHODS:In a retrospective study,UWFA images were obtained using an ultra-widefield imaging device in 42 normal eyes of 42 *** and peripheral steered images were used to define the edge of retinal vasculature by a certified *** length from the center of the optic disc to the edge of retinal vascularity(RVL)in each quadrant and the total retinal vascular perfusion area(RVPA)were determined by the grader using OptosAdvance *** density of retinal vascularity(RVD)was quantified in different zones of central-steered images using Image J ***:Among 42 healthy eyes,the values for mean RVL in each quadrant were 19.007±0.781 mm(superior),18.467±0.869 mm(inferior),17.738±0.622 mm(nasal)and 24.241±1.336 mm(temporal).The mean RVPA was 1140.117±73.825 mm^(2).The mean RVD of the total retina was 4.850%±0.638%.RVD varied significantly between different retina zones(P<0.001),and significant differences existed in the RVD values for total retinal area in patients over 50 years old compared to those under 50 years old(P=0.033).No gender difference was ***:The UWFA device can be a promising tool for analyzing the overall retinal vasculature and may provide a better understanding of retinal vascular morphology in normal *** may be related to lower RVD.

Risk factors of regression and undercorrection in photorefractive keratectomy: a case-control study

AIM: To determine risk factors of regression and undercorrection following photorefractive keratectomy (PRK) in myopia or myopic astigmatism. METHODS: A case -control study was designed in which eyes with an indication for re-treatment (AT) were defined as cases; primary criteria for RT indication, as assessed at least 9mo postoperatively, included an uncorrected distance visual acuity (UDVA) of 20/30 or worse and a stable refraction for more than 3mo. Additional considerations included optical quality symptoms and significant higher order aberrations (HOAs). Controls were chosen from the same cohort of operated eyes which had complete post-operative follow up data beyond 9mo and did not need RT. The cohort included patients who had undergone PRK by the Tissue-Saving (TS) ablation profile of Technolas 217z100 excimer laser (Bausch & Lomb, Rochester, NY, USA). Mitomycin C had been used in all of the primary procedures. RESULTS: We had 70 case eyes and 158 control eyes, and they were comparable in terms of age, sex and follow -up time (P values: 0.58, 1.00 and 0.89, respectively). Pre-operative spherical equivalent of more than -5.00 diopter (D), intended optical zone (OZ) diameter of less than 6.00 mm and ocular fixation instability during laser ablation were associated with AT indications (all P values -5.00 D), smaller OZ (<6.00 mm) and unstable fixation during laser ablation of PRK for myopia and myopic astigmatism were found to be strong predictors of undercorrection and regression.

Implantation of trabecular micro-bypass stent using a novel “landing strip” technique

AIM：To describe a novel technique of creating a landing strip within the trabecular meshwork to guide trabecular micro-bypass stent（iStent） implantation in patients who underwent ***：Thirty-four eyes from 30 patients who underwent i Stent implantation after phacoemulsification from May 2014 to February 2015 were included in our retrospective study. All iStents were implanted via the ＂landing strip＂ technique. A 25-gauge microvitreoretinal blade was used to bisect the trabecular meshwork to less than 1 clock-hour, effectively creating a landing strip. The iStent applicator was pressed along the landing strip and then the stent was released into the trabecular meshwork. RESULTS：Of the 34 eyes with iStent implantation, 27（79.4%） eyes had primary open-angle glaucoma, 6（17.6%） eyes had pseudoexfoliation glaucoma, and 1（2.9%） eye had ocular hypertension. At 6-month follow-up（n=17）, the mean number of hypotensive medications decreased from 2.2±1.2 at baseline to 0.8±1.3（P=0.05） and mean intraocular pressure decreased from 19.7±4.1 mm Hg at baseline to 16.7±2.1 mm Hg（P=0.58）. Two eyes（5.9%） required subsequent trabeculectomy. CONCLUSION：The ＂landing strip＂ technique appears to be an effective way to assist with iS tent implantation.

Clinical analysis of tear film after lamellar keratoplasty

AIM: To study the tear film stability after lamellar keratoplasty METHODS: Five female and eight male patients with lamellar keratoconus, aged from 18 to 32, were involved. After lamellar keratoplasty, Schirmer I test (S I t), tear break-up time (BUT) test, fluorescein staining test were used to judge the effect of the surgery at different time point. RESULTS: The S 1 t were greatly increased in 7 clays post operation (11.86+/-2.28 -25.14+/-1.97, 19.86+/-1.61) (P0.05). The mean break-up time decreased in 7 days post operation (5.00+/-1.31 -2.71+/- 0.18, 2.57+/- 0.20, 2.71+/- 0.36, 2.43+/- 0.20) (P <0.05). The mean scores of fluorescence increased post-operatively (0.14+/- 0.14 - 8.00+/- 0.00, 8.00+/- 0.00, 8.00+/- 0.00, 7.57+/- 0.20) (P<0.01). CONCLUSION: Lamellar keratoplaty influence the tear film stability, artificial tears and improving corneal epithelium cured medicine should be used after surgery.

Factors affecting visual outcome of myopic choroidal neovascularization treated with verteporfin photodynamic therapy

AIM: To evaluate the visual outcomes of choroidal neovascularization (CNV) secondary to pathological myopia and the impact of novel risk factors affecting the final visual outcome. ·METHODS: Interventional case series of 18 consecutive patients with pathological myopia treated with photodynamic therapy (PDT). Inclusion criteria were spherical equivalent -6D or worse or features of pathological myopia on retinal examination. The main outcome measure was final best -corrected visual acuity (BCVA). ·RESULTS: Of 18 eyes, 13 (72.2%) avoided moderate visual loss (≥3 lines of LogMAR BCVA) and 5 eyes (27.8%) improved by at least 1 line after 1 year. Patients with LogMAR BCVA ≤0.3 (Snellen equivalent 20/40) at one year were younger than those with BCVA >0.3 (mean age 39.0 vs 61.6 years, P =0.001). A higher proportion of eyes with greatest linear dimension (GLD) of ≤1 000μm avoided moderate visual loss (100% vs 50%, P =0.026). Among patients who were treated within 2 weeks of visual symptoms, 88.9% avoided the loss of 3 or more lines compared to 55.6% for those who presented later. The mean improvement in LogMAR BCVA of those with GLD ≤1 000μm was +0.12 compared to a loss of 0.55 LogMAR units for those with GLD >1 000μm (P =0.02). Visual outcomes were not associated with gender or refractive error.·CONCLUSION: Good visual outcome in myopic CNV is associated with younger age, smaller lesion size and earlier initiation of treatment. These factors are relevant for ophthalmologists considering treatment options for myopic CNV.

Wave-Front Aberrations of Cornea and Crystalline Lens

The aberrations for anterior corneal surface, posterior corneal surface and complete eyes are measured and calculated. The aberrations for the crystalline lens are obtained by subtracting the aberrations in cornea from that in complete eyes. It is shown that the combination between the crystalline lens and the cornea can be either a compensatory or an additive process. The combination between the anterior and the posterior corneal surfaces on the aberrations is complicated, compensatory or additive. The anterior corneal surface contributes mainly lower-order aberrations （astigmatism） of images, while the posterior corneal surface and the crystalline lens contribute mainly higher-order aberrations.

Age-related macular degeneration(AMD)is a leading cause of central vision loss *** progression of dry AMD from early to intermediate stages is primarily characterized by increasing drusen formation and adverse impact on outer retinal *** stage AMD consists of either geographic atrophy(GA),the non-exudative(dry)AMD subtype,or choroidal neovascularization,the exudative(wet)AMD *** is characterized by outer retinal and choroidal atrophy,specifically the photoreceptor layer,RPE,and *** remains to be discovered regarding the pathogenesis of AMD progression and subsequent development of *** the functionality of all three layers is closely linked,the temporal sequence of events that end up in atrophy is important in the understanding of the pathogenic pathway of the *** advent of OCTA,and particularly of swept-source technology,has allowed for depth-resolved imaging of retinal vasculature and the *** the use of OCTA,recent studies demonstrate that choriocapillaris flow alterations are closely associated with the development and progression of *** changes may even possibly offer predictive value in determining progression of *** article reviews studies demonstrating choriocapillaris changes in dry AMD and summarizes the existing literature on the potential role of the choriocapillaris as a key factor in the pathogenesis of AMD.
