Improved Growth and Stress Tolerance in the Arabidopsis oxtl Mutant Triggered by Altered Adenine Metabolism

Plants perceive and respond to environmental stresses with complex mechanisms that are often associ- ated with the activation of antioxidant defenses. A genetic screen aimed at isolating oxidative stress-tolerant lines of Arabidopsis thaliana has identified oxtl, a line that exhibits improved tolerance to oxidative stress and elevated temperature but displays no apparent deleterious growth effects under non-stress conditions. Oxtl harbors a muta- tion that arises from the altered expression of a gene encoding adenine phosphoribosyltransferase （APT1）, an enzyme that converts adenine to adenosine monophosphate （AMP）, indicating a link between purine metabolism, whole-plant growth responses, and stress acclimation. The oxtl mutation results in decreased APT1 expression that leads to reduced enzymatic activity. Correspondingly, oxtl plants possess elevated levels of adenine. Decreased APT enzyme activity dir- ectly correlates with stress resistance in transgenic lines that ectopically express APT1. The metabolic alteration in oxtl plants also alters the expression of several antioxidant defense genes and the response of these genes to oxidative chal- lenge. Finally, it is shown that manipulation of adenine levels can induce stress tolerance to wild-type plants. Collectively, these results show that alterations in cellular adenine levels can trigger stress tolerance and improve growth, leading to increases in plant biomass. The results also suggest that adenine might play a part in the signals that modulate responses to abiotic stress and plant growth.

The origin of multicellularity is considered an evolutionary milestone,as cellular differentiation and cooperation have allowed the emergence of specialized and unique functions.Some crucial biological processes involve only a few cells,making it difficult to detect specific transcriptional signatures in classical bulk tissue studies.Important information often remains hidden in the sum of thousands of cells,and thus localized studies are essential.Although plant responses to microbes are well known to occur highly localized(Emonet et al.,2021),approaches to describe their impact at a cellular level have been established only very recently.Applying single-cell transcriptomics,Verbon et al.,2023 addressed this topic and disclosed heterogeneous responses of root cells when colonized by the beneficial bacterium Pseudomonas simiae.Focusing on Arabidopsis,we here compare this study with some groundbreaking methodological advances and provides perspectives on upcoming endeavors(Figure 1).

Few-shot learning has been proposed and rapidly emerging as a viable means for completing various tasks.Recently,few-shot models have been used for Named Entity Recognition(NER).Prototypical network shows high efficiency on few-shot NER.However,existing prototypical methods only consider the similarity of tokens in query sets and support sets and ignore the semantic similarity among the sentences which contain these entities.We present a novel model,Few-shot Named Entity Recognition with Joint Token and Sentence Awareness(JTSA),to address the issue.The sentence awareness is introduced to probe the semantic similarity among the sentences.The Token awareness is used to explore the similarity of the tokens.To further improve the robustness and results of the model,we adopt the joint learning scheme on the few-shot NER.Experimental results demonstrate that our model outperforms state-of-the-art models on two standard Fewshot NER datasets.

Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops,including watermelon,whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants,accurate identification of mb-mRNAs is challenging owing to an array of technical problems.Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions,respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline.We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock,subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance.This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress.

Porous biomaterials which provide a structural and biological support for cells have immense potential in tissue engineering and cell-based therapies for tissue repair.Collagen biomaterials that can host endothelial cells represent promising tools for the vascularization of engineered tissues.Three-dimensional collagen scaffolds possessing controlled architecture and mechanical stiffness are obtained through freeze–drying of collagen suspensions,followed by chemical cross-linking which maintains their stability.However,cross-linking scaffolds renders their biological activity suboptimal for many cell types,including human umbilical vein endothelial cells(HUVECs),by inhibiting cell–collagen interactions.Here,we have improved crucial HUVEC interactions with such cross-linked collagen biomaterials by covalently coupling combinations of triple-helical peptides(THPs).These are ligands for collagen-binding cell-surface receptors(integrins or discoidin domain receptors)or secreted proteins(SPARC and von Willebrand factor).THPs enhanced HUVEC adhesion,spreading and proliferation on 2D collagen films.THPs grafted to 3D-cross-linked collagen scaffolds promoted cell survival over seven days.This study demonstrates that THP-functionalized collagen scaffolds are promising candidates for hosting endothelial cells with potential for the production of vascularized engineered tissues in regenerative medicine applications.

Long non-coding RNAs(lncRNAs)are important regulators of diverse biological processes,especially skeletal muscle cell differentiation.Most of the lncRNAs identified to date are localized in the nucleus and play regulatory roles in gene expression.The cytoplasmic lncRNAs are less well understood.We previously identified a long intergenic non-coding RNA(linc-RNA)activator of myogenesis(Linc-RAM)that directly binds MyoD in the nucleus to enhance muscle cell differentiation.Here,we report that a substantial fraction of Linc-RAM is localized in the cytoplasm of muscle cells.To explore the molecular functions of cytoplasmic Linc-RAM,we sought to identify Linc-RAM-binding proteins.We report here that Linc-RAM physically interacts with glycogen phosphorylase(PYGM)in the cytoplasm.Knockdown of PYGM significantly attenuates the function of Linc-RAM in promoting muscle cell differentiation.Loss-of-function and gain-of function assays demonstrated that PYGM enhances muscle cell differentiation in an enzymatic activity-dependent manner.Finally,we show that the interaction between Linc-RAM and PYGM positively regulates the enzymatic activity of PYGM in muscle cells.Collectively,our findings unveil a molecular mechanism through which cytoplasmic Linc-RAM contributes to muscle cell differentiation by regulating PYGM activity.Our findings establish that there is crosstalk between lncRNAs and cellular metabolism during myogenic cell differentiation.

Hepatic mitochondrial dysfunction contributes to the progression of nonalcoholic fatty liver disease(NAFLD).However,the factors that maintain mitochondrial homeostasis,especially in hepatocytes,are largely unknown.Hepatocytes synthesize various high-level plasma proteins,among which albumin is most abundant.In this study,we found that pre-folding albumin in the cytoplasm is completely different from folded albumin in the serum.

Fruit set is inhibited by adverse temperatures,with consequences on yield.We isolated a tomato mutant producing fruits under non-permissive hot temperatures and identified the causal gene as SIHB15A,belonging to classⅢhomeodomain leucine-zipper transcription factors.SIHB15A,loss-of-function mu-tants display aberrant ovule development that mimics transcriptional changes occurring in fertilized ovules and leads to parthenocarpic fruit set under optimal and non-permissive temperatures,in field and greenhouse conditions.Under cold growing conditions,SIHB15A is subjected to conditional haploinsufficiency and recessive dosage sensitivity controlled by microRNA 166(miR166).Knockdown of SIHB15A alleles by miR166 leads to a continuum of aberrant ovules correlating with parthenocarpic fruit set.Consistent with this,plants harboring an Slhb15a-miRNA166-resisiant allele developed normal ovules and were unable to set parthenocarpic fruit under cold conditions.DNA affinity purification sequencing and RNA-sequencing analyses revealed that SIHB15A is a bifunctional transcription factor expressed in the ovule integument.SIHB15A binds to the promoters of auxin-related genes to repress auxin signaling and to the promoters of ethylene-related genes to activate their expression.A survey of tomato genetic biodiversity identified pat and pat-1,two historical parthenocarpic mutants,as alleles of SIHB15A.Taken together,our findings demonstrate the role of SIHB15A as a sentinel to prevent fruit set in the absence of fertilization and provide a mean to enhance fruiting under extreme temperatures.

Short-Time Preparation of NaOH-Activated Carbon from Sugar Cane Bagasse Using Microwave Plasma Heating

The microwave induced argon plasma was applied to the preparation of NaOH-activated carbon from sugar cane bagasse. The distinguished feature of the heating technique with this cold plasma is the short operation time. The carbonization and the activation process were finalized in one step within 3 min. The prepared activated carbon with NaOH ratio 3 to bagasse characterized using N2 adsorption of type IV (IUPAC classification) to give specific surface area of 1980 m2/g and mesopore volume of 0.73 ml/g. It also showed a higher specific capacitance of 201 F/g in 1 M H2SO4 solution (with standard three electrodes) than the corresponding one by the conventional heating, previously reported. The other features were the absence of oxygen groups and the presence of carbon centered stable radicals, detected by ESR spectra, on the surface.
