SARS-CoV-2 Pooled Testing Methodology for PCR Testing Applied in Private Laboratory in Armenia

Since the beginning of COVID-19 pandemics many countries were facing challenges with testing capacity recourse limitations. Throughout the waves of the pandemic countries were trying to address the existing constrains exploring solutions to increase the testing capacity with more cost-effective approaches. Pooled methodology was one of the methods which many have validated and used. It is evident that in case of pooled sample testing the sensitivity becomes lower, however the variation highly depends on the pool size as well as the incidence rate at the certain point. Armenia as well as many other countries has adopted regulations for mandatory COVID-19 PCR testing for all the travelers. Current study aimed to explore the efficiency of COVID-19 pooled PCR testing for nasopharyngeal swabs of individuals with no symptoms in a time period with good epidemiological state of the infection. Nasopharingeal swab samples from individuals were collected. The manual extraction of RNAs of samples was performed after pooling up to 5 samples. The pools with Cycle Threshold (CT) of < 37 were considered positive and were retested individually. In total 28,015 samples were grouped in 667 pools of which 57 were positive. The total number of positive samples was 65. The median difference (CT-pool–CT samples) was 2.4 (ranging from–3.0 to 8.9). The correlation of CT of pools and positive samples was positive. The correlation coefficient r = 0.84, P < 0.000, 95% CI range 0.7423 to 0.9243). The total economic saving when using pools compared to the individual testing was 72%. The minor difference between CT values of pools and samples can be explained by the dilution effect in the pool. However, the positive correlation between the values as well as the amount of cost saving demonstrate that pooling on nasopharyngeal samples for COVID-19 PCR testing can be a good method for efficient screening with significant resource saving. Application of a water quality model for determining instream aeration station location and operational rules:A case study

Instream aeration has been used as a supplement to secondary treatment or a substitute for tertiary treatment for meeting dissolved oxygen （DO） standards in rivers. Many studies have used water quality models to determine the number, location, and capacity of instream aeration stations （IASs） needed to meet DO standards in combination with other pollution control measures. DO concentrations have been improved in the North Shore Channel and North Branch Chicago River by the Devon Avenue IAS for more than 35 years. A study was initiated to determine whether it was better to rehabilitate or relocate this station and to determine appropriate operational guidance for the IAS at the selected location. A water quality model capable of simulating DO concentrations during unsteady flow was used to evaluate the proper location for an IAS and operational guidance for this IAS. Three test years, a dry year, a wet year, and an extreme year, were considered in the evaluation. The study found that the Devon Avenue IAS should be rehabilitated as this location performed as well as or better than any of 10 alternative locations. According to the new operational guidance for this IAS, the amount of time with blowers operating could be substantially reduced compared to traditional operations while at the same time the attainment of the DO standards could be increased. This study shows that a carefully designed modeling study is key to effective selection, location, and operation of IASs such that attainment of DO standards can be maximized while operation hours of blowers can be minimized.

Body-fitted mesh generation has long been the bottleneck of simulating fluid flows involving complex geometries. Immersed boundary methods are non-boundary-conforming methods that have gained great popularity in the last two decades for their simplicity and flexibility, as well as their non-compromised accuracy. This paper presents a summary of some numerical algori- thms along the line of sharp interface direct forcing approaches and their applications in some practical problems. The algorithms include basic Navier-Stokes solvers, immersed boundary setup procedures, treatments of stationary and moving immersed bounda- ries, and fluid-structure coupling schemes. Applications of these algorithms in particulate flows, flow-induced vibrations, biofluid dynamics, and free-surface hydrodynamics are demonstrated. Some concluding remarks are made, including several future research directions that can further expand the application regime of immersed boundary methods.

Patients with a diagnosis of polycystic ovary syndrome （PCOS） are on the rise. About 4%-12% of women are currently estimated to have this condition. It is hypothesized that PCOS appears in women who have long-standing insulin resistance （1R）, which leads to high androgen and testosterone levels; this ultimately disrupts their menstrual cycles. Some researchers attribute IR to genetic factors, although there have been only minute changes in the human genome in the past 20 000 years. However, even with a stable gene pool, genes can be turned on and offby the environment, food and air quality and toxin exposure.

Biological Dose Estimation Model for Proton Beam Therapy

Purpose: The recommended value for the relative biological effectiveness (RBE) of proton beams is currently assumed to be 1.1. However, there is increasing evidence that RBE increases towards the end of proton beam range that may increase the biological effect of proton beam in the distal regions of the dose deposition. Methods: A computational approach is presented for estimating the biological effect of the proton beam. It includes a method for calculating the dose averaged linear energy transfer (LET) along the measured Bragg peak and published LET to RBE conversion routine. To validate the proposed method, we have performed Monte Carlo simulations of the pristine Bragg peak at various beam energies and compared the analysis with the simulated results. A good agreement within 5% is observed between the LET analysis of the modeled Bragg peaks and Monte Carlo simulations. Results: Applying the method to the set of Bragg peaks measured at a proton therapy facility we have estimated LET and RBE values along each Bragg peak. Combining the individual RBE-weighted Bragg peaks with known energy modulation weights we have calculated the RBE-weighted dose in the modulated proton beam. The proposed computational method provides a tool for calculating dose averaged LET along the measured Bragg peak. Conclusions: Combined with a model to convert LET into RBE, this method enables calculation of RBE-weighted dose both in pristine Bragg peak and in modulated beam in proton therapy.

Secure and efficient outsourced computation in cloud computing environments is crucial for ensuring data confidentiality, integrity, and resource optimization. In this research, we propose novel algorithms and methodologies to address these challenges. Through a series of experiments, we evaluate the performance, security, and efficiency of the proposed algorithms in real-world cloud environments. Our results demonstrate the effectiveness of homomorphic encryption-based secure computation, secure multiparty computation, and trusted execution environment-based approaches in mitigating security threats while ensuring efficient resource utilization. Specifically, our homomorphic encryption-based algorithm exhibits encryption times ranging from 20 to 1000 milliseconds and decryption times ranging from 25 to 1250 milliseconds for payload sizes varying from 100 KB to 5000 KB. Furthermore, our comparative analysis against state-of-the-art solutions reveals the strengths of our proposed algorithms in terms of security guarantees, encryption overhead, and communication latency.

During the past 30 years, there has been spectacular growth in the use of risk analysis and risk management tools developed by engineers in the financial and insurance sectors. The insurance, the reinsurance, and the investment banking sectors have enthusiastically adopted loss estimation tools developed by engineers in developing their business strategies and for managing their financial risks. As a result, insurance/reinsurance strategy has evolved as a major risk mitigation tool in managing catastrophe risk at the individual, corporate, and government level. This is particularly true in developed countries such as US, Western Europe, and Japan. Unfortunately, it has not received the needed attention in developing countries, where such a strategy for risk management is most needed. Fortunately, in the last five years, there has been excellent focus in developing ＂Insur Tech＂ tools to address the much needed ＂Insurance for the Masses＂, especially for the Asian Markets. In the earlier years of catastrophe model development, risk analysts were mainly concerned with risk reduction options through engineering strategies, and relatively little attention was given to financial and economic strategies. Such state-of-affairs still exists in many developing countries. The new developments in the science and technologies of loss estimation due to natural catastrophes have made it possible for financial sectors to model their business strategies such as peril and geographic diversification, premium calculations, reserve strategies, reinsurance contracts, and other underwriting tools. The Surface Mining Control and Reclamation Act （SMCRA） was enacted in 1977 and was the overriding federal regulation governing mining and reclamation of surface coal mines in the United States of America （USA）. Many of the newest surface mines in the USA, at that time, were in the western portion of the nation. Wyoming surface coal mines numbered approximately 20 and were located throughout the state in the coal bearing regions, generally in the south/southwestern portion of the state and the northeastern corner. The Office of Surface Mining Reclamation and Enforcement （OSMRE） is the federal agency tasked with oversight of the implementation of SMCRA. Individual states developed statewide programs that met or exceeded the requirements of SMCRA and, thus, obtained primacy over coal mine mining and reclamation within their boundaries as long as those regulatory conditions were maintained. OSMRE retained oversight on the Indigenous Nations programs within the USA. Much information has been learned on the reclamation side of SMCRA, its regulations and state programs since its passage. This paper and presentation will present some of the basic changes made in reclamation programs over the last 40 years in the State of Wyoming.
