Quantification of In Vivo Epidermal Keratinocyte Architecture Associated with the Signs of Skin Aging and the Skin Benefit Evaluation by Application of Galactomyces Ferment Filtrate (Pitera)-Containing Skin Care Product

Background: Aged skin exhibits visual alterations such as wrinkles, rough texture, pore dilation, and dull skin tone, as well as physiological aging, namely, decreased hydration and increased transepidermal water loss (TEWL). Recent advances in coherence tomography have also revealed that skin aging affects in vivo epidermal keratinocyte architecture. However, the interconnectivity between spatial architectural aging and visual/physiological aging parameters remains largely unknown. Purpose: To elucidate whether the tomographic keratinocyte architectural aging is correlated with visual and physiological skin aging parameters and to quantitatively evaluate the improvements of the architectural, visual, and physiological aging parameters by the daily treatment of the skin care formula containing Galactomyces Ferment Filtrate (GFF, 8X PiteraTM). Method: We measured the in vivo keratinocyte cellular architecture with two-photon stereoscopic tomography obtaining by-layer epidermal section images in 78 Asian females of various ages. Visual aging parameters were analyzed using a portable image capture system. Hydration and TEWL were also assessed. The anti-aging effects of GFF-containing skin moisturizer (SK-II LXP CreamTM) were also examined in two studies after twice-daily application for 2 (N = 35) and 4 (N = 32) weeks. Results: As for the keratinocyte cellular architecture, skin aging was significantly associated with decreased cell density and increased cell uniformity. These architectural aging parameters were significantly correlated with visual and physiological aging parameters, namely, rough texture, wrinkles, pore dilation, dull skin tone, dehydration, and increased TEWL. The strong interconnectivity allowed us to develop formulae to estimate the keratinocyte architecture from visual aging parameters. Six Amino Acids among Natural Moisturizing Factors Responsible for Skin Hydration: Improvement and Anti-Aging of Skin by Galactomyces Ferment Filtrate-PiteraTM Containing Skin Moisturizer

Background: Natural moisturizing factors (NMFs) are filaggrin-derived components in the cornified layer that are critical for maintaining healthy skin moisturization and barrier function. However, studies have reported conflicting findings on the relationship between NMF levels and aging, while few studies have investigated this relationship clinically. To fill this research gap, we determined the levels of major NMF components such as free amino acids, pyrrolidone carboxylic acid, and urocanic acids, and individually verified their relationships with skin hydration, barrier function, age, and skin aging. Purpose: The objective of this study was to clinically investigate the relationship between NMF components levels and skin aging in facial skin. The main NMF components were obtained from facial skin and quantified. We then selected NMF components showing strong relationships to skin hydration, and analyzed the relationships of the levels of these selected NMF components with signs of skin aging, namely, texture, pores, wrinkles, and dullness (L-value). We also examined the efficacy of treatment with a skin care formula (SK-II Facial Treatment Essence, called SK-II FTE hereafter) including Galactomyces ferment filtrate (GFF, PiteraTM) on the selected NMF component levels associated with skin hydration and barrier function, and the signs of skin aging of texture, pores, wrinkles, and dullness (L-value). Method: We conducted two clinical trials in this research. In Study 1, we measured 23 NMF components using tape-stripped cornified layer to quantify them via an HPLC method in 196 Asian females aged 20 to 59 (mean S.D., 38.6 9.4). Facial visual aging parameters [texture, pores, wrinkles, and dullness (L-value)], as well as elasticity (R7), skin hydration, and TEWL, were quantified using facial skin imaging and skin physical property measurement devices. An experimental and numerical study of packing, compression, and caking behaviour of detergent powders

This paper presents an experimental and numerical study of the packing, compression, and caking behaviour of spray dried detergent （SDD） powders with a two-fold aim： an experimental process of observation and evaluation of the packing, compression and caking behaviour of SDD powders, and a numerical approach based on discrete element modelling （DEM）. The mechanical properties, including the stress-strain response and the corresponding porosity change as a function of consolidation stress in a confined cylinder, the stress-strain response during unconfined shearing and the cake strength as a func- tion of consolidation stress, were evaluated and compared for different SDD powders using an extended uniaxial tester （Edinburgh Powder Tester - EPT）. The experiments using EPT showed excellent repro- ducibility in the measurement of packing, compression and caking behaviour and were therefore very useful for describing the handling characteristics of these powdered products including screening new products and different formulations. It was found that the sample with higher moisture had lower bulk porosity but higher compressibility and cake strength. The porosity, compressibility and cake strength were found to vary across different size fractions of the same sample. The larger sieve-cut samples had higher initial bulk porosity, compressibility and cake strength. It is revealed that moisture plays a sig- nificant role in packing, compression, and shearing behaviour of the powder. Three-dimensional DEM modelling using a recently developed elasto-plastic adhesive-frictional contact model showed that the contact model is able to capture the detergent behaviour reasonably well and can be used to model complex processes involving these powders.

Effect of Non-reactive Powder Particle Properties on Dry Agglomeration in a High Shear Mixer

The effect of non-reactive powder particle properties on the detergent dry agglomeration process in a high shear mixer was investigated. Three types of micron-scale silica were chosen as the non-reactive fine powders and a semi-solid alkyl ethyl ethoxy sulfate(AES) paste with ultra-high viscosity was chosen as the binder. The granules were characterized using massbased granule size distribution, scanning electron microcopy, and bulk density tests. The results revealed that powder particle size plays a leading role in agglomeration behavior. A decrease in the median particle size results in enhanced dispersion of silica particles in the AES paste binder droplets, which leads to the formation of uniform granules that are slightly affected by compacting forces. Agglomerate quality, using silica with high oil absorption as well as optimum particle size, was satisfactory, and the product exhibited a smaller median particle size, narrower size distribution, and superior anti-caking capacity under the same liquid-to-solid ratio(L/S).

Regime Map of Dry Neutralization Agglomeration Process Containing Non-Reactive Ingredient on a Laboratory Scale

This study described a regime map for dry neutralization agglomeration. Based on the map, the effects of selected key parameters, such as ingredient composition, operation temperature, agitation speed, and size of Na_2CO_3 particles, were investigated using a laboratory-scale mixer, and properties of the agglomeration product were analyzed, including particle size distribution, Hunter color, and flowability. Torque curves evolving during the process were correlated with the system flowability. Three distinguishable regimes were indicated, dry, wet, and transitional, and the agitation speed was found to have a different influence on the agglomeration process for the three regimes. Furthermore, the influence of temperature on reactive agglomeration significantly differed from that in agglomeration processes in which the binder was non-reactive.

The diffusion kinetics of a molecular probe-rhodamine B-in ternary aqueous solutions containing poly(vinyl alcohol),glycerol,and surfactants was investigated using fluorescence correlation spectroscopy and dynamic light *** show that the diffusion characteristics of rhodamine B in such complex systems is determined by a synergistic effect of molecular crowding and intermolecular interactions between chemical *** presence of glycerol has no noticeable impact on rhodamine B diffusion at low concentration,but significantly slows down the diffiision of rhodamine B above 3.9%(w/v)due to a dominating steric inhibition ***,introducing surfactants(cationic/nonionic/anionic)to the system results in a decreased diffusion coefficient of the molecular *** solutions containing nonionic surfactant,this can be explained by an increased crowding *** ternary poly(vinyl alcohol)solutions containing cationic or anionic surfactant,surfactant-polymer and surfactant-rhodamine B interactions alongside the crowding effect of the molecules slow down the overall diffiisivity of rhodamine *** results advance our insight of molecular migration in a broad range of industrial complex formulations that incorporate multiple compounds,and highlight the importance of selecting the appropriate additives and surfactants in formulated products.

ZnO nanofluids-A potential antibacterial agent

In this work, ZnO nanofluids were produced by a medium mill with a pH value of about 7.2 and characterized by Nano-Sizer and SEM. After milling, ZnO nanofluids were formed with an average particle size of 198.4 nm. The ZnO nanofluids used for testing were stored for different periods (1-, 90- and 120-day) and kept in different conditions (under the light and in the dark). The antibacterial activities of these ZnO nanofluids were evaluated by estimating the reduction ratio of the bacteria treated with ZnO. The results showed that the ZnO nanofluid stored for 120 days under the light had the best antibacterial behavior against Escherichia coli DH5a. SEM images suggest that an interaction between the ZnO particles and the E. coli bacteria cells caused by electrostatic forces might be a mechanism.

Oral rinses containing chemotherapeutic agents, such as cetylpyridinium chloride （CPC）, can alleviate plaque-induced gingival infections, but how oral microbiota respond to these treatments in human population remains poorly understood. Via a double- blinded, randomised controlled trial of 91 subjects, the impact of CPC-containing oral rinses on supragingival plaque was investigated in experimental gingivitis, where the subjects, after a 21-day period of dental prophylaxis to achieve healthy gingivae, received either CPC rinses or water for 21 days. Within-subject temporal dynamics of plaque microbiota and symptoms of gingivitis were profiled via 16S ribosomal DNA gene pyrosequencing and assessment with the Mazza gingival index. Cetylpyridinium chloride conferred gingival benefits, as progression of gingival inflammation resulting from a lack of dental hygiene was significantly slower in the mouth rinse group than in the water group due to inhibition of 17 gingivitis-enriched bacterial genera. Tracking of plaque a and β diversity revealed that CPC treatment prevents acquisition of new taxa that would otherwise accumulate but maintains the original biodiversity of healthy plaques. Furthermore, CPC rinses reduced the size, local connectivity and microbiota-wide connectivity of the bacterial correlation network, particularly for nodes representing gingivitis- enriched taxa. The findings of this study provide mechanistic insights into the impact of oral rinses on the progression and maturation of dental plaque in the natural human population.
