Product identification and toxicity change during oxidation of methotrexate by ferrate and permanganate in water

Accompanying an annual increase in cancer incidence,the global use of anticancer drugs has remarkably increased with their worldwide environmental prevalence and ecological *** this study,the oxidation of methotrexate(MTX),a typical anticancer drug with ubiquitous occurrence and multi-endpoint toxicity,by ferrate(VI)(Fe(VI))and permanganate(Mn(VII)))was investigated in ***(VI)exhibited a higher reactivity with MTX(93.34 M^(−1) s^(−1))than Mn(VII)(3.01 M^(−1) s^(−1)) at pH *** introduction of Cu(II)and Fe(III)at 1.0 mM improved the removal efficiency of 5.0μM MTX by 100.0μM Fe(VI)from 80%to 95%and 100%after 4 min,*** oxidized products(OPs)were identified during oxidative treatments,while OP-191 and OP-205 were characterized as specific products for Fe(VI)*** ketonization of the L-glutamic acid moiety and cleavage of the peptide bond of MTX were ***,a multi-endpoint toxicity evaluation indicated no genotoxicity,neurotoxicity,or endocrine-disrupting effects of MTX and its ***,serious developmental toxicity in zebrafish larvae was observed in the treated MTX *** on the acute and chronic aquatic toxicity prediction,OP-190,OP-192,OP-206,and OP-208 were deemed toxic or very toxic compared to harmful ***,the reduced biodegradability index from 0.15(MTX)to−0.5 to−0.2(OP-192,OP-206,and OP-468)indicated the formation of lower biodegradable ***,this study suggests that Fe(VI)and Mn(VII)oxidation are promising treatments for remediating anticancer drug-contaminated ***,the environmental risks associated with these treatments should be considered in the evaluation of water safety.

Emulsion‑based evolution of Escherichia coli for higher growth yield on D‑xylose identifies central role of cyclic AMP

D-xylose is an abundant sugar found in plant biomass and can be used as a renewable feedstock for the microbial production of diverse biofuels and ***,D-xylose metabolism is slow in many industrial microorganisms,at least as compared to glucose *** surprisingly,a number of approaches have been developed for improving D-xylose metabolism in diverse *** this work,we applied a previously developed evolution strategy based on media-in-oil emulsions for improving the growth yield of Escherichia coli NCM3722 on *** 30 rounds of evolutions,we isolated multiple mutants with increased growth yield on *** addition,we also observed similar increases in the growth *** mutants were selected for whole-genome *** mutants had an amber stop mutation in adenylate cyclase,which truncates nearly 60%of the ***,the ability of this mutant to grow on xylose indicated that truncated enzyme,lacking the C-terminal regulatory domain,is still *** other mutant had a point mutation in the cyclic AMP receptor protein(CRP),near the high affinity binding site for cyclic *** mutations,when intro-duced into wild type ***,were able to increase the growth yields at levels similar to the isolated *** addition to D-xylose,these mutant strains and their genetic mimics also exhibited higher growth rates and yields on glucose,lactose,and *** results suggest that the improved growth rates and yields are due to changes in the production and sensing of intracellular cyclic AMP concentrations and also suggest native concentrations are suboptimal with respect to the growth rate and yield under the growth conditions ***,these results may prove useful for engineering strains of *** for high-density fermentations or protein production.

The Brodmann area(BA)-based map is one of the most widely used cortical maps for studies of human brain functions and in clinical practice;however,the molecular architecture of BAs remains *** present study provided a global multiregional proteomic map of the human cerebral cortex by analyzing 29 *** 29 BAs were grouped into 6 clusters based on similarities in proteomic patterns:the motor and sensory cluster,vision cluster,auditory and Broca’s area cluster,Wernicke’s area cluster,cingulate cortex cluster,and heterogeneous function *** identified 474 cluster-specific and 134 BA-specific signature proteins whose functions are closely associated with specialized functions and disease vulnerability of the corresponding cluster or *** findings of the present study could provide explanations for the functional connections between the anterior cingulate cortex and sensorimotor cortex and for anxiety-related function in the sensorimotor *** brain transcriptome and proteome comparison indicates that they both could reflect the function of cerebral cortex,but show different *** proteomic data are publicly available at the Human Brain Proteome Atlas(***).Our results may enhance our understanding of the molecular basis of brain functions and provide an important resource to support human brain research.

The actin-bundling protein Fascin-1 modulates ciliary signalling

Primary cilia are microtubule-based cell organelles important for cellular communication. Since they are involved in the regulation of numerous signalling pathways, defects in cilia development or function are associated with genetic disorders, collectively called ciliopathies. Besides their ciliary functions, recent research has shown that several ciliary proteins are involved in the coordination of the actin cytoskeleton. Although ciliary and actin phenotypes are related, the exact nature of their interconnection remains incompletely understood. Here, we show that the protein BBS6, associated with the ciliopathy Bardet–Biedl syndrome, cooperates with the actin-bundling protein Fascin-1 in regulating filopodia and ciliary signalling. We found that loss of Bbs6 affects filopodia length potentially via attenuated interaction with Fascin-1. Conversely, loss of Fascin-1 leads to a ciliary phenotype, subsequently affecting ciliary Wnt signalling, possibly in collaboration with BBS6. Our data shed light on how ciliary proteins are involved in actin regulations and provide new insight into the involvement of the actin regulator Fascin-1 in ciliogenesis and cilia-associated signalling. Advancing our knowledge of the complex regulations between primary cilia and actin dynamics is important to understand the pathogenic consequences of ciliopathies.

GPS tracking of non-breeding ravens reveals the importance of anthropogenic food sources during their dispersal in the Eastern Alps

In many songbirds, the space use of breeders is well studied but poorly understood for non-breeders. In common ravens, some studies of non-breeders indicate high vagrancy with large individual differences in home range size, whereas others show that up to 40% of marked non-breeders can be regularly observed at the same anthropogenic food source over months to years. The aim of this study was to provide new insights on ravens＇ behavior during dispersal in the Eastern Alps. We deployed Global Positioning System （GPS） loggers on 10 individuals to gather accurate spatial and temporal information on their movements to quantify： 1） the dimension of the birds＇ space use （home range size with seasonal effects and daily/long-term travel distances）, 2） how long they stayed in a dispersal stage of wandering as opposed to settling temporarily, and 3） their destination of movements. We recorded movements of up to 40km per hour, more than 160km within 1 day and more than 11,000 km within 20 months, indicating high vagrancy. Switching frequently between temporarily settling and travelling large distances in short time intervals leads to extensive home ranges, which also explains and com- bines the different findings in the literature. The destinations are rich anthropogenic food sources, where the birds spent on average 75% of their time. We discuss how ravens may find these ＂feeding hot spots＂ and which factors may influence their decision to stay/leave a site. The strong dependence on anthropogenic resources found in this population may have implications for site management and conservation issues.

CENP-K and CENP-H may form coiled-coils in the kinetochores

Kinetochores are large proteinaceous structure on the surface of chromosomes’ primary constriction during mitosis. They link chromosomes to spindle microtubules and also regulate the spindle assem- bly checkpoint, which is crucial for correct chromosome segregation in all eukaryotes. The better known core networks of kinetochores include the KMN network (K, KNL1; M, Mis12 complex; N, Ndc80 complex)and CCAN (constitutive centromere-associated network). However, the detailed molecular mechanism of the kinetochore protein network remains unclear. This study demonstrates that CENP-H and CENP-K form quite stable subcomplex by TAP (tandem affinity purification) with HEK 293 cells which express TAP-CENP-K, with the ratio of purified CENP-H and CENP-K being close to 1︰1 even with high salt. Bioinformatic analysis suggests that CENP-H and CENP-K are enriched with coiled-coil regions. This implies that CENP-H and CENP-K form heterodimeric coiled-coils. Furthermore, the func- tional regions which form the complex are respectively located on their N- and C-terminals, but the association between the C-terminals is more complex. It is possible that this is the first identified het- erodimeric coiled-coils within the inner kinetochore, which is directly involved in the attachment be- tween kinetochores and the spindle microtubules.

Lipid Profile Altered in Phenanthrene Exposed Zebrafish Embryos with Implications for Neurological Development and Early Life Nutritional Status

Lecithotrophic fish embryos rely on finite maternally deposited yolk resources for early *** exposure can disrupt the uptake of yolk resources with consequences for *** this study,we investigate the impacts of altered yolk utilization on fish embryos using the cardiotoxic compound *** embryos were exposed to a cardiotoxic concentration of phenanthrene beginning at 6 hpf(hours post-fertilization)until a maximum of 72 *** were stained with Oil Red O to visualize neutral *** then used a nontargeted approach to profile lipids in 24 and 72 hpf embryos after phenanthrene *** assess changes in lipid movement within the embryo,the yolk sac was dissected from the body at 24 and 72 hpf and analyzed separately from the body at 72 ***,total metabolites were significantly reduced in the yolk sac,and staining for neutral lipids was reduced in the embryo body at 72 *** result is consistent with significant reductions in triglycerides in both the embryo body and yolk,indicating a limited contribution of impaired cardiac function to lipid mobilization at the dose ***,lysophosphatidylcholines and lysophosphatidylethanolamines were significantly increased in the 72 hpf embryo *** pathway analysis indicated that changes to these lysophospholipids could be linked to a disease model associated with inflammation and neuron demyelination consistent with previously observed injuries to neuronal and eye development in fish embryos and larvae.
