External quality Assurance Scheme in a National Reference laboratory for HIV Testing in South India

As the complexity of the chemical constitution of traditional Chinese medicines (TCMs),it is now a trend to establish a method of using multi-components analysis for the effective quality control of ***,the limited availability of multiple reference substances hinders the wide popularization of this method for the routine quality control of *** an easy-available single component which contained in the analyte as internal/external standard to determine multiple analogues should be an optional *** this paper,we select Rhubarb rhizome as an example,using emodin,an easy-available active component,as external standard to determine directly its own content in Rhubarb,meanwhile as internal standard to determine simultaneously the other six *** with the results obtained by external standard method,this alternative method was found in our laboratory to have no significant difference verified by the F-test(p = 95%,n = 6).However,on account of the weak robustness caused by some fluctuation of relative response factors from different laboratories,such a method can only serve as an expedient measure for solving the lack of chemical standards,and first relative response factors have to be determined before this method can be used without reference *** the corresponding chemical standards being available as well as being acceptable cost/effective,the external standard method for simultaneous determination of multiple components(SDMC) of TCMs will prevail.

Background: The Department of Experimental Medicine functions as the National Reference laboratory (NRL) for HIV testing covering 11 Medical Colleges (State Reference Laboratories SRL) & 723 subcenters i.e. Integrated Counseling & Testing Centres and Blood Banks. The External quality Assurance Scheme (EQAS) in NRL implements quality control (QC) Testing, Proficiency panel testing and training programs. Materials & Method: 9419 samples (4393 HIV negative/5026 HIV positive) were tested for QC. All the samples were tested using HIV rapid test (CombAids) and HIV positives alone were tested using Tridot and EIA Comb. The QC samples consisted of 20% negative and all positives. All the 723 subcenters were provided with 5 coded plasma samples (3 reactive & 2 negative) for proficiency testing using rapid tests. The aliquot panel (500 μl) were provided twice a year for testing to monitor the laboratory performance. Results: Out of 9419 samples tested for QC, 9371 (99.49%) reported correct results and 48 (0.50%) discordant results. Out of 48 samples 26 (0.27%) were false positives and 22 (0.23%) false negative. Mislabeling, sample contamination, leaking vials, transcriptional errors, tests that were not performed correctly were identified. For proficiency testing 91.8% reported test results. 645 (97.13%) reported correct results & 19 (2.86%) incorrect results. Out of 19 samples 7 (1.05%) were false positive & 12 (1.80%) were false negative. Hands on training were provided and the 19 discordant centers reported correct results on retesting. Conclusion: Significant progress in establishing a well coordinated HIV laboratory network of NRL and SRLs had been developed. However the HIV testing and quality Assurance needs to be strengthened towards certification.

The quality variety wines which are produced in specified regions of Slovakia are mostly watched category of wine in European Union. quality of the measurement for appropriate parameter in wine sample is given by uncertainty. Therefore, protocol of analysis has to have an uncertainty of measurement and should be made in accredited laboratory. In Slovakia, there are six wine growing regions （Tokaj, east Slovakia region, south Slovakia region, middle Slovakia region, Nitriansky region and Little-Carpathian region） where the quality wines with protected designation of origin are produced. Central controlling and Testing Institute in Agriculture in Bratislava is a state organization, which has an accredited laboratory for testing wine products before introduction into the market. This article presents the results of the authors＇ measurements in proficiency testing FAPAS （food analysis performance assessment scheme） which were organized by Central Science laboratory in York （England） in 2011. The authors add also interlaboratory comparison in 2012 between Slovak accredited laboratories. The results show very good agreement. quality control in the wine sector presents very important feature in the market, therefore, using certified reference materials and checking the accredited laboratories with interlaboratory comparisons is very helpful. Testing of wine in the authors＇ laboratory is performed by EN ISO/IEC： 17025. The results show that z-score lies not outside the range ±2 and this is a chance that the authors＇ results are actually acceptable.

Background: Groundwater is an important source of water. Since the control and removal of pollution are expensive, it is essential to identify the possible sources of contamination and to correctly classify groundwater on the basis of its intrinsic and integrated vulnerability. Objectives: To group ground water chemical ions and heavy metals parameters into similar groups. Method: The investigation made use of standard analytical procedures. All sampling, conservation, transportation and analysis followed standard procedures described in APHA (2012). To prevent degradation of the organic substances, all obtained samples were transferred to the laboratory, while kept in an icebox. Results: Sampling records from the same area are generally assigned to the same cluster during hierarchical cluster analysis (HCA). The cluster diagram shows the grouping of the heavy metal in the study area during wet and dry seasons. It reveals that 5 distinct clusters were identified for wet season and 4 clusters were identified during dry season. Also, it reveals that 5 distinct clusters were identified for wet season and for dry season, 4 distinct clusters were identified. Conclusion: The findings of this study are significant for policymakers and agencies in terms of dealing with the issues identified to enhance sustainable livelihood practices in the oil rich Niger Delta region of Nigeria. Therefore, decision-makers should take proper initiatives to get local people aware of the endangered zones before use, as drinking water is key to good health. Similarly, multinational oil companies will find it useful in their quest for viable social corporate responsibility and remediation plans in their respective host communities. The method proved to be a useful and objective tool for environmental planning.

Objective:To analyze the registration information of Corona Virus Disease 2019(COVID-19)related clinical trials from all the clinical trial registry accepted by the International Committee of Medical Journal Editors(ICMJE).Methods:All the database of ICMJE-accepted clinical trial registry platform were searched for COVID-19 related clinical trials that registered from December 8,2019 to February 19,***:All the database totally contained data of 209 COVID-19 related clinical trials,including 66(31.6%)traditional Chinese medicine(TCM)related studies and 143(68.4%)non-TCM related studies.176(84.2%)clinical trials were registered in ChiCTR,as 33(15.8%)were registered in *** were 152(72.7%)intervention trials,including 9(4.3%in total)preventive trials,136(65.1%in total)therapeutic trials and 7(3.3%in total)convalescent ***:At the present stage,clinical studies are still aiming at exploring the effective treatment of COVID-19,and the existing treatments need more high-quality clinical evidence *** Chinese medicine may have played an important role in all stages of the epidemic preventing and control,and the enthusiasm of carrying out scientific research has also been greatly *** relevant policies of clinical trial registration should be popularized among researchers,including traditional Chinese medicine *** the methodological quality of clinical trials’design urges improving.

[Objectives]The effects of water chestnut powder on the gel properties and quality of silver carp surimi were investigated.[Methods]The surimi gel was prepared by adding 0%,1%,2%,3%,4%and 5%of water chestnut powder to the surimi of silver *** gel properties,water-holding capacity,cooking loss,whiteness value,puncture and texture profile analysis(TPA)indexes of surimi products were determined to assess the effects of adding different amounts of water chestnut powder on surimi gel.[Results]The results showed that the gel strength,breaking force and depression distance of surimi products increased first and then decreased with the increase of the addition of water chestnut *** with the control group,the hardness and chewiness of surimi gel in TPA could be significantly improved by adding water chestnut *** the addition of water chestnut powder was 2%,the maximum water-holding capacity was 83.68%;the cooking loss rate was the lowest;and the whiteness value was the *** 2%of water chestnut powder could significantly improve the gel properties of silver carp surimi and obtain a good gel product.[Conclusions]This study provides a scientific basis for the effective utilization of water chestnut resources and the development of new fish surimi products.

Impact of Crude Oil Storage Tank Emissions and Gas Flaring on Air/Rainwater quality and Weather Conditions in Bonny Industrial Island, Nigeria

This study investigated the effects of gaseous emissions from crude storage tank and gas flaring on air and rainwater quality in Bonny Industrial Island. Ambient air quality parameters, rainwater and weather parameters were collected at 60 m, 80 m, 100 m, 200 m and control plot for 4 weeks at the Bonny. Rainwater parameters were investigated using standard laboratory tests. Data analyses were done using Analysis of variance, pairwise t-test and Pearson’s correlation statistical tools. Results show that emission rates, volatile organic compound (VOC) noise and flare temperature decreased with increasing distance from flare points and crude oil storage tanks. Findings further revealed the emission rates varied significantly with distance away from the gas flaring point (F = 6.196;p = 0.004). The mean concentration of pollutants between gas flare site and crude oil storage tank showed that CO (0.02 ± 0.001 - 0.002 ±0.001), SPM (0.011 ± 0.001 - 0.01 ± 0.001), VOC (0.005 ± 0.001 - 0.01 ± 0.001) and NO2 (0.04 ± 0.001 - 0.005 ± 0.000) had significant variations (p > 0.05) with CO, O3 and NO2 having higher concentrations at the gas flare site while SPM, and VOC were higher around the crude oil storage tank site. Wind turbulence was higher around the gas flaring point (4.93 TKE) than the crude oil storage tank (4.55 TKE). Similarly, there was significant variation in the sun radiation, precipitation, and wind speed caused by gas flaring (1582.25 w/m2, 436.25 mm, 0.53 m/s) and crude oil storage tank (1536.25 w/m2, 3.91.41 mm, 0.51 m/s). There were also significant variations in flared temperature (F = 22.144;p = 0.001);NO2 (F = 8.250;p = 0.001), CO (F = 6.000;p = 0.004) and VOC (F = 5.574;p = 0.006) with distance from the gas flaring point. Various studies have been conducted in the past by researchers who have proposed diverse solutions to control the premature degradation noted on flexible pavements. It turns out that the binder content remains a parameter so far not mastered and its optimization remains an imperative to produce a quality mix capable of meeting the technical and economic requirements sought in a formulation study. This article mainly aimed to optimize the hydrocarbon binder content of a 0/14 granular class semi-granular bituminous concrete using the Marshall and Duriez method. In the first part of the study, the different constituents were characterized according to the standards in force. The second allowed to describe the methodology used to determine the mixture and calculated the theoretical binder content. A theoretical mixture, entering the reference zone was defined at first. Following the characterization of the mixture in the laboratory, the granulometry curve of the mixture was inserted into the reference zone. This mixture was then preserved for the rest of the study. Then, it was theoretically determined bitumen contents ranging from 5.13% to 5.75% for wealth modules ranging from 3.3% to 3.7%. Thus, the Marshall method was used to characterize the performance of the mixture as a function of the binder content. The results obtained led to the selection of two grades of binders 5.13% and 5.28% for the study of water resistance using the Duriez method. On the basis of the results obtained on the Duriez test, the mixture with a binder content of 5.28% satisfies a good part of the criteria selected. This content corresponds to a wealth module of 3.4%.

A laboratory information system (LIS) established in a microbiology department has the potential to play an important role in the quality of microbiology data such as culture of blood, urine, stool, pus swab samples etc. Such data could be effectively utilised to measure the burden of antimicrobial resistance among patients presented to various hospitals and clinics with an episode of an infectious illness of bacterial origin. A variety of clinical and epidemiological investigations are conducted using culture data and the presence of an electronic system such as LIS enhances such investigations and improves the reliability of measures of antimicrobial resistance owing to improved data quality as well as completeness of data gathered as opposed to paper based system. Therefore to improve surveillance of antimicrobial resistance in South Africa, there is a need to reinforce the functionality of the LIS in both public and private microbiology laboratories as this will help to improve internal quality control methodologies.
