Comprehensive analysis of the gut microbiome and posttranslational modifications elucidates the route involved in microbiota-host interactions

The gut microbiome interacts with the host to maintain body homeostasis,with gut microbial dysbiosis implicated in many diseases.However,the underlying mechanisms of gut microbe regulation of host behavior and brain functions remain unclear.This study aimed to elucidate the influence of gut microbiota on brain functions via post-translational modification mechanisms in the presence or absence of bacteria without any stimulation.We conducted succinylome analysis of hippocampal proteins in germ-free(GF)and specific pathogen-free(SPF)mice and metagenomic analysis of feces from SPF mice.These results were integrated with previously reported hippocampal acetylome and phosphorylome data from the same batch of mice.Subsequent bioinformatics analyses revealed 584 succinylation sites on 455 proteins,including 54 up-regulated succinylation sites on 91 proteins and 99 down-regulated sites on 51 proteins in the GF mice compared to the SPF mice.We constructed a panoramic map of gut microbiota-regulated succinylation,acetylation,and phosphorylation,and identified cross-talk and relative independence between the different types of post-translational modifications in modulating complicated intracellular pathways.Pearson correlation analysis indicated that 13 taxa,predominantly belonging to the Bacteroidetes phylum,were correlated with the biological functions of post-translational modifications.Positive correlations between these taxa and succinylation and negative correlations between these taxa and acetylation were identified in the modulation of intracellular pathways.This study highlights the hippocampal physiological changes induced by the absence of gut microbiota,and proteomic quantification of succinylation,phosphorylation,and acetylation,contributing to our understanding of the role of the gut microbiome in brain function and behavioral phenotypes.

Fractional Angular Momentum of an Atom on a Noncommutative Plane

The mechanism of obtaining the fractional angular momentum by employing a trapped atom which possesses a permanent magnetic dipole moment in the background of two electric fields is reconsidered by using an alternative method. Then, we generalize this model to a noncommutative plane. We show that there are two different mechanisms,which include cooling down the atom to the negligibly small kinetic energy and modulating the density of electric charges to the critical value to get the fractional angular momentum theoretically.

Polarization visualization for low-irradiance regions by perceptually uniform color space

Currently,polarization visualization strategies are accomplished by mapping polarization information into a perceptually uniform color appearance model CAM02-UCS.However,the deviation of the CAM02-UCS space from the lightness prediction results in an inaccurate match between the polarization information and the perceptual information.In this paper,we propose a novel polarization visualization strategy based on the perceptual uniform space Jzazbz.The polarization visualization be completed by placing the polarization information into the lightness Jz,colorfulness Cz and hue angle hz channels of the Jzazbz space.The experimental results show that the proposed method can significantly improve the lightness of the low irradiance and high polarization region,hence more polarization information can be sensed by human visual system.

Therapeutics of Integrative Medicine Ameliorate Immunological Disorders of the Nervous System: A Meta-Analysis

Objective: The objective of the study is to evaluate the therapeutic efficacy of integrative medicine in the treatment of immunological disorders of the nervous system. Methods: The Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines were adopted to conduct this study, which included randomized controlled trials with a confirmed diagnosis of multiple sclerosis(MS), myasthenia gravis(MG), and Guillain–Barre syndrome(GBS), all of which were treated with integrative medicine. The effective rate, recurrent frequency, and disease score were used as the markers of outcome variables for the meta-analysis. Results: A total of 48 randomized control trials were included. The effective rates of treatment with integrative medicine were noticeably higher than those with Western medicine alone for the three diseases. The recurrence frequency for MS and the recurrence rate for MG treated with integrative medicine were reduced more than those with Western medicine alone. The Extended Disability Status Scale, acetylcholine receptor antibody, and Hughes scores for MS, MG, and GBS, respectively, treated with integrative medicine were significantly lower than those with Western medicine alone. The risks of bias in the literature evaluation showed that the quality of the included studies was not high. Conclusions: Compared to treatment with Western medicine alone, integrative medicine might ameliorate immunological disorders of the nervous system.

A Comprehensive Evaluation on the Devonian-Carboniferous Boundary Stratotype Candidate Section in Nanbiancun, Guilin, Guangxi

This paper presents a comprehensive evaluation on the Devonian-Carboniferous Boundary (DCB) sectionin Nanbiancun. Guilin. Guangxi on the basis of Palaeontological, biostratigraphical and sedimentarypetrological studies as well as microfacies analysis. REE and trace element analysis and isotopic dating. The DCB at this section conforms to the definition of this boundary and essentially measure up the criteriaand conditions for selection of the boundary stratotype. Up to 14 fossil groups are contained in the section. Thesection presents a general biostratigraphical framework with multispecies fossils zones, assemblage zones andmultiple groups of fossils and has latent preponderant conditions for magnetostratigraphic andchronostratigraphic studies. The boundary and point occur in a persistent and continuous monofacialcarbonate succession. and the section is located in a tourist city with convenient communication, where variousfacies of DCB sections are developed. allowing the correlation within a small area. So it is an ideal candidateboundary stratotype.

Effectiveness of Liu-zi-jue exercise on coronavirus disease 2019 in the patients: a randomized controlled trial

OBJECTIVE: The aim of this study was to examine the effect of Liu-zi-jue exercise on the respiratory symptoms, quality of life, and mental health of patients with mild coronavirus disease 2019(COVID-19). METHODS: A single-center randomized controlled trial was conducted with 104 patients with mild COVID-19. The patients were randomly assigned to the Liu-zi-jue plus conventional therapy group and conventional therapy group. The outcome measures included Modified Borg Dyspnea Scale(MBDS) score, Patient Health Questionnaire-9(PHQ-9) score, Fatigue Scale-14(FS-14), respiratory symptoms, and vital signs. Data were collected on the first and sixth days of hospitalization and on the discharge day. RESULTS: Repeated-measures ANOVA revealed that the whole scales all showed a downward trend in the two groups(all P < 0.05). The results of single-group effect suggested that the whole scale score in the intervention group was significantly lower than that in the control group at the sixth day of hospitalization. Compared with the control group, only the MBDS, expectoration, and FS-14 scores showed significantly lower scores at the discharge day(P < 0.001, P = 0.011, P = 0.002). Comparison within the group showed that all the variables were significantly different at the three time points with a decreasing trend(P < 0.05), except for the PHQ-9 and expectoration scores(P = 0.331, 0.052). All patients’ vital signs remained within a stable normal range throughout the hospital stay. CONCLUSION: The results suggested that Liu-zi-jue exercise as a complementary and alternative therapy showed beneficial effects on improving the symptoms(shortness of breath, fatigue, and cough), quality of life, and mental health of patients with mild COVID-19.

OBJECTIVE:To investiage the possible mechanism underlying the effect of the Jianpi Qutan Fang(健脾祛痰方,JPQT)on Atherosclerosis(AS)which is the main pathological process of most cardiovascular diseases that affect millions of adults worldwide.METHODS:In the present study,rats were fed with a high-fat-diet(HFD)with vitamin D3 for 16 weeks and were orally administered atorvastatin treatment and different doses of JPQT.Histopathological changes and ultrastructural changes in the aorta were evaluated through hematoxylin-eosin staining and transmission electron microscopy(TEM),respectively.Suppressor of cytokine signaling 1(SOCS1)/Janus kinase 1(JAK1)/signal transducer and activator of transcription 1(STAT1)signaling pathways were detected through Western blotting.RESULTS:JPQT treatment decreased the lipid levels of triglyceride,low-density lipoprotein,and cholesterol,the inflammatory cytokine levels of interleukin 1 beta(IL-1β),IL-6 and IL-8 in rat serum,but increased high-density lipoprotein and IL-10 serum levels.JPQT treatment ameliorated pathological changes in the aorta of AS model rats.Moreover,JPQT upregulated SOCS1 protein expression and down-regulated phosphorylated protein expression levels of p-JAK1 and p-STAT1.CONCLUSION:These results suggest that JPQT induces anti-atherosclerosis effects through anti-inflammatory and inhibiting JAK/STAT signaling pathways in HFD fed rats.

AIM To evaluate antihepatoma effect ofantisense phosphorothioate oligodeo-xyribonucleotides (S-ODNs) targeted to alpha-fetoprotein (AFP) genes in vitro and in nudemice.METHODS AFP gene expression was examinedby immunocytochemical method or enzyme-linked immunosorbent assay. Effect of S-ODNson SMMC-7721 human hepatoma cell growth invitro was determined using microculturetetrazolium assay. In vivo antitumor activitiesof S-ODNs were monitored by measuring tumorweight differences in treated and control micebearing SMMC-7721 xenografts. Induction of cellapoptosis was evaluated by fluorescence-activated cell sorter (FACS) analysis.RESULTS Antisense S-ODN treatment led toreduced AFP gene expression. Specificantisense S-ODNs, but not control S-ODNs,inhibited the growth of heaptoma cells in vitro.In vivo. only antisense S-ODNs exhibitedobvious antitumor activities. FACS analysisrevealed that the growth inhibition by antisenseS. ODNs was associated with their cell apoptosisinduction.CONCLUSION Antisense S-ODNs targeted toAFP genes inhibit the growth of human hepatomacells and solid hepatoma, which is related totheir cell apoptosis induction.

CsTT8 regulates anthocyanin accumulation in blood orange through alternative splicing transcription

A homologous gene of basic-helix-loop-helix AtTT8 in Arabidopsis thaliana was identified in juice sac cells of pulp tissues from blood orange(Citrus sinensis cv‘Tarocco’),which was designated as CsTT8 in this study.Additionally,the mRNA levels of TT8 with the full-length open reading frame were significantly higher in‘Tarocco’than in mutant fruit lacking pigment in pulp or peel tissues.However,an alternative splicing transcript,Δ15-TT8,with the fourth exon skipped,was also identified from transcripts different in length from that in‘Tarocco’.The mRNA levels ofΔ15-TT8 were higher in mutant fruit lacking pigment in pulp or peel tissues than in the wild type.Therefore,the TT8/Δ15-TT8 mRNA level ratio was found to be crucial for sufficient pigment in either pulp or peel tissues.TT8 from blood orange fruit demonstrated the capacity for nucleus localization and binding to other proteins.In contrast,Δ15-TT8,lacking the fourth exon,lost its ability to interact with RUBY1 and to localize at the nucleus.Using a dual luciferase reporter assay and transient overexpression in tobacco,we proved that two regulatory complexes formed by a functional TT8 with different MYB(v-myb avian myeloblastosis viral oncogene homolog)-type partners significantly promoted expression of an anthocyanin biosynthetic gene and a proton pumping gene,leading to anthocyanin and citrate production.Our findings suggest that TT8,rather than dysfunctionalΔ15-TT8,is possibly involved in modulating anthocyanin biosynthesis and its transport into vacuoles by proton gradients.However,increased mRNA levels of the dysfunctional alternative splicing transcript may act as a negative feedback to downregulate TT8 expression and limit anthocyanin accumulation in blood oranges.
