Elenoside increases intestinal motility

AIM： TO study the effects of elenoside, an arylnaphthalene lignan from Justicia hyssopifolia, on gastrointestinal motility in vivo and in vitro in rats. METHODS： Routine in vivo experimental assessments were catharsis index, water percentage of boluses, intestinal transit, and codeine antagonism. The groups included were vehicle control （propylene glycol-ethanolplant oil-tween 80）, elenoside （i.p. 25 and 50 mg/kg）, cisapride （i.p. 10 mg/kg）, and codeine phosphate （intragastric route, 50 mg/kg）. In v/tro approaches used isolated rat intestinal tissues （duodenum, jejunum, and ileum）. The effects of elenoside at concentrations of 3.2 ×10^4, 6.4 ×10^4 and 1.2 ×10^3 mol/L, and cisapride at 10^6 mol/L were investigated. RESULTS： Elenoside in vivo produced an increase in the catharsis index and water percentage of boluses and in the percentage of distance traveled by a suspension of activated charcoal. Codeine phosphate antagonized the effect of 25 mg/kg of elenoside. In vitro, elenoside in duodenum, jejunum and ileum produced an initial decrease in the contraction force followed by an increase. Elenoslde resulted in decreased intestinal frequency in duodenum, jejunum, and ileum. The in vitro and in vivo effects of elenoside were similar to those produced by cisapride. CONCLUSION： Elenoside is a lignan with an action similar to that of purgative and prokinetics drugs. Elenoside, could be an alternative to cisapride in treatment of gastrointestinal diseases as well as a preventive therapy for the undesirable gastrointestinal effects produced by opioids used for mild to moderate pain.

A hydrogel based on chitosan,collagen,hydroxypropyl-c-cyclodextrin and polyethylene glycol was developed and *** incorporation of nano-hydroxyapatite and pre-encapsulated hydrophobic/hydrophilic model drugs diminished the porosity of hydrogel from 81.6262.25%to 69.9863.07%.Interactions between components of hydrogel,demonstrated by FTIR spectroscopy and rheology,generated a network that was able to trap bioactive components and delay the burst *** thixotropic behavior of hydrogel provided adaptability to facilitate its implantation in a minimally invasive *** profiles from microspheres included or not in hydrogel revealed a two-phase behavior with a burst-and a controlled-release *** same release rate for microspheres included or not in the hydrogel in the controlled-release period demonstrated that mass transfer process was controlled by internal *** diffusion coefficients,Deff,that describe internal diffusion inside microspheres,and mass transfer coefficients,h,*** contribution of hydrogel to mass transfer,were determined using‘genetic algorithms’,obtaining values between 2.64·10^(-15) and 6.67·10^(-15)m^(2)/s for Deff and 8.50·10^(-10) to 3.04·10^(-9) m/s for *** proposed model fits experimental data,obtaining an R2-value ranged between 95.41 and 98.87%.In vitro culture of mesenchymal stem cells in hydrogel showed no manifestations of intolerance or toxicity,observing an intense proliferation of the cells after 7 days,being most of the scaffold surface occupied by living cells.

Advanced strategies for marine antifouling based on nanomaterial-enhanced functional PDMS coatings

Marine biofouling seriously affects human marine exploitation and transportation activities, to which marine antifouling(AF) coatings are considered to be the most cost-effective solution. Since the mid-20th century, human beings have dedicated their efforts on developing AF coatings with long cycle and high performance, leading to a large number of non-target organisms? distortion, death and marine environmental pollution. Polydimethylsiloxane(PDMS), is considered as one of the representative environment-friendly AF materials thanks to its non-toxic, hydrophobic, low surface energy and AF properties. However, PDMS AF coatings are prone to mechanical damage, weak adhesion strength to substrate, and poor static AF effect, which seriously restrict their use in the ocean. The rapid development of various nanomaterials provides an opportunity to enhance and improve the mechanical properties and antifouling properties of PDMS coating by embedding *** on our research background and the problems faced in our laboratory, this article presents an overview of the current progress in the fields of PDMS composite coatings enhanced by different nanomaterials, with the discussion focused on the advantages and main bottlenecks currently encountered in this field. Finally, we propose an outlook, hoping to provide fundamental guidance for the development of marine AF field.

Recurrence of inflammatory pseudotumor in the distal bile duct: Lessons learned from a single case and reported cases

Inflammatory myofibroblastic tumors （IMTs） or inflammatory pseudotumors （IPs） have been extensively discussed in the literature. They are usually found in the lung and upper respiratory tract. However, reporting of cases involving the biliopancreatic region has increased over recent years. Immunohistochemical study of these lesions limited to the pancreatic head or distal bile duct seems to be compatible with those observed in a new entity called autoimmune pancreatitis, but usually intense fibrotic reaction （zonation） predominates producing a mass. When this condition is limited to the pancreatic head, the common bile duct might be involved by the inflammatory process and jaundice may occur often resembling adenocarcinoma of the pancreas. We have previously reported a case of IMT arising from the bile duct associated with autoimmune pancreatitis which is an extremely rare entity. Four years after Kaush-Whipple resection, radiological examination on routine follow-up revealed a tumor mass, suggesting local recurrence. Ultrasound-guided FNA confirmed our suspicious diagnosis. This present case, as others, suggests that persistent follow-up is necessary in order to prevent irreversible liver damage at this specific location.

Optimized combinations of statins and azoles against Acanthamoeba trophozoites and cysts in vitro

Objective: To evaluate the combination of several statins(atorvastatin, fluvastatin and simvastatin) and azoles(voriconazole, posaconazole and itraconazole) against Acanthamoeba spp. Methods: The efficiency of the different drug combinations against the trophozoite stage of different Acanthamoeba strains were evaluated by Alamar Blue assay. Effect on the cyst stage was observed by inverted microscope. Cytotoxicity of combinations of azoles and statins was evaluated by measuring the release of lactate dehydrogenase from a murine macrophage cell line. Results: Combinations of any of the tested statins and voriconazole or posaconazole were more efficient in inhibiting Acanthamoeba compared to statins or azoles individually. The drug combinations at the combined inhibitory concentrations 50% showed lower toxicity compared to that of the compounds ***: The combinations of statins together with voriconazole and posaconazole are more efficient than these drugs alone, and these combinations have lower cytotoxicity in mammalian cell lines.

Hydrogeochemical and isotopic assessment of the origin of NO_(3)^(−)and N-NH_(3)contents in the aquifer located in a closed lacustrine volcano-sedimentary basin in the metropolitan area of Mexico City

To explain the presence and spatial distribution of NO_(3)^(−)and N-NH_(3)in the Aquifer of the Metropolitan Area of Mexico City(AMAMC),a hydrogeochemical and isotopic analysis using^(13)C DIC(as well as the stable isotopes^(18)O and^(2)H)in groundwater was *** aquifer is located in an old closed lacustrine volcano-sedimentary basin;some wells hosted in the semi-confined zone contain high N-NH_(3)concentrations,while others present NO_(3)^(−)contents in the recharge zones(hosted in an oxidizing environment).In this study,a change in the isotopic signature(primarily in^(18)O and^(2)H)was observed from the recharge zones to the basin center in some of the wells with high NO_(3)^(−)concentrations,this behavior can be attributed to evaporation during the incorporation of recently infiltrated *** addition,the results for^(13)C(along with ^(2) H)in wells with the highest N-NH_(3)concentrations exhibited an atypically broad range of *** indicated the occurrence of hydrogeochemical and/or biochemical processes in the aquifer(in an oxidizing or reducing environment),such as organic degradation,bacterial decomposition(primarily in the ancient lake Texcoco and which acts as a natural sink for carbon,nitrogen,sulfur,and phosphorus),besides rock weathering and dissolution,which may be responsible for a very marked isotopic modification of the^(13)C(and,to a lesser extent,2 H).Methanotrophic bacterial activity and methanogenic activity may be related to N-NH_(3)removal processes by oxidation and residual water incorporation respectively,whereas the increase in the NO_(3)^(−)content in some wells is due to the recent contribution of poor-quality water due to contamination.

Temporal dynamics of shallow subtidal meiobenthos from a beach in Tenerife (canary islands, northeast Atlantic Ocean)

A shallow subtidal （3 m deep） meiofaunal assemblage in Los Abrigos Bay, Tenerife, canary islands was sam- pled during May 2000-April 2001. The main aims were to （1） find temporal variations in meiofaunal assem- blage structure and overall abundance, as well as in the most abundant meiofaunal species throughout the study period, and （2） identify environmental variables （sedimentary composition, organic matter content, and total nitrogen） that better explain meiofaunal assemblage structure during the study year. The most abundant species were the nematodes Siphonolaimus sp. 2 and Catanema sp., which represented 46.2% of the overall meiofaunal abundances and varied significantly throughout the study duration. Overall meio- faunal abundance and the most abundant taxonomic groups （nematodes, copepods, and oligochaetes） showed significant temporal variations during the study period. Nematodes overwhelmingly dominated during the study period, ranging from 78% in May to 97.34% in February. Multivariate analyses showed seasonality in meiofaunal community structure during the study year, with the lowest abundances in May.
