Antioxidant Capacity in Vanilla Extracts Obtained by Applying Focused Microwaves

ORAC method and a continuous flow injection method based on Folin-Ciocalteau reaction (FI-FC) were used for determining the antioxidant activity in extracts obtained by using focused microwaves. Analysis of the antioxidant capacity (AC) of the main compounds of vanilla (vanillin, p-hydroxybenzaldehyde, p-hydroxybenzoic acid and vanillic acid) was also carried out. Vanilla extracts obtained by using focused microwaves had a higher AC (between 72% and 117%) than the obtained by conventional methods. Vanillin had a linear correlation with the antioxidant capacity of the extracts and it is the most influential compound in the antioxidant power. The AC calculated by the ORAC method and the FI-FC method had a ratio 2:1 because of different kinetics and reaction mechanisms of the antioxidants with the reagents, so it is necessary more than one method to establish the antioxidant power in food. On base on the results of the present study microwaves energy can be used to obtain vanilla extracts to improve the AC of them.

Cardiovascular disease(CVD) is a prevalent condition in general population and the first cause of death overall. Klotho, a pleiotropic protein related to longevity that acts as a co-receptor of the fibroblast growth factor 23, has been proposed as a key regulator of the development of CVD. In the few clinical studies made, it has been observed a relationship between low levels of soluble Klotho and the occurrence and severity of CVD, as well as a reduction of cardiovascular risk when they are high. Also, different polymorphisms of human Klotho gene have been related to the incidence of cardiovascular events. Moreover, several experimental studies indicate that this protein acts in the maintenance of vascular homeostasis. Klotho improves endothelial dysfunction through promotion of NO production and mediates antiinflammatory and anti-aging effects such as suppression of adhesion molecules expression, attenuation of nuclear factor-kappa B or inhibition of Wnt signaling. Furthermore,this protein is related to the attenuation of vascular calcification as well as prevention of cardiac hypertrophy. The expression of this protein in the vascular wall implies a new scenario for the treatment of vascular disorders. The purpose of this review is to provide an overview of the relationship between the Klotho protein and CVD, in addition to its role in the maintenance of functional vascular integrity.

Computational Aspects of Dam Risk Analysis： Findings and Challenges

In recent years, risk analysis techniques have proved to be a useful tool to inform dam safety manage- ment. This paper summarizes the outcomes of three themes related to dam risk analysis discussed in the Benchmark Workshops organized by the International Commission on Large Dams Technical Com- mittee on ＂Computational Aspects of Analysis and design of Dams.＂ In the 2011 Benchmark Workshop, estimation of the probability of failure of a gravity dam for the sliding failure mode was discussed. Next, in 2013, the discussion focused on the computational challenges of the estimation of consequences in dam risk analysis. Finally, in 2015, the probability of sliding and overtopping in an embankment was an- alyzed. These Benchmark Workshops have allowed a complete review of numerical aspects for dam risk analysis, showing that risk analysis methods are a very useful tool to analyze the risk of dam systems, including downstream consequence assessments and the uncertainty of structural models.

AIM: To characterize hydraulic right ventricle (RV) af- terload by pulmonary arterial pressure waveform analy- sis in an acute pulmonary hypertension (PH) model. METHODS: Pulmonary artery (PA) flow and pressure were recorded in six anesthetized sheep. Acute iso- baric PH was induced by phenylephrine (active) and PA mechanical constriction (passive). We estimated the amplitude of the forward and reflected pressure waves according to the inflection point. In most cases the in- flection pressure was smooth, thus the inflection point was defined as the time at which the first derivative ofpulmonary arterial pressure reached its first minimum. We calculated the input and characteristic (Z C , time- domain Li method) impedances, the capacitance index (stroke volume/pulse pressure), the augmentation index (AI) (reflected pressure/pulse pressure), the frac- tional pulse pressure (pulse pressure/mean pressure) and the wasted energy generated by the RV due to wave reflection during ejection (E W ). RESULTS: Pulse pressure, fractional pulse pressure, AI and Z C increased and capacitance index decreased during passive PH with respect to control (P dex did not change and E W and the AI decreased during active PH. Pulse pressure correlated with E W and Z C and the AI was cor- related with E W (r > 0.6, P ysis al- lows the quantification of the dynamic RV afterload. Prospective clinical studies will be necessary to validate this time-domain approach to evaluate the dynamic RV afterload in chronic PH.

Hollow conduits of natural or synthetic origins have shown acceptable regeneration results in short nerve gap repair;however,results are still not comparable with the current gold standard technique“autografts”.Hollow conduits do not provide a successful regeneration outcome when it comes to critical nerve gap *** the lumen of conduits with different extracellular materials and cells could provide a better biomimicry of the natural nerve regenerating environment and is expected to ameliorate the conduit *** this study,we evaluated nerve regeneration in vivo using hollow chitosan conduits or conduits enriched with fibrin-collagen hydrogels alone or with the further addition of adipose-derived mesenchymal stem cells in a 15 mm rat sciatic nerve transection *** changes in the hydrogel consistency and structural stability in vivo led to a failure of nerve regeneration after 15 ***,the molecular assessment in the early regeneration phase(7,14,and 28 days)has shown an upregulation of useful regenerative genes in hydrogel enriched conduits compared with the hollow *** composed of fibrin-collagen were able to upregulate the expression of soluble NRG1,a growth factor that plays an important role in Schwann cell *** further enrichment with adipose-derived mesenchymal stem cells has led to the upregulation of other important genes such as ErbB2,VEGF-A,BDNF,c-Jun,and ATF3.

Growth-associated protein 43 plays a key role in neurite outgrowth through cytoskeleton *** have previously demonstrated that structural damage of peripheral nerves induces growth-associated protein 43 upregulation to promote growth cone ***,the limited regenerative capacity of the central nervous system due to an inhibitory environment prevents major changes in neurite outgrowth and should be presumably associated with low levels of growth-associated protein 43 ***,central alterations due to peripheral nerve damage have never been assessed using the growthassociated protein 43 *** this study,we used the tubulization technique to repair 1 cm-long nerve gaps in the rat nerve injury/repair model and detected growth-associated protein 43 expression in the peripheral and central nervous ***,histological analysis of the regeneration process confirmed an active regeneration process of the nerve gaps through the conduit from 10 days *** growth-associated protein 43 expression profile varied across regions and follow-up times,from a localized expression to an abundant and consistent expression throughout the regeneration tissue,confirming the presence of an active nerve regeneration ***,spinal cord changes were also histologically assessed,and no apparent changes in the structural and cellular organization were observed using routine staining ***,remarkable differences and local changes appeared in growth-associated protein 43 expression at the spinal cord level,in particular at 20 days post-repair and ***-associated protein 43 protein was first localized in the gracile fasciculus and was homogeneously distributed in the left posterior *** findings differed from the growth-associated protein 43 pattern observed in the healthy control,which did not express growth-associated protein 43 at these *** results revealed a differential expression in growth-associate

There has been significant progress in our understanding of the pathobiology, epidemiology and prognosis of pulmonary vascular disease and, over the past few years, there has been an explosion of clinical therapeutic trials for pulmonary arterial hypertension (PAH). The increasing number of different conditions now associated with PAH and the appearance of new diagnostic techniques have led to a need for a systematic diagnostic approaches and a new disease classification, which has resulted in notable improvements in the quality and efficacy of clinical care. We appreciate traditional resting right heart catheterization techniques (which still remain the gold standard for diagnosing PAH and managing patients on therapy) and look forward to novel invasive techniques (e.g. intravascular ultrasound) that add greatly to our understanding of right ventricle and pulmonary circulation, and for the interpretation of data from clinical trials as well.
