Mastitis is a common disease that affects women during lactation， posing a threat to the health of both mothers and infants. Recent studies have shown that insufficient nutrient intake increase the risk of mastitis. Phlorizin (PHZ) is one of the nutrients present in apples. This study uses Lipopolysaccharide (LPS)-induced mastitis mice and LPS+ATP-stimulated mouse mammary epithelial cells (mMECs) as research objects to explore the effect and mechanism of PHZ on mastitis. Different to in vitro beliefs， our findings demonstrated that PHZ significantly reduced inflammation and protected the blood–milk barrier (BMB) in vivo. Additionally， we observed that oral administration of PHZ regulated the intestinal flora and exhibited prebiotic functions. However， the anti-inflammatory effect of PHZ was not solely dependent on the intestinal flora， as antibiotic disruption of the intestinal flora does not completely abolish the improvement of mastitis by PHZ. Further mechanistic research revealed that the anti-inflammatory properties of PHZ were attributed to its metabolism into phloretin (PHT). Moreover， our results demonstrated that PHT reduced inflammation and protected the BMB by promoting autophagy to prevent the pyroptosis of mMECs. This study provides a theoretical basis for reducing inflammation in lactating women by consuming fruits， such as apples， that contain PHZ.

Aniba rosaeodora essential oil(RO) has been traditionally used in natural medicine as a substitute for antibiotics due to its notable antidepressant and antibacterial properties. Salmonella, a prevalent pathogen in foodborne illnesses, presents a major challenge to current antibiotic treatments. However, the antibacterial efficacy and mechanisms of action of RO against Salmonella spp. remain underexplored. This study aims to elucidate the chemical composition of RO, evaluate its antibacterial activity and mechanisms against Salmonella in vitro, and further delineate its anti-inflammatory mechanisms in vivo during Salmonella infection. Gas chromatography-mass spectrometry(GC-MS) was utilized to characterize the chemical constituents of RO. The antibacterial activity of RO was assessed using minimal inhibitory concentration(MIC) and time-kill assays. Various biochemical assays were employed to uncover the potential bactericidal mechanisms. Additionally, mouse and chick models of Salmonella infection were established to investigate the prophylactic effects of RO treatment. RO exhibited significant antibacterial activity against both Gram-positive and Gram-negative bacteria, with an MIC of 4 mg·mL^(-1) for Salmonella spp. RO treatment resulted in bacterial damage through the disruption of lipid and purine metabolism. Moreover, RO reduced injury and microbial colonization in infected mice and chicks. RO treatment also modulated the host inflammatory response by inhibiting proinflammatory pathways. In conclusion, our findings demonstrate that RO is effective against Salmonella infection, highlighting its potential as an alternative to antibiotics for antibacterial therapy.

The coronavirus disease 2019(COVID-19) outbreak caused by the severe acute respiratory syndrome coronavirus-2(SARS-CoV-2) developed into a global health emergency. Systemic microthrombus caused by SARS-CoV-2 infection is a common complication in patients with COVID-19. Cardiac microthrombosis as a complication of SARS-CoV-2 infection is the primary cause of cardiac injury and death in patietns with severe COVID-19. In this study, we performed single-cell sequencing analysis of the right ventricular free wall tissue from healthy donors,patients who died during the hypercoagulable period of characteristic coagulation abnormality(CAC), and patients who died during the fibrinolytic period of CAC. We collected 61,187 cells enriched in 24 immune cell subsets and 13 cardiac-resident cell subsets. We found that in the course of SARS-CoV-2 infected heart microthrombus, MYO1EhighRASGEF1Bhighmonocyte-derived macrophages promoted hyperactivation of the immune system and initiated the extrinsic coagulation pathway by activating chemokines CCL3, CCL5. This series of events is the main cause of cardiac microthrombi following SARS-CoV-2 infection. In a SARS-CoV-2 infected heart microthrombus, excessive immune activation is accompanied by an increase in cellular iron content, which in turn promotes oxidative stress and intensifies intercellular competition. This induces cells to alter their metabolic environment, resulting in increased sugar uptake via the glycosaminoglycan synthesis pathway. In addition, high levels of reactive oxygen species generated by elevated iron levels promote increased endogenous malondialdehyde synthesis in a subpopulation of cardiac endothelial cells. This exacerbates endothelial cell dysfunction and exacerbates the coagulopathy process.

The Ebola virus(EBOV)is a member of the Orthoebolavirus genus,Filoviridae family,which causes severe hemorrhagic diseases in humans and non-human primates(NHPs),with a case fatality rate of up to 90%.The development of countermeasures against EBOV has been hindered by the lack of ideal animal models,as EBOV requires handling in biosafety level(BSL)-4 ***,accessible and convenient animal models are urgently needed to promote prophylactic and therapeutic approaches against *** this study,a recombinant vesicular stomatitis virus expressing Ebola virus glycoprotein(VSV-EBOV/GP)was constructed and applied as a surrogate virus,establishing a lethal infection in *** infection with VSV-EBOV/GP,3-week-old female Syrian hamsters exhibited disease signs such as weight loss,multi-organ failure,severe uveitis,high viral loads,and developed severe systemic diseases similar to those observed in human EBOV *** animals succumbed at 2–3 days post-infection(dpi).Histopathological changes indicated that VSV-EBOV/GP targeted liver cells,suggesting that the tissue tropism of VSV-EBOV/GP was comparable to wild-type EBOV(WT EBOV).Notably,the pathogenicity of the VSV-EBOV/GP was found to be species-specific,age-related,gender-associated,and challenge ***,equine anti-EBOV immunoglobulins and a subunit vaccine were validated using this ***,this surrogate model represents a safe,effective,and economical tool for rapid preclinical evaluation of medical countermeasures against EBOV under BSL-2 conditions,which would accelerate technological advances and breakthroughs in confronting Ebola virus disease.

Polycystic ovarian syndrome （PCOS） is the most principal reason for infertility in reproductive women， but no versatile treatment is feasible. Although trans-10-Hydroxy-2-decenoic acid （10H2DA） exhibits the anti-inflammatory， anti-cancerous and immunomodulatory properties， its role in improving PCOS remains elusive. This present study examined the effects of 10H2DA on the estrous cycle， ovarian morphology， reproductive hormone， glucose and insulin tolerances of PCOS mice， analyzed the binding between 10H2DA and free fatty acid receptor 4 （FFAR4）， and explored the regulation of 10H2DA on the c-Jun N-terminal kinase （JNK）/JUN （Jun proto-oncogene） signaling， cytosolic calcium ion (Ca2+)， endoplasmic reticulum （ER） Ca2+release， mitochondrial Ca2+， mitochondrial function， autophagy， lysosomal acidification， mitochondrial fission， secretion function and apoptosis of ovarian granulosa cells （GCs） using enzyme-linked immunosorbent assay， flow cytometry. western blotting， molecular docking， various fluorescent probes or plasmid， etc. The results found that 10H2DA alleviated the symptom of PCOS mice through improving the secretion dysfunction of GCs and protecting GCs against apoptosis. After binding to FFAR4 and inactivation of JNK/JUN signaling， 10H2DA restricted the release of ER Ca2+through targeting ryanodine receptors （RYRs） which had been identified as the downstream targets of JUN， diminished cytosolic Ca2+accumulation and prevented Ca2+influx into mitochondria via adjusting the mitochondrial calcium uptake 1 （MICU1） and mitochondrial calcium uniporter （MCU）. Concurrently， 10H2DA kept the integrity of mitochondrial function through maintaining mitochondrial Ca2+homeostasis and suppressed the discharge of mitochondrial reactive oxygen species （mtROS） into the cytosol through blocking the mitochondrial permeability transition pore opening to avoid the lipid peroxidation and ameliorate the secretory function and apoptosis of GCs. Coronaviruses are widely transmissible between humans and animals, causing diseases of varying severity. Porcine enteric alphacoronavirus(PEAV) is a newly-discovered pathogenic porcine enteric coronavirus in recent years, which causes watery diarrhea in newborn piglets. The host inflammatory responses to PEAV and its metabolic regulation mechanisms remain unclear, and no antiviral studies have been reported. Therefore, we investigated the pathogenic mechanism and antiviral drugs of PEAV. The transcriptomic analysis of PEAV-infected host cells revealed that PEAV could upregulate lipid metabolism pathways. In lipid metabolism, steady-state energy processes, which can be mediated by lipid droplets(LDs), are the main functions of organelles. LDs are also important in viral infection and inflammation. In infected cells, PEAV increased LD accumulation, upregulated NF-κB signaling, promoted the production of the inflammatory cytokines IL-1β and IL-8, and induced cell death. Inhibiting LD accumulation with a DGAT-1 inhibitor significantly inhibited PEAV replication, downregulated the NF-κB signaling pathway, reduced the production of IL-1β and IL-8, and inhibited cell death. The NF-κB signaling pathway inhibitor BAY11-7082 significantly inhibited LD accumulation and PEAV replication. Metformin hydrochloride also exerted anti-PEAV effects and significantly inhibited LD accumulation, downregulated the NF-κB signaling pathway, reduced the production of IL-1β and IL-8, and inhibited cell death. LD accumulation in the lipid metabolism pathway therefore plays an important role in the replication and pathogenesis of PEAV, and metformin hydrochloride inhibits LD accumulation and the inflammatory response to exert anti-PEAV activity and reducing pathological injury. These findings contribute new targets for developing treatments for PEAV infections.

The recent concurrent emergence of H5N1,H5N6,and H5N8 avian influenza viruses(AIVs)has led to significant avian mortality *** 2020,frequent human-animal interactions have been *** gain insight into the novel H5 subtype AIVs(i.e.,H5N1,H5N6 and H5N8),we collected 6102 samples from various regions of China between January 2021 and September 2022,and identified 41 H5Nx *** analyses on the evolution and biological properties of these isolates were *** analysis revealed that the 41 H5Nx strains belonged to clade 2.***.4.4b,with 13 related to H5N1,19 to H5N6,and 9 to *** based on global 2.***.4.4b viruses showed that all the viruses described in this study were likely originated from H5N8,exhibiting a heterogeneous evolutionary history between H5N1 and H5N6 during 2015–2022 worldwide.H5N1 showed a higher rate of evolution in 2021–2022 and more sites under positive selection pressure in 2015–*** antigenic profiles of the novel H5N1 and H5N6 exhibited notable *** hemagglutination inhibition assay suggested that some A(H5N1)viruses may be antigenically distinct from the circulating H5N6 and H5N8 *** challenge assays demonstrated that the H5N8 virus(21GD001_H5N8)displayed the highest pathogenicity in mice,followed by the H5N1 virus(B1557_H5N1)and then the H5N6 virus(220086_H5N6),suggesting a heterogeneous virulence profile of H5 AIVs in the mammalian *** on the above results,we speculate that A(H5N1)viruses have a higher risk of emergence in the ***,these findings unveil a new landscape of different evolutionary history and biological characteristics of novel H5 AIVs in clade 2.***.4.4b,contributing to a better understanding of designing more effective strategies for the prevention and control of novel H5 AIVs.

Surrogate modeling for unsaturated infiltration via the physics and equality-constrained artificial neural networks
