Witness for resolution: post-conflict quadratic affiliation in semi-free ranging pigs

In social mammals,post-conflict resolution can involve the reunion of former opponents(reconciliation),spontaneous/solicited post-conflict affliation of a third party with either opponent(triadic contacts),and affiliation between other individuals(hereafter bystanders;quadratic contacts).Quadratic contacts-possibly informing complex cognitive abilities-have been neglected in post-conflict studies.We investi-gated quadratic affliation in semi-free ranging pigs Sus scrofa,at the ethical farm Parva-Domus(Cavagnolo,Italy).Kinship was known.We collected behavioral data on adult pigs(n=104)via video recordings(43 h)followed by video analyses.Affiliative and anxiety behaviors between bystanders were collected under post-conflict(PC;following a conflict between non-bystanders)and matched-control(MC;no conflict)conditions.Quadratic affiliation was present in pigs,as bystanders affliated more in PC than MC,and such affiliation was followed by a decrease in the anxiety behaviors of both the interacting bystanders.Thus,quadratic contacts may be partly aimed at reducing one's own anxiety(intrinsic regulation).Quadratic affiliation was highest between closely related bystanders,which suggests that such affiliation may be most effective when close kin is involved.Quadratic affiliation was lowest after reconciliation and spontaneous triadic contacts.This suggests that direct peacemaking between opponents and spontaneous triadic contacts with close kin may most likely replace quadratic affiliation.Hence,pigs can be influenced by the negative events that affect other pigs-but not themselves-and their response may be modulated by social factors.Such non-random quadratic affiliation may point toward the presence of elements of social appraisal abilities inpigs.

Repercussion of salinity on hematological parameters and tissue morphology of gill and kidney at early life of tilapia

This study was attempted to reveal the changes in three hematological parameters such as blood hemoglobin,number of white blood cells(WBCs)and number of red blood cells(RBCs)and one biochemical(e.g.,glucose)parameter along with the changes in histomorphological structures of gills and kidney of Nile tilapia fry reared in saline water.Two experimental set up were used for two different age groups-5 days-after-hatch(5DAH)and 15 days-after-hatch(15DAH)-to test a novel hypothesis:if a little difference in the age during the first exposure to saline water could produce any variation in the results.They were placed into 6 plastic drums having six different salinity levels:0,5,10,15,20 and 25.After 60 days,at least 5 individuals were taken out from each drum to observe the above-hematological and histomorphological parameters.A two-way-ANOVA revealed that,except for hemoglobin,all the observed parameters have been affected the effect of age×salinity interaction(Ppresent as gill degenerations,hyperplasia was prevalent across almost all salinity levels.On the other hand,glomerular necrosis,pyknotic nucleus,hyaline droplets degeneration,shrinking of the glomeruli,etc.were observed in kidney structures.By assisting to understand the effects of salinity stress,all these data on tilapia fry will help to determine the prospects and scope of culturing the species in high-salinity water.

Low-density lipoprotein receptor-related protein 2(LRP2)is a multifunctional endocytic receptor expressed in epithelial cells.In mammals,it acts as an endocytic receptor that mediates the cellular uptake of cholesterol-containing apolipoproteins to maintain lipid homeostasis.However,little is known about the role of LRP2 in lipid homeostasis in insects.In the present study,we investigated the function of LRP2 in the migratory locust Locusta migratoria(LmLRP2).The mRNA of LmLRP2 is widely distributed in various tissues,including integument,wing pads,foregut,midgut,hindgut,Malpighian tubules and fat body,and the amounts of LmLRP2 transcripts decreased gradually in the early stages and then increased in the late stages before ecdysis during the nymphal developmental stage.Fluorescence immunohistochemistry revealed that the LmLRP2 protein is mainly located in cellular membranes of the midgut and hindgut.Using RNAi to silence LmLRP2 caused molting defects in nymphs(more than 60%),and the neutral lipid was found to accumulate in the midgut and surface of the integument,but not in the fat body,of dsLmLRP2-treated nymphs.The results of a lipidomics analysis showed that the main components of lipids(diglyceride and triglyceride)were significantly increased in the midgut,but decreased in the fat body and hemolymph.Furthermore,the content of total triglyceride was significantly increased in the midgut,but markedly decreased in the fat body and hemolymph in dsLmLRP2-injected nymphs.Our results indicate that LmLRP2 is located in the cellular membranes of midgut cells,and is required for lipid export from the midgut to the hemolymphand fat body in locusts.

Understanding the underlying mechanisms and links between genome evolution and adaptive innovations stands as a key goal in evolutionary studies.Poplars,among the world’s most widely distributed and cultivated trees,exhibit extensive phenotypic diversity and environmental adaptability.In this study,we present a genus-level super-pangenome comprising 19 Populus genomes,revealing the likely pivotal role of private genes in facilitating local environmental and climate adaptation.Through the integration of pangenomes with transcriptomes,methylomes,and chromatin accessibility mapping,we unveil that the evolutionary trajectories of pangenes and duplicated genes are closely linked to local genomic landscapes of regulatory and epigenetic architectures,notably CG methylation in gene-body regions.Further comparative genomic analyses have enabled the identification of 142202 structural variants across species that intersect with a significant number of genes and contribute substantially to both phenotypic and adaptive divergence.We have experimentally validated a∼180-bp presence/absence variant affecting the expression of the CUC2 gene,crucial for leaf serration formation.Finally,we developed a user-friendly web-based tool encompassing the multi-omics resources associated with the Populus super-pangenome(http://***).Together,the present pioneering super-pangenome resource in forest trees not only aids in the advancement of breeding efforts of this globally important tree genus but also offers valuable insights into potential avenues for comprehending tree biology.

A new diatom genus Spargeria gen.nov.(Bacillariophyceae)from Xizang,China,and the description of two new species

During our investigation of diatom biodiversity in Xizang,two species exhibited unique morphological features discriminative from all previously known genera.Herein we describe these two species and describe as new the genus,Spargeria gen.nov.The new genus features narrow to wide rectangular valves,narrow valve mantles,filiform raphe branches that occur on the valve face only,terminal raphe fissures straight or slightly deflected to same side,bow-tie shaped central areas,chambered striae present on the valve face only,being absent from the mantle,wider striae near the axial area and very narrow near the margin,multiseriate striae with small and round areolae that are occluded externally.Comparatively,Spargeria zhuii sp.***.has larger and robust valves,radiate striae,with one divergent stria near the apices,while Spargeria chenia sp.***.is smaller,with narrow valves,striae slightly radiate in the middle,becoming convergent or parallel near apices.This new genus belongs to the family Pinnulariaceae,and it was compared and contrasted with other genera of this family.Our work suggests the need for continued studies to document the biodiversity of diatoms in Xizang.

Nuclear phylogenomics of angiosperms and insights into their relationships and evolution

Angiosperms(flowering plants) are by far the most diverse land plant group with over 300,000 species. The sudden appearance of diverse angiosperms in the fossil record was referred to by Darwin as the “abominable mystery,”hence contributing to the heightened interest in angiosperm evolution. Angiosperms display wide ranges of morphological, physiological,and ecological characters, some of which have probably influenced their species richness. The evolutionary analyses of these characteristics help to address questions of angiosperm diversification and require well resolved phylogeny. Following the great successes of phylogenetic analyses using plastid sequences,dozens to thousands of nuclear genes from next-generation sequencing have been used in angiosperm phylogenomic analyses, providing well resolved phylogenies and new insights into the evolution of angiosperms. In this review we focus on recent nuclear phylogenomic analyses of large angiosperm clades, orders, families,and subdivisions of some families and provide a summarized Nuclear Phylogenetic Tree of Angiosperm Families. The newly established nuclear phylogenetic relationships are highlighted and compared with previous phylogenetic results. The sequenced genomes of Amborella,Nymphaea, Chloranthus, Ceratophyllum, and species of monocots, Magnoliids, and basal eudicots, have facilitated the phylogenomics of relationships among five major angiosperms clades. All but one of the 64 angiosperm orders were included in nuclear phylogenomics with well resolved relationships except the placements of several orders. Most families have been included with robust and highly supported placements, especially for relationships within several large and important orders and families.Additionally, we examine the divergence time estimation and biogeographic analyses of angiosperm on the basis of the nuclear phylogenomic frameworks and discuss the differences compared with previous analyses. Pyrroloquinoline quinone:a potential neuroprotective compound for neurodegenerative diseases targeting metabolism

Pyrroloquinoline quinone is a quinone described as a cofactor for many bacterial dehydrogenases and is reported to exert an effect on metabolism in mammalian cells/tissues.Pyrroloquinoline quinone is present in the diet being available in foodstuffs,conferring the potential of this compound to be supplemented by dietary administration.Pyrroloquinoline quinone’s nutritional role in mammalian health is supported by the extensive deficits in reproduction,growth,and immunity resulting from the dietary absence of pyrroloquinoline quinone,and as such,pyrroloquinoline quinone has been considered as a“new vitamin.”Although the classification of pyrroloquinoline quinone as a vitamin needs to be properly established,the wide range of benefits for health provided has been reported in many studies.In this respect,pyrroloquinoline quinone seems to be particularly involved in regulating cell signaling pathways that promote metabolic and mitochondrial processes in many experimental contexts,thus dictating the rationale to consider pyrroloquinoline quinone as a vital compound for mammalian life.Through the regulation of different metabolic mechanisms,pyrroloquinoline quinone may improve clinical deficits where dysfunctional metabolism and mitochondrial activity contribute to induce cell damage and death.Pyrroloquinoline quinone has been demonstrated to have neuroprotective properties in different experimental models of neurodegeneration,although the link between pyrroloquinoline quinone-promoted metabolism and improved neuronal viability in some of such contexts is still to be fully elucidated.Here,we review the general properties of pyrroloquinoline quinone and its capacity to modulate metabolic and mitochondrial mechanisms in physiological contexts.In addition,we analyze the neuroprotective properties of pyrroloquinoline quinone in different neurodegenerative conditions and consider future perspectives for pyrroloquinoline quinone’s potential in health and disease.

Stabilization and detection of labile reaction intermediates in supramolecular containers

Detection and observation of reactive intermediates is an essential step in investigation of reaction pathways.However,most reactive intermediates are unstable and present at low concentrations;their short lifetimes make them difficult to detect and characterize.Supramolecular containers offer opportunities for the stabilization and characterization of those labile species,through isolation from the media and protection inside the cavity of the host.In this review,we summarize the examples of labile reaction intermediates that are stabilized and characterized with the help of supramolecular containers.The container compounds include carcerands,deep cavitands and amide naphthotubes.We focus on unstable guest species-cyclobutadiene,benzocyclopropenone,o-benzyne,1,2,4,6-cycloheptatetraene,anti-Bredt's olefin,fluorophenoxycarbene,O-acylisoamide,and hemiaminalthat act as intermediates in certain organic reactions.

Metabolic reprogramming of the inflammatory response in the nervous system:the crossover between inflammation and metabolism

Metabolism is a fundamental process by which biochemicals are broken down to produce energy(catabolism) or used to build macromolecules(anabolism). Metabolism has received renewed attention as a mechanism that generates molecules that modulate multiple cellular responses. This was first identified in cancer cells as the Warburg effect, but it is also present in immunocompetent cells. Studies have revealed a bidirectional influence of cellular metabolism and immune cell function, highlighting the significance of metabolic reprogramming in immune cell activation and effector functions. Metabolic processes such as glycolysis, oxidative phosphorylation, and fatty acid oxidation have been shown to undergo dynamic changes during immune cell response, facilitating the energetic and biosynthetic demands. This review aims to provide a better understanding of the metabolic reprogramming that occurs in different immune cells upon activation, with a special focus on central nervous system disorders. Understanding the metabolic changes of the immune response not only provides insights into the fundamental mechanisms that regulate immune cell function but also opens new approaches for therapeutic strategies aimed at manipulating the immune system.

Ensemble Wind Power Prediction Interval with Optimal Reserve Requirement

Wind power prediction interval(WPPI)models in the literature have predominantly been developed for and tested on specific case studies.However,wind behavior and characteristics can vary significantly across regions.Thus,a prediction model that performs well in one case might underperform in another.To address this shortcoming,this paper proposes an ensemble WPPI framework that integrates multiple WPPI models with distinct characteristics to improve robustness.Another important and often overlooked factor is the role of probabilistic wind power prediction(WPP)in quantifying wind power uncertainty,which should be handled by operating reserve.Operating reserve in WPPI frameworks enhances the efficacy of WPP.In this regard,the proposed framework employs a novel bi-layer optimization approach that takes both WPPI quality and reserve requirements into account.Comprehensive analysis with different real-world datasets and various benchmark models validates the quality of the obtained WPPIs while resulting in more optimal reserve requirements.
