The role of the molecular chaperone heat shock protein A2 （HSPA2） in regulating human sperm-egg recognition

One of the most common lesions present in the spermatozoa of human infertility patients is an idiopathic failure of sperm-egg recognition. Although this unique cellular interaction can now be readily by-passed by assisted reproductive strategies such as intracytoplasmic sperm injection （ICSI）, recent large-scale epidemiological studies have encouraged the cautious use of this technology and highlighted the need for further research into the mechanisms responsible for defective sperm-egg recognition. Previous work in this field has established that the sperm domains responsible for oocyte interaction are formed during spermatogenesis prior to being dynamically modified during epididymal maturation and capacitation in female reproductive tract. While the factors responsible for the regulation of these sequential maturational events are undoubtedly complex, emerging research has identified the molecular chaperone, heat shock protein A2 （HSPA2）, as a key regulator of these events in human spermatozoa. HSPA2 is a testis-enriched member of the 70 kDa heat shock protein family that promotes the folding, transport, and assembly of protein complexes and has been positively correlated with in vitro fertilization （IVF） success. Furthermore, reduced expression of HSPA2 from the human sperm proteome leads to an impaired capacity for cumulus matrix dispersal, sperm-egg recognition and fertilization following both IVF and ICSI. In this review, we consider the evidence supporting the role of HSPA2 in sperm function and explore the potential mechanisms by which it is depleted in the spermatozoa of infertile patients. Such information offers novel insights into the molecular mechanisms governing sperm function.

Poor semen quality is an attribute of the human condition that we sharewith our closest primate ancestors and may be part of our evolutionary inheritance. This characteristic may also reflect the lack of selection pressure on male fertility in advanced societies that have gone through the demographic transition. In the future, such trends may be further exacerbated by the widespread use of ART, which will impair the elimination of low fecundity genotypes from the human gene pool. Environmental pollutants may also be influencing male fertility, however, neither the chemical identity of these factors nor their mechanism-of-action are fully understood.

This paper summarises the results of a combined numerical,statistical and experimental study concerned with the use of dissimilar walling systems on the external parts of a given building *** rational behind this“hybrid wall”concept,as opposed to conventional approaches where identical walls are used in a building envelope,is to achieve a more effective distribution of thermal mass across the envelope and,hence,improve the overall thermal per-formance of the *** effectiveness of the“hybrid wall”concept was investigated using a series of hypothetical building modules of common Australian residential constructions,namely Light Weight(LW),Brick Veneer(BV),Reverse Brick Veneer(RBV)and Cavity Brick(CB).These designs were examined numerically using a commercial energy rating tool known as“AccuRate”,statistically using JMP software and experimentally using a novel bench-scale setup developed as part of this *** performance of each design was evaluated by its energy *** numerical predictions and experimental data highlighted that the east and west walls have the most impact on the energy consumption under Australian climatic *** was found that considerable reductions in the energy consumption could be achieved in cases where the hybrid wall concept was implemented through the use of high ther-mal mass insulated walls on the east and west sides of the building envelope.

Reaction of nitrous oxide with methane to synthesis gas:A thermodynamic and catalytic study

The aim of the present study is to explore the coherence of thermodynamic equilibrium predictions with the actual catalytic reaction of CH4 with N2O,particularly at higher CH4 *** this purpose,key process variables,such as temperature(300℃-550℃) and a molar feed ratio(N2O/CH4 = 1,3,and 5),were altered to establish the conditions for maximized *** experimental study was conducted over the Co-ZSM-5 catalyst in a fixed bed tubular reactor and then compared with the thermodynamic equilibrium compositions,where the equilibrium composition was calculated via total Gibbs free energy minimization *** results suggest that molar feed ratio plays an important role in the overall reaction products *** for N2O conversions,and irrespective of N2O/CH4feed ratio,the thermodynamic predictions coincide with experimental data obtained at approximately 475℃-550℃,indicating that the reactions are kinetically limited at lower range of *** example,theoretical calculations show that the H2 yield is zero in presence of excess N2O(N2O/CH4= 5).However over a Co-ZSM-5 catalyst,and with a same molar feed ratio(N2O/CH4) of 5,the H2yield is initially 10%at 425℃,while above450℃ it drops to ***,H2yield steadily increases with temperature and with the level of CH4 conversion for reactions limited by N2O concentration in a reactant *** maximum attainable(from thermodynamic calculations and at a feed ratio of N2O/CH4=3) H2yield at 550℃ is 38%,whereas at same temperature and over Co-ZSM-5,the experimentally observed yield is about 19%.Carbon deposition on Co-ZSM-5 at lower temperatures and CH4 conversion(less than 50%) was also *** higher temperatures and levels of CH4conversion(above 90%),the deposited carbon is suggested to react with N2O to form CO2.

Kanatsu-Shinohara and colleagues continue to reveal the secrets of the rare mammalian spermatogonial stem cells. Their most recent study offers a new approach by applying principles from hematopoietic stem cell research to demonstrate that cells which form a cobble- stone-like underlay beneath testicular somatic cells in culture are spermatogonial stem cells. Utilization of mouse models and cell cultures shows how the chemokine, CXCL12, fits into the signalling cascade which governs the fate of these cells and hence is essential to male fertility.

Short-wave infrared organic phototransistors with strong infrared-absorbing polytriarylamine by electron-transfer doping

Short-wavelength infrared(SWIR)sensors have attracted keen attention due to the increasing necessity in a variety of scientific and industrial applications,including biomedical and information technology *** conventional SWIR sensors are made of inorganic materials with rigid and brittle characteristics,organic materials with a discrete SWIR absorption are required for flexible SWIR sensors in the flexible electronics ***,we demonstrate that a polytriarylamine,poly[N,N′-bis(4-butylphenyl)-N,N′-bis(phenyl)benzidine](PolyTPD),can absorb almost full range of SWIR wavelength(λ=1000–3200 nm)after 48 h doping with tris(pentafluorophenyl)borane(BCF).The spectroscopic characterization disclosed that an electron transfer from PolyTPD to BCF created a new low energy level(gap)state leading to the SWIR absorption in the BCF-doped PolyTPD *** phototransistors(OPTRs)with the BCF-doped PolyTPD films as a gate-sensing layer could detect the SWIR light with a reasonable photoresponsivity of~538 mAW^(−1)(λ=1500 nm),~541 mAW^(−1)(λ=2000 nm),and~222 mAW^(−1)(λ=3000 nm).The present breakthrough SWIR-OPTR technology can pave a way for further advances in SWIR-absorbing organic materials and flexible SWIR sensors.

Motion characteristics of binary solids in a liquid fluidised bed with inclined plates

Rectangular inclined channels prove promising for solid classification based on the principle of parti- cle differential sedimentation. In the present work, we investigated the motion characteristics of binary solids in a modified fluidised bed （mFB） with inclined plates. We developed a theoretical model for the particle motion behaviour that accounts for the average solid volume fraction in the inclined channel and interactions between binary solids. The experimental system was designed to be consistent with the idealised theoretical arrangements to maximise the measurement accuracy. The experimental particles were mixtures of silica sand particles of sizes 425-710 i^m and 710-880/~m, respectively. Specifically, we investigated the flow hydrodynamics of the binary suspension in terms of the settling length of both par- ticle species and the bed expansion behaviour. We also analysed the utilisation factor and the separation efficiency of the mFB. The results showed that the average solid volume fraction in the inclined channel fluctuated slightly for a given total solid inventory. The utilisation factor and separation efficiency of the system decreased when increasing either the fluidisation velocity or the solid inventory. The prediction results were in good agreement with the experimental data with an absolute deviation of less than 15%.

Exposed facet-controlled N2 electroreduction on distinct Pt3Fe nanostructures of nanocubes, nanorods and nanowires

Understanding the correlation between exposed surfaces and performances of controlled nanocatalysts can aid effective strategies to enhance electrocatalysis, but this is as yet unexplored for the nitrogen reduction reaction （NRR）. Here, we first report controlled synthesis of well-defined Pt3Fe nanocrystals with tunable morphologies （nanocube, nanorod and nanowire） as ideal model electrocatalysts for investigating the NRR on different exposed facets. The detailed electrocatalytic studies reveal that the Pt3Fe nanocrystals exhibit shape-dependent NRR electrocatalysis. The optimized Pt3Fe nanowires bounded with high-index facets exhibit excellent selectivity （no N2H4 is detected）, high activity with NH3yield of 18.3μg h-1mg-1cat(0.52μg h-1cm-2ECSA; ECSA:electrochemical active surface area) and Faraday efficiency of 7.3%at-0.05 V versus reversible hydrogen electrode, outperforming the{200}facet-enclosed Pt3Fe nanocubes and{111}facet-enclosed Pt3Fe nanorods. They also show good stability with negligible activity change after five cycles. Density functional theory calculations reveal that, with high-indexed facet engineering, the Fe-3d band is an efficient d-d coupling correlation center for boosting the Pt 5d-electronic exchange and transfer activities towards the NRR.

Surface engineering at the interface of core/shell nanoparticles promotes hydrogen peroxide generation

Hydrogen peroxide(HO), an environmentally friendly oxidant, has already been widely used in many chemical synthesis and industrials as an alternative to replace traditional oxidants including chlorinated oxidizers and strong acids. However, the conventional synthesis method confronts intense energy cost,tedious separation procedures and high cost, which is not competitive with traditional oxidants. Although direct HOsynthesis from Hand Ois a green and atomically economic reaction, satisfactory activity and desirable selectivity still remain formidable challenges. Herein, for the first time, a class of Pd@NiO-x nanoparticles(NPs)(x = 1, 2, 3 and 4) with a unique core@shell interface structure has been created to achieve high activity, selectivity and stability for the direct HOsynthesis. A precise thermal annealing on Pd@Ni-x NPs revealed that the resulting Pd@Ni O-x NPs exhibited the volcano-like activity toward direct HOsynthesis as a function of annealing temperature and time. By tuning the composition of Pd@Ni O-x NPs and the reaction condition, the efficiency of HO-2 synthesis could be well optimized with 5 wt%Pd@NiO-3/TiOexhibiting the highest productivity(89 mol/(kgh)) and selectivity(91%) to HOas well as excellent stability, making it one of the best catalysts for direct HOsynthesis reported to date.

Partially hydroxylated ultrathin iridium nanosheets as efficient electrocatalysts for water splitting

Ultrathin two-dimensional（2D） materials have attracted considerable attention for their unique physicochemical properties and promising applications; however, preparation of freestanding ultrathin 2D noble metal remains a significant challenge. Here, for the first time, we report use of a wet-chemical method to synthesize partially hydroxylated ultrathin Ir nanosheets（Ir-NSs） of only five to six atomic layers’ thickness. Detailed analysis indicates that the growth confinement effect of carbon monoxide and the partially hydroxylated surface play a critical role in formation of the ultrathin structure. The ultrathin Ir-NSs exhibit excellent performance for both the hydrogen evolution reaction and oxygen evolution reaction in a wide p H range, outperforming the state-of-the-art Pt/C and IrO;, respectively. Density-functional theory calculations reveal that the partial hydroxylation not only enhances the surface electron transfer between Ir-sites and intermediate O-species, but also guarantees efficient initial activation of bond cleavage of H-O-H for first-step H;O splitting. This, ultimately, breaks through barriers to full water splitting, with efficient electron transfer essentially maintained.
