Identification and Cloning of Differentially Expressed Genes Involved in the Interaction Between Potato and Phytophthora infestans using a Subtractive Hybridization and cDnA-AFLP Combinational Approach

Using a subtractive hybridization （SH）/cDnA-AFLP combinational approach, differentially expressed genes involved in the potato-Phytophthora infestans interaction were identified. these included genes potentially controlling pathogenesis or avr genes in P. infestans as well as those potentially involved in potato resistance or susceptibility to this pathogen. Forty-one differentially expressed transcript, derived fragments （tDFs）, resulting from the interaction, were cloned and sequenced. two tDFs, suggested as potential pathogenicity factors, have sequence similarity to n-succinyl diaminopimelate aminotransferase and a transcriptional regulator, tetr family gene, respectively. two other tDFs, suggested as potential avr genes, have sequence similarity to an ESt sequence from Avr41Cf.41Avr91Cf- 9 and a P. infestans avirulence-associated gene, respectively. Genes＇ expression and origin were confirmed using Southern blots, northern blots and qrt-PCr, he., potential resistance gene DL81 was induced at 12 hpi in the moderately resistant cultivar, whereas it was down-regulated as early as 6 hpi in the susceptible cultivar. On the other hand, DL21 was induced at 6 hpi （3.38-fold） in response to the highly aggressive isolate （US8） and strongly up-regulated thereafter （25.13-fold at 120 hpi.）, whereas it was only slightly up-regulated in response to the weakly aggressive isolate US11 （3.82-fold at 96 hpi）, suggesting its potential involvement as a susceptibility gene.

the benefits of regular physical activity are well ***,few studies have examined the effectiveness of integrating physical activity(PA)into curricula within a post-secondary *** investigate the incorporation of PA into medical curriculum,we developed a series of optional exercise-based review sessions designed to reinforce musculoskeletal(MSK)anatomy course *** synchronous sessions were co-taught by a group fitness instructor and an anatomy *** fitness instructor would lead students through both strength and yoga style exercises,while the anatomy instructor asked questions about relevant anatomical structures related to course material previously *** the sessions,participants were asked to evaluate the classes on their self-reported exam preparedness in improving MSK anatomy knowledge,PA levels,and mental *** participants completed surveys;a majority agreed that the classes increased understanding of MSK concepts(90.0%)and activity levels(97.7%).Many(70.0%)felt that the classes helped reduce *** majority of respondents(90.0%)agreed that the classes contributed to increased feelings of social ***,medical students saw benefit in PA based interventions to supplement MSK course *** with increasing activity levels and promoting health behaviours,integrating PA into medical curriculum may improve comprehension of learning material,alleviate stress and foster social connectivity among medical students.

A competitive finite-difference method will be constructed and used to solve a modified deterministic model for the spread of herpes simplex virus type-2 (HSV-2) within a given population. the model monitors the transmission dynamics and control of drug-sensitive and drug-resistant HSV-2. Unlike the fourth-order runge-Kutta method (rK4), which fails when the discretization parameters exceed certain values, the novel numerical method to be developed in this paper gives convergent results for all parameter values.

the insertion losses of silicon oxynitride (SiOn) waveguides have been measured in the 1550 nm wavelength *** waveguide structure consisted of a 2.0μm SiOn waveguide core with a refractive index of 1.50, a 0.5μm SiO2 uppercladding and a 5.0μm SiO2 lower cladding with a refractive index of 1.45. It was found that the wavelength-dependentinsertion losses of the waveguide were greatly reduced by annealing, and the loss was decreased more than 5.7 dB/cm at1550 nm after annealing at optimum conditions. the former was attributed to the reduction of the absorption caused by n-H and Si-H vibration modes, and the latter was due to the improvement of the interface roughness and homogeneity in the waveguides after annealing.

Cowpea protein isolate(CPI)was subjected to various dry and wet heat pretreatments followed by sequential digestion with pepsin and pancreatin;the undigested residues were isolated as the indigestible cowpea proteins(ICPs).All the ICPs exhibited in vitro bile acid-binding capacity but ICP from the slow cooling-induced gelation had the highest yield(68%)and was used for rat feeding experiments to determine effect on plasma total cholesterol(tC).Groups consisting of 3 male and 3 female Sprague-Dawley rats each were fed hypercholesterolemic diets that contained casein only or casein that was partially substituted with ICP of CPI for 6 *** showed diet that contained 5%(w/w)ICP was more effective in preventing tC increase(1.8 mmol/L)when compared to increases of 9.34 and 4.15 mmol/L for CPI and casein only diets,respectively.

In grassland ecosystems,burrowing mammals create disturbances,providing habitat for animal species and increasing plant community *** investigated whether seedling assemblages on richardson's ground squirrel Urocitellus richardsonii mounds result from seed rearrangement or environmental changes that favor germination of certain species over *** test whether ground squirrels rearrange the seed bank by burrowing,we compared seed compositions among mounds,burrows,and undisturbed *** test whether ground squirrels change environmental conditions,we compared soil nitrate and bare ground cover on and off *** also compared seedlings that germinated on mounds with seedlings that germinated on artificial disturbances from which we removed aboveground *** nitrate and bare ground cover were significantly higher on mounds than artificial *** seed richness and abundance did not differ among mounds,burrows,and undisturbed soil,seedling richness was reduced on mounds relative to artificial *** disturbance favors seedlings that can capitalize on bare ground availability but are less able to immobilize nitrate,as opposed to perennial species that immobilize more nitrate but take longer to *** results suggest that richardson's ground squirrels act as ecosystem engineers,although future research following succession on ground squirrel mounds is necessary to understand how they influence plant communities past the seedling stage.
