Anti-inflammatory and antinociceptive activities of Rhipicephalus microplus saliva

To evaluate the antinociceptive and anti-inflammatory activities and the toxic effects of Rlhipicephalus microplus saliva for elucidating the modulation mechanism between arthropod saliva and ***:For saliva collection,engorged ticks were obtained from a controlled bovine infestation and collected by natural *** ticks were fixed and injected pilocarpine 0.2%for induction of *** was collected,lyophilized and stored at-80℃.Cytotoxic activity was assessed by the hemolysis method(25,50,100,200 and 300μg/mL)and MTT cell viability assay(2.5,5,10,20 and 40μg/mL)for 24,48 and 72 ***-inflammatory activity was evaluated using the method of neutrophil migration to the peritoneal cavity of mice at doses of l0,15 and 20 mg/kg;antinociceptive activity was assessed using the acetic acid-induced writhing test,and formalin-induced paw-licking in mice at dose of 15 mg/***:Saliva did not cause erythrocytes hemolysis at any concentration tested,as well as did not decrease cell viability in the MTT *** inhibited neutrophil migration by 87%and 73%at doses of l5 and 20 mg/kg,*** the nociceptive tests,saliva presented analgesic activity of69.96%in the abdominal writhing test,and of 84.41%in the formalin *** study proves that Rlhipiceplhalus microplus saliva has significant in vivo anti-inflammatory and antinociceptive *** data presented herein support the development of further studies to elucidate the active principles of Rhipicephalus microplus saliva and its mechanism of action and,in future,to develop novel anti-inflammatory and analgesic drugs.

Aquatic exercise program-modulated oxidative stress markers in patients with Parkinson's disease

Parkinson's disease is a neurodegenerative *** stress,i.e.,the imbalance between the generation of reactive oxygen species and the antioxidant defense capacity of the body,plays an important role in the pathogenesis of this *** exercise can regulate oxidative *** purpose of this study was to analyze the short-and long-term effects of an aquatic exercise program on oxidative stress levels in patients with Parkinson's *** aquatic exercise program was carried out during 1 month with two sessions per week(1 hour/session).Blood samples were collected at four different time points:pre-intervention,immediately,48 hours,and 30 days after the first session of aquatic exercise *** results revealed that water-based programs modulated antioxidant enzyme activity,increased superoxide dismutase activity,reduced catalase activity,and increased the ratio of superoxide dismutase activity to catalase activity in patients with Parkinson's *** with pre-intervention and 48 hours after the first session of aquatic exercise program,superoxide dismutase activity was higher and catalase activity was lower immediately and 30 days after the first *** results demonstrated that aquatic exercise program could modulate oxidative stress,mainly by the effect of antioxidant enzyme *** results could better help understand the target of oxidative stress in Parkinson's *** study was approved by the Ethics Committee of Centro Universitário Metodista IPA(approval No.1.373.911)on August 9,2019 and registered with REBEC(registration number:RBR-6 NJ4 MK).

An in vivo approach to the reported effects of phenolic acids and flavonoids on the pancreaticα-amylase activity

Reduction in the activity of theα-amylases which hydrolyze starch and glycogen has been proposed for diminishing the incidence of obesity and *** plant extracts contain inhibitors ofα-amylases andα-glucosidases and several molecules with this activity have been *** present work attempts at investigating if several natural compounds(flavonoids and phenolic acids)that have been reported to be inhibitors of those hydrolytic enzymes can also inhibit starch absorption in *** starch tolerance test in mice was used for assaying starch absorption after administration of 100 mg/kg of five flavonoids(epicatechin,catechin,rutin,quercetin,naringenin)and five phenolic acids(gallic,syringic,vanillic,sinapic,ferulic),which are known in vitro inhibitors ofα-*** the flavonoids,only rutin inhibited starch absorption in mice,even though its effect was only 33%of that one found for the positive control *** the five phenolic acids tested only syringic acid produced a clear diminution in starch absorption,which was 47%of that one found with *** the other hand,other three phenolic acids,namely gallic,sinapic and ferulic acid,caused delay in the absorption of starch,which was characterized by a shift in the time peak of maximal blood glucose concentration,with no modification in the excess of glucose that effectively entered the *** at detecting eventual synergisms by administrating mixtures of flavonoids and phenolic acids were *** results recommend careful translation of in vitro data on the inhibition ofα-amylases andα-glucosidases to the in vivo conditions.

Paternal physical exercise modulates global DNA methylation status in the hippocampus of male rat offspring

It is widely known that maternal physical exercise is able to induce beneficial improvements in offspring cognition; however, the effects of paternal exercise have not been explored in detail. The present study was designed to evaluate the impact of paternal physical exercise on memory and learning, neuroplasticity and DNA methylation levels in the hippocampus of male offspring. Adult male Wistar rats were divided into two groups: sedentary or exercised fathers. The paternal preconception exercise protocol consisted of treadmill running, 20 minutes daily, 5 consecutive days per week for 22 days, while the mothers were not trained. After mating, paternal sperm was collected for global DNA methylation analysis. At postnatal day 53, the offspring were euthanized, and the hippocampus was dissected to measure cell survival by 5-bromo-2′-deoxiuridine and to determine the expression of synaptophysin, reelin, brain-derived neurotrophic factor and global DNA methylation levels. To measure spatial memory and learning changes in offspring, the Morris water maze paradigm was used. There was an improvement in spatial learning, as well as a significant decrease in hippocampal global DNA methylation levels in the offspring from exercised fathers compared with those from sedentary ones; however, no changes were observed in neuroplasticity biomarkers brain-derived neurotrophic factor, reelin and 5-bromo-2′-deoxiuridine. Finally, the global DNA methylation of paternal sperm was not significantly changed by physical exercise. These results suggest a link between paternal preconception physical activity and cognitive benefit, which may be associated with hippocampal epigenetic programming in male offspring. However, the biological mechanisms of this modulation remain unclear.

Arbuscular mycorrhizal fungi（AMF） are important during revegetation of mining sites, but few studies compared AMF community in revegetated sites with pristine adjacent ecosystems. The aim of this study was to assess AMF species richness in a revegetated iron-mining site and adjacent ecosystems and to relate AMF occurrence to soil chemical parameters. Soil samples were collected in dry and rainy seasons in a revegetated iron-mining site（RA） and compared with pristine ecosystems of forest（FL）, canga（NG）,and Cerrado（CE）. AMF species were identified by spore morphology from field and trap cultures and by LSU r DNA sequencing using Illumina. A total of 62 AMF species were recovered, pertaining to 18 genera and nine families of Glomeromycota. The largest number of species and families were detected in RA, and Acaulospora mellea and Glomus sp1 were the most frequent species. Species belonging to Glomeraceae and Acaulosporaceae accounted for 42%–48% of total species richness. Total number of spores and mycorrhizal inoculum potential tended to be higher in the dry than in the rainy season, except in RA. Sequences of uncultured Glomerales were dominant in all sites and seasons and five species were detected exclusively by DNA-based identification. Redundancy analysis evidenced soil p H,organic matter, aluminum, and iron as main factors influencing AMF presence. In conclusion, revegetation of the iron-mining site seems to be effective in maintaining a diverse AMF community and different approaches are complementary to reveal AMF species, despite the larger number of species being identified by traditional identification of field spores.

Critically ill cirrhotic patients have high in-hospital mortality and utilize significant health care resources as a consequence of the need for multiorgan *** this fact,their mortality has decreased in recent decades due to improved care of critically ill ***-on-chronic liver failure(ACLF),sepsis and elevated hepatic scores are associated with increased mortality in this population,especially among those not eligible for liver *** score is superior to another in the prognostic assessment of these patients,and both liver-specific and intensive care unit-specific scores have satisfactory predictive *** sequential assessment of the scores,especially the Sequential Organ Failure Assessment(SOFA)and Chronic Liver Failure Consortium(CLIF)-SOFA scores,may be useful as an auxiliary tool in the decision-making process regarding the benefits of maintaining supportive therapies in this population.A CLIF-ACLF>70 at admission or at day 3 was associated with a poor prognosis,as well as SOFA score>19 at baseline or increasing SOFA score>*** studies addressing the prognostic assessment of these patients are necessary.

The aim of the current study was to determine the nature of the seasonal variability of the Suspended Particulate Matter(SPM) fluxes from the drainage basin to the estuary in a macrotidal region(Northeastern Brazil),and the estuarine response to a seawater intrusion regarding sediment deposition,which will support the understanding of the global transport of materials at the continent-ocean *** structure data was acquired using a Conductivity,Temperature,and depth(CTD) probe with a sampling frequency of 4 *** particulate material was measured by gravimetric measurements applied to exact filtered volume *** outflows were measured through the use of an Acoustic Doppler Current Profiler(ADCP) with frequency of 1.5 *** horizontal thermal and saline gradients varied from warmer and less saline waters(2014) to cooler and saline waters(2015).The gradient behavior when linked to volume transport and SPM flows,suggests a minimization of the fluvial flows in 2015,easing the advance of coastal water(CW) towards the inner estuary,leading to an inversion of the baroclinic pressure *** bottom saline front,generated by the entrance of coastal water masses,caused an increase in SPM concentrations due to increased fluid density,resuspension of previously deposited sediment,and erosion of *** concentrations of SPM indicate higher volume transport suggesting a hydraulic barrier due to the change/inversion of the baroclinic pressure gradient,resulting in water and material *** deposition was observed during neap tide,while during spring tide the material is resuspended,increasing the concentration,generating cycles of deposition and erosion during the neap-spring *** sediment in suspension that reach the estuary,even with low fluvial volume,stay in this environment forming new islands because of *** deposition rates or sediment cycling,if generated by the hydraulic barrier,may indicate that the flo

Aims Our objective was to quantify the contributions of the seed bank and the established vegetation to the species composition,functional composition and diversity,and discuss the implications of these differences in regeneration and persistence of floodplain plant *** We sampled all ground cover vegetation up to 1.5 m height and seed bank in 25 plots(10 m×1 m)distributed across five sites in dry and rainy seasons in a periodically flooded savanna in the Pantanal wetland,*** evaluated the soil seed bank by seedling emergence *** Findings The seed bank species had traits that conferred regeneration to the communities,while persistence traits characterized the *** seed bank had higher functional richness and lower functional evenness than the *** existence of different plant traits between seed bank and vegetation allowed the coexistence of species with functionally contrasting persistence and regeneration traits,which may help maintain functional *** may allow the community to be more resilient when dealing with different environmental filters such as drought,fire and flood.
