Dear Editor After 14-years of development, the first prophylactic vaccine against the Hepatitis E virus （HEY） has been marketed since 2012 （Wu et al., 2012）o However, the neutralizing epitopes of HEY are not completely defined. E2s, a protruding homodi- mer domain of HEV capsid protein, is responsible for inter- acting with host cells to initiate infection （Li et al., 2009; Li et al., 2005）. It was shown that two monoclonal antibodies （mAb）, 8Cll and 8H3, could neutralize the infectivity of HEV in rhesus, and the two mAbs bind to different neutralizing conformational epitopes on E2s （Zhang et al., 2005）.

We constructed a prokaryotic expression vector expressing the Mycobacterium tuberculosis protein TB16.3,as well as 3 other proteins,including TB15.3,CFP-10,and Rv2626C,which were purified and analyzed for their effectiveness as detection *** TB16.3genes of *** H37Rv genomic DNA were amplified by polymerase chain reaction,inserted into the expression vector p ET-30a,and expressed in Escherichia *** enzyme-linked immunosorbent assay was used to detect the 4 *** *** *** bacteria expressing TB16.3 and the 3 other proteins were constructed and found mainly to be *** recombinant TB16.3 proteins,serum samples of 118 tuberculosis(TB)patients and 96healthy controls were ***,specificity,and adjusted concordance rate for the TB16.3 antibody were 72.9,86.5,and 79.6%,*** positive rate of Rv2626C antibody in TB patients(44.1%)was significantly lower than that in normal controls(75.0%,χ2=20.8,P<0.01).TB15.3 and TB16.3 were used for simultaneous detection and showed sensitivity,specificity,and repeatability rates of 69.4,96.9,and 83.7%.The antibody positive rate and specificity for patients with lung disease was 9.6 and 90.4%,***15.3 and TB16.3 were mixed and detected *** with the results for TB15.3,the sensitivity,specificity,and concordance rates were 82.2,95.9,and 88.9%,*** concordance rate was the highest value *** genes were cloned into a host strain and expressed *** TB16.3 recombinant protein may be used as a new serological antigen for tuberculosis diagnosis.

We constructed a Mycobacterium tuberculosis vector expressing CFP-10 and Rv2626c to examine the expression of these proteins in Escherichia coli as well as their *** CFP-10 and Rv2626c genes were amplified from tuberculosis H37Rv genomic DNA using polymerase chain *** were ligated into the expression vector PET30a and expressed in *** tag nickel column chromatography was used to purify the recombinant *** enzyme-linked immunosorbent assay(ELISA)was used for *** our ***-engineered bacteria containing a CFP10 and Rv2626c plasmid,the target protein was found mainly to be in the soluble *** formed mixed antigens of the recombinant CFP10 and Rv2626c *** results showed that in 214 blood samples,the positive rate was 77.1%.The target gene was successfully expressed in the host *** antigens of the recombinant CFP-10 and Rv2626c proteins can be used as a combination antigen in the serological diagnosis of tuberculosis.

Background The clinical presentations of frontotemporal dementia(FTD)are diverse and overlap with other neurological *** are,as of today,no biomarkers in clinical practice for diagnosing the ***,we aimed to find protein markers in cerebrospinal fluid(CSF)from patients with FTD,presymptomatic mutation carriers and *** Antibody suspension bead arrays were used to analyse 328 proteins in CSF from patients with behavioural variant FTD(bvFTD,n=16)and progressive primary aphasia(PPA,n=13),as well as presymptomatic mutation carriers(PMC,n=16)and non-carriers(NC,n=8).A total of 492 antibodies were used to measure protein levels by direct labelling of the CSF *** findings were further examined in an independent cohort including 13 FTD patients,79 patients with Alzheimer’s disease and 18 healthy *** We found significantly altered protein levels in CSF from FTD patients compared to unaffected individuals(PMC and NC)for 26 *** analysis show patterns of separation between unaffected individuals and FTD patients,especially for those with a clinical diagnosis of *** most statistically significant differences in protein levels were found for VGF,TN-R,NPTXR,TMEM132D,PDYN and *** with FTD were found to have higher levels of TN-R and NF-M,and lower levels of VGF,NPTXR,TMEM132D and PDYN,compared to unaffected *** main findings were reproduced in the independent *** In this pilot study,we show a separation of FTD patients from unaffected individuals based on protein levels in *** investigation is required to explore the CSF profiles in larger cohorts,but the results presented here has the potential to enable future clinical utilization of these potential biomarkers within FTD.
